1. Isolation, Characterization And Pathogenesis Of Capripox Virus

The economic losses occur in terms of mortality, reduced productivity and lower quality of wool and leather. The clinical manifestations of the disease include high temperature, lesions skin in the form of macules, papules, vesicles, pustule and scabs on hairless areas of the body. The disease is highly contagious having high morbidity and mortality in the infected herds. The present study was conducted to document the prevalence of goat pox disease in the different regions of Punjab. The study was based on clinical manifestation of the disease in various collecting spots including slaughter houses, cattle and hide markets and tanneries. The prevalence of goat pox at slaughter houses in different regions was 9.93% in arid region followed by 8.69% and 7% in southern and northern irrigated regions respectively. The prevalence of pox disease in sheep was highest (8.54%) in the northern irrigated region, 7.69% and 6.62% in arid and southern irrigated regions respectively. The prevalence of pox recorded in the hide markets shows a trend of high presence 7.29% in arid region followed by 6.22% and 3.84% in southern and northern irrigated regions. Whereas in sheep the overall prevalence was 0.51 %, 4.44% and 1.66% in northern irrigated, arid and southern irrigated regions. In tanneries the pox lesions were identified on the basis of method as adopted in hide markets. The overall prevalence of pox in goat was 3.96%, 4.06% and 4.09% while in sheep 9.58%, 2.41 % and 10% in northern irrigated, arid and southern irrigated regions. The overall prevalence of pox disease in goat was 5%, 5.79% and 5.34% in Northern irrigated, arid and southern irrigated regions respectively. Where as in sheep, pox was 3.133%, 4.11 % and 2.67% in Northern irrigated, arid and southern irrigated regions respectively. The highest trend of incidence of disease was present in the arid regions followed by southern and northern regions. The slaughter houses shows high incidence of disease as compared to cattle and hide market and tanneries. The result was significant (P<0.05) among the regions and samples collecting spots. A total of 100 samples consisting of 55 scabs and 45 skin tissues were randomly selected from the different collecting spots of the three regions. The scabs and skin tissue samples were processed on dehydrated minimum essential media tor virus isolation. The virus was isolated on Vero cell line culture and its characteristics were observed on the basis of specific cytopathic effects. All 55 scab samples consisting 20 from cattle markets, 20 from slaughter house and 15 from hide market and tannery were tested through cell culture. The cell culture positive result for scabs was 60% cattle markets, 20% hide market and tannery and 40% slaughter house. All 45 skin tissue samples including 5 from cattle markets and tannery, 20 from hide market and 20 from slaughter house were subjected to virus isolation on Vero cell line. The cell culture positive result for skin tissue samples was 100% cattle markets, 30% hide market and tannery and 60% slaughter house. In this way the total cell culture result for scabs and skin tissue samples from all areas become 41.82% and 51.11 % respectively. The isolated virus was confirmed through peR. All the collected samples were also analyzed through peR in order to compare the two techniques for disease diagnosis. Out of 40 samples from slaughter houses 18 scabs and 15 tissues sample were positive through peR with 82.5%. Out of 25 samples collected from cattle markets consisting of 20 scabs and 5 skin tissues, 17 of scabs and 5 skin tissues were positive with 92%. Similarly a total of 35 samples out of which 15 were scabs and 20 were skin tissues collected from hide markets and tanneries. The peR of 7 scabs and 14 skin tissues was positive with 60%. In this way the total peR result for scabs and skin tissue from all areas was 42% and 34% respectively. In the 3rd study of the present project the isolated virus was inoculated in to experimental animal to study the detail pathogenesis. The disease followed the same pattern as in the natural outbreak. But however the routes of inoculation affect the severity of the disease. During the study the diseased animals were periodically slaughter at weekly interval after the appearance of 1 st clinical signs. The detailed lesions were observed in different visceral organs and the tissues were collected and preserved in 10% formalin. The tissues were processed for histopathology and immunohistochemical examination. The IHC was successfully optimized for the detection of viral antigen in the tissues of skin, lung and lymph nodes. Secondly to evaluate the efficacy of composted poultry litter/dead birds in broiler quail ration. The experiment was conducted at Poultry Research and Training Centre and Avian Research and Training Center, UVAS, Lahore in two different phases. The first phase was of 8 weeks duration in which composting of dead birds was doneusing advance windrow pile technique and proximate analysis of the composted material wascarried out. During the second phase, a quail ration was formulated according to dietary recommendations of NRC (1994) with inclusion of 0, 2, 4 and 6% compost and fed to quails,For this purpose, a total of 1200 day old Japanese broiler quails were randomly divided into 4 different experimental groups (A, B, C, and D). Group A was control and group B, C, and D contained 2, 4, and 6% composted ration respectively. The birds in each group were replicated six timescomprising 50 birds in each replicate. After 4 weeks of age three birds per replicate were slaughtered and their slaughtering parameters were recorded. The data thus obtained were analyzed through ANOVA in completely randomized design (Steelet al.1997) and means were compared by Duncan's Multiple Range (DMR) test (Duncan, 1955) using SAS (Statistical Analysis System) version 9.1. In production performance feed intake, body weight, body weight gain and FCR showed positive response when fed different levels of composted diet while mortality % remained unaffected throughout the experimental period. In slaughtering parameters live body weight (g), carcass weight %, dressing Weight %, Giblet weight %, Gizzard weight % and Heart weight % showed positively when fed different levels of composted diet while liver weight % remained unaffected throughout the experimental period. 3. Prevalence And Chemotherapy Of Various Endoparasites In Pheasants

A total of 200 faecal samples (100 from Lahore Zoo & 100 from Jallo Park Lahore) were collected from pheasants. Faeces were examined by using direct smear and centrifuge floatation technique. Forty six out of 200 Pheasants were found to be infected with nematodes. The overall prevalence of nematodes was 23%, while 20% & 26% prevalence was reported for Lahore Zoo & Jallo Park Lahore respectively. The overall species wise prevalence of nematodes reported as, Capillaria, Ascaridia, Coccidia, Trichostrongylus and Heterakis 23.9%, 30.4% 19.6%, 2.2% and 23.95 respectively. Then forty five birds were selected for chemotherapeutic trial by using fenbendazole and tetramisole and checked the efficacy of these drugs against the nematodes. The decrease in mean of EPG in faecal samples was calculated before and after treatment. All the Pheasants in group A were treated with tetramisole and mean EPG was 1000 at day 0 (before treatment). The reduction in mean EPG after treatment was observed 293, 220 and 367 on days 3, 7 and 18 respectively and tetramisole efficacy was 71%, 78% and 63%. The lowest (63%) tetramisole efficacy was calculated after 7 day, likewise highest (78%) on day 7 after treatment. The overall percentage efficacy of tetramisole was calculated 71% during this drug trial. On the other hand in group B the mean EPG was 961 before treatment (on day 0). The mean EPG was calculated 226, 193 and 327 on days 3, 7 and 21 respectively after fenbendazole treatment, indicating the egg load reduction 76%, 80% and 65% respectively. In group B, the highest reduction load (80%) of eggs was calculated on day 7 after medication, on contrary 65% on day 18 (post-treatment). The significant difference (p<0.05) was seen in EPG before and after treatment with anthelmintic in group A and B. The efficacy of fenbendazole was found highly significant (p<0.02) than tetramisole. Study was conducted on twelve healthy clogs with same age and body weight. The dogs were divided into three groups i.e A, B and C' comprising of four animals each. The dogs in group A and B were subjected to Y-U Antral advancement flap pyloroplasty and inverted pylorus duodenal plasty respectively whereas dogs in group C were kept as control. All the dogs were kept at surgery section department of C.M.S (Clinical Medicine and Surgery), UVAS Lahore throughout the experimental period for two months postoperatively. Prior to surgery, extreme cautions were observed to assure a successful attempt of this operation. The two techniques were evaluated on the basis of best effectiveness of the techniques as a relief to gastric outlet obstruction i.e. improvement in lurninal diameter post-operatively, development of adhesions, ease in surgery, healing at the operation site and least post operative complications. The evaluation of the procedures used were made on the basis of clinical observation, gastric emptying time, contrast radiography and post euthanasia assessment. The results of this study clearly indicated that the efficacy of Inverted pylorus cluodenal plasty was better than Y-U antral acivancenient flap pyloroplasty in tenns of convenience, short duration of operation, less post operative complications, increased the diameter of lumen and to provide better gastric drainage in short period of time. 6. Effects Of Infectious Bursal Desease Vaccine And Vaccination Schedule In Imunity Induceds By Newcastle Disease

Infectious bursal disease (IBD) is a viral disease producing suppression in humoral immune response causing degeneration of bursa of Fabricius. Different vaccines are available in the market for mass scale immunization of chickens. The study was carried out to compare the immunosuppressive effects of intermediate strain, hot strain and complex IBDV vaccines on immunity induced by NDV vaccine. Vaccine efficacy was studied by measuring the induced humoral antibody level using HI (Haemagglutination inhibition) test for NDV and indirect ELISA (enzyme-linked immunosorbent assay) (Kirkegaard' & Perry Laboratories - KPL) to detect antibodies against IBDV. The parameters used to evaluate the effects of IBDV vaccine on broiler chicks were immune response to NDV vaccination, weight of lymphoid organs such as bursa of Fabricius, thymus and spleen, post virulent NDV challenge and FCR. The results showed that IBDV vaccinated groups A, B, C, D and E had lower HI antibody profile, higher bursa, spleen and thymus body weight ratio, poor FCR and higher post challenge mortality than NDV vaccinated group F. The HI serum antibody profile revealed that the groups vaccinated with IBDV hot strain had significantly lower antibody titer as compared to the intermediate strain of IBDV vaccinated birds. In addition to that hot strain vaccine found to be more damaging to the bursa, spleen and thymus than the intermediate strain vaccine. The hot strain had adverse effects on the feed conversion ratio of birds as well. The challenge with virulent NDV revealed that IBDV vaccine treated groups were overall higher mortality than the only NDV vaccinated chickens. The study suggested the use of intermediate strain as vaccine since it had least interference with the antibody production against ND. However, hot strain used in this study had adversely affected the NDV HI. titer and caused more damage to the lymphoid organs and reduced feed conversion ratio (FCR). 7. Detection And Taxonomy Of Zoonotic Soil-Borne Helminths Of Cats And Dogs In Lahore

Material type: book Book; Format: print ; Literary form: not fiction Publisher: 2009Dissertation note: To find out the prevelance of parasitic eggs/larvae of zoonotic importance, 160 faecal samples, 80 soil samples and 80 herbage samples from each of the selected park during four months i.e. from 1St June to 30th September 2007 were collected and examined. The public parks included; 1) Nasir Park, 2) Iqbal Park. Thus in total 640 samples were examined for prevelance of parasitic eggs/larvae of zoonotic importance including faeces, soil and herbage. At the same time 160 faecal samples of pet dogs and cats were collected from the Pet Centre of University of Veterinary and Animal Sciences, Lahore and were examined for the presence of zoonotic parasites. These samples were processed and examined according to the techniques i.e. direct smear method, flotation and Baerman techniques. Five samples of each material were collected every week. The effect of meteorological data was also studied. Overall infection of the parks showed that 118 (37%) out of 320 faecal samples, 44 (27.5%) out of 160 soil samples and 81(51%) out of 160 herbage samples were found positive for parasitic eggs/larvae whereas 243 (38%) out of total 640 samples were found positive for parasitic contamination. One hundred and eighteen (37%) out of 320 faecal samples were found positive in which 72 (61%) were positive for A ncylostoma caninum, 23 (19%) for Toxocara canis, 15 (13%) for Diplidium caninum and 8 (7%) were found positive for others. Out of 160 soil samples 44 (27.5%) were found positive for parasitic infection. Amongst these 31 (70%) were positive for Ancylostoma caninum, 9 (20%) were positive for Toxocara canis and 4 (10%) were positive for other nematodes. Eighty one (51%) out of 160 samples was positive for parasitic infection. Amongst these 48 (59%) were positive for Ancylostoma caninum, 21 (26%) for Toxocara canis and 12 (15%) for others were found positive. The total 243 (38%) out of 640 were found positive for parasitic contamination. Amongst those 53 (22%) were positive for Toxocara canis, 151(62%) for Ancylostoma caninum, 15 (6%) for Diplidium caninum and 24 (10%) for others including Tinia saginata, Strongyloids etc. To find out the prevalence of zoonotic parasites in pet dogs and cats 160 faecal, 80 for dogs and cats each, samples were collected form the pet centre of University of Veterinary & Animal Sciences, Lahore. Among dogs 25 (31%) out of 80 faecal samples were positive for parasitic eggs/larvae and while in cats 23 (29) out of 80 samples were positive for parasitic eggs/larvae. Material type: book Book; Format: print Publisher: 2008Dissertation note: An epidemiological study of babesiosis in dogs was conducted at Pet center, University of Veterinary and Animal Sciences, Lahore, for one year and information on age, sex and breed was gathered. It was found that from a total number of 6204, dogs up to two years of age were more susceptible than other age groups (2-4, 4-6 and above 6 years).The data regarding genders revealed that males were more prone to the disease than female dogs. As far as the breeds were concerned, crossbred dogs were more susceptible followed by Pointers, Alsatians, German shepherds and Bull terriors.Hot and humid months (June to September) have greater impact on the occurrence of disease. The study regarding identification of ticks revealed that Rhiphicephalus sanguinus is the predominant vector of the disease in Pakistan. Molecular studies were conducted to characterize and identify the species responsible for canine babesiosis in Pakistan. In this regard, a nested polymerase chain reaction-Restriction fragment length polymorphism (PCR-RFLP) technique was employed on different specimens (Blood, Body tissues and Ticks). For this purpose blood samples were collected from twenty four chronically infected dogs and applied on the Flinders Technologies Associates (FTA) cards for transportation to Australia. Different body tissues (Liver, Spleen, Kidney, Intestine, Bone marrow and Pancreas) were procured after euthanizing the two dogs and DNA was extracted, for further studies. Similarly, the eighty eight ticks were also collected from the infested dogs in the 70% ethanol for transportation to Australia. A nested PCR-RFLP assay was used for the detection and differentiation of Piroplasm species on the basis of the 1 8S ribosomal RNA gene. The assay potentially amplified and identified Babesia gibsoni as the main canine piroplasm. Similar assays on the DNA extracted from body tissues and ticks revealed Babesia gibsoni as the main piroplasm. The PCR was found to have a high detection limit (equivalent to i0 dilution), when using the DNA extracted from blood applied to FTA cards, body tissues and ticks. A new technique was developed for extraction of DNA from FTA cards and tick, in this technique, instead of using the FTA specified punching machine, we used scalpel blades, and so the rest of the chemicals used are'generally and easily available. The same protocol was used for extraction of DNA from ticks, only chemicals used in different quantities with different spinning times. Both of which, resulted in cost reduction, less effort and speedy DNA extraction. 9. Prevalance And Anthelmentic Activity Of Indigenous Plants Against Trichostrongylus Of Sheep In District Zhob

A total 240 gastrointestinal tracts of sheep from district Zhob abattoir were collected. These samples were isolated and identified for trichostrongylid nematodes. The overall prevalence was 50%(120/240), of which 39.1% in male and 60.8% in female sheep were recorded at slaughter-house. Plants were collected from area of district Zhob. These plants were Identified and authenticated by botanist .The crude aqueous methanolic extract of the plants were used for in-vivo studies. Eighty sheep of either sex, aged between three to six months and naturally infested with Trichostrongylid nematodes (including trichostrongylius spp. I-Iaemonchus contortus, cooperia etc) were selected and managed separately for the experiment. These sheep were divided into 4 groups A ,B,C and D. Group A was contained 10 sheep and was untreated and considered as control. Thirty (30) were kept in Group B, were further sub divided into three equal groups i.e. BI, B2 and B3 and treated with different levels of Chenopodium album @ 1, 2 and 3 g/kg body weight respectively. Group C having thirty (30) was also sub divided into three equal groups i.e. Cl, C2 and C3 and treated with different levels of Artemisia brevifolia @ 1, 2 and 3g/kg body weight respectively. Group D was treated with Levamisole @ 7.5 mg/kg body weight. Faecal egg count reduction was criterion for evaluation. Faecal samples were collected before treatment on day 0 and on day 3, 5, 7, 10 and 14 post treatments for EPG count. The results of EPG for animals in group B1 at day 0 was 1325. This rate was reduced to 1250, 1125, 995 and 702 at day 3, 7, 10 and 14, respectively. Similarly EPG of sheep in B2 were 1280 at day 0 and reduced to 1205, 1202, 1001 and finally to 690 at day 3, 7, 10 and 14 respectively. Best results in B group against the nematodes were in B3 in which Chenopodium album was used 3g/kg bw. The results were 1250, 1231, 1145, 590 EPG at day 0, 3, 7, 10 and 14. The sheep in group Cl showed 1203 EPG at day 0, when treated with Ig/kg bwArtemisia brevifolia, the EPG was reduced to 1173, 1115, 700 and 528 on day 3, 7, 10 and 14 respectively. Second level of treatment C2 of Artemisia brevifolia which was given @ 2g/kg bw initially contained reduced to 1202 EPG, on day 3, 7, 10 and 14 the EPG counts were 1020, 631, 546 and 459, respectively. Highest dose of Artemisia brevfolia was 3g/kg body weight to sheep in group C3. On day zero the EPG count was 1196. On day 3, 7, 10 and 14th day the EPG count decreased to 1079, 905, 528 and 396 respectively. The sheep in group D, treated with recommended dose of Levamisole showed 1138 EPG prior to medication, which reduced remarkably to 681, 536, 357 and 147 on day 3, 7, 10 and 14 respectively. Although no untoward effects of plants were observed but best EPG reduction results (87.08 %) were observed in Levamisole as compared to treatment of 3gm/kg b.w Chenopodium album (51 .03 %) and Artemisia hrevfolia 3g/kgbw (66.88 %). CONCLUSIONS A wide variety of plants are naturally available in the Indo-Pakistan subcontinent which possess narrow or broad spectrum anthelmintic activities. No doubt this is true in other regions of the world as well where gastrointestinal parasitism is an important problem in livestock keeping, and the availability of commercial drugs may be limited. Conventionally, trichostrongylids has been tackled with use of synthetic anthelmintic but owning to development of anthelmintic resistance against major groups of anthelmintics viz., benzimidazole, Levamisole and avermectins, people are looking for alternatives to synthetic chemicals. The phytochemical analysis of these plants and controlled anthelmintic trials along with contemporary knowledge of parasite control strategies may offer new opportunities for effective and economical control of parasitic diseases. So these plants can be better alternative for synthetic chemicals. Quality control extracts of Artemisia brevifolia and Chenopodium album or possibly isolated bioactive compounds could be a promising alternative to conventional anthelmintics fbr the treatment of gastrointestinal trichostrongylids of small ruminants in the future. Such a treatment could be used in control strategies against gastro intestinal nernatodes in organic and conventional production systems. Further research is needed for studies on the bio active constituents, as well as on the reproducibility, dosage, application regime. toxicity and effectiveness of Artemisia brevifolia and C'henopodium a/bums in other host species and against other economically important gastro intestinal nematodes species. RECOMMENDATIONS It is recommended that further research could be carried out on large sample size in different seasons of the year and large number of plants, identification of active principles of plants with proven anthelmintic activity, standardization of dose and toxicity studies for drug development. In addition to this, large number of samples of the same plant from different geographic areas should be subjected to experimentation keeping in view the possibility of differences in chemical composition of soils. 10. Inter Population Variation In The Indian Flying (Pteropus Giganteus)Of Nwfp And Punjab-Pakistan

Material type: book Book; Format: print Publisher: 2010Dissertation note: The present study was designed to investigate the morphometric variations in the Indian flying fox populations inhabiting NWFP and Punjab. Intensive field efforts resulted in finding out three roosts of Indian flying fox (Pteropus giganteus) at NWFP (one each in Peshawar, Charsadda and Mardan districts and two in Punjab (one each in Gujranwala and Kasur districts). Four specimens each, from the three bat roosts of NWFP and, three specimens each from two bat roosts of Punjab were captured and euthanized to study the inter-population variations in the Indian flying fox (Pteropus giganteus). The results of the inter-population variations revealed that bat samples collected from Charsadda district were not significantly different from those collected from Peshawar district. They were also not different significantly from those collected from Kasur district. The Peshawar bats also showed no significant variation with respect to their cranial depth, breadth of upper incisors, maximum dentary length and mandibular tooth row with those collected from Gujranwala as well as other districts of Charsadda, Mardan and Ksur too. The bat sample collected from Charsadda district were also not significantly different from those collected from Mardan district with respect to their body weight, length of 3 metacarpal, length of l phalanyx on third metacarpal, length of 2' phalanyx on metacarpal, length 41 metacarpal, wingspan and length of femur, respectively. Whereas these specimens were not different from Gujranwala specimens in all the norphometric parameters measured. Mardan bat specimens were also not difibrent from Gujranwala bat specimens with respect to their mastoid breadth, length of the incisor of the upper jaw and mandibular tooth row, respectively. The specimens collected from Kasur district were not significantly different from Peshawar, Mardan, and Gujranwala with respect to the length of the desired parameters taken. Inter- population variations in all other comparisons were statistically similar. Combined inter-populations variations in the body weight, external body and cranial measurements of the Indian flying fox (Pteropus giganteus) captured from Punjab and NWFP. Of the 22 parameters studied, both populations were not significantly different ft)r any parameter. There were also no significant variations in the length of the Claw (C), Length oF the 5th metacarpal (5tI1 met) and the Condylobasal length (CBL). While the cranial depth (CR1)). Breadth of the incisors of upper jaw (J2J2), maximum dentary length (MDL) and mandibular tooth row length (MBTR) were also not different significantly in these specimens captired from two populations of Punjab and NWFP. A comparison of the two populations revealed that NWFP population had no significantly broader ear, longer femur, and deeper cranium, broader incisors of the upper jaw, longer dentary bone and longer mandibular tooth row with respect to Punjab. The body weight, external body and cranial measurements of the male and female Indian flying fox (Pleropus giganteus) populations captured from Punjab and NWFP were also had no 1significant variations in all the 22 parameters measured. Nonspecific, self-resolving acute gastritis or gastroenteritis is the most common GI disease seen in small animals. Congenital pyloric stenosis caused by congenital muscular hypertrophy also leads to pyloric outflow Obstruction. Acquired pyloric stenosis may be the result of external compression of the pylorus, caused, for example, by a pancreatic or hepatic tumor. Pyloric obstruction is common in humans and dogs. The only conclusive diagnostic test for pyloric stenosis is a satisfactory response to surgery, Ramstedt's pyloromyotomy being the simple and safest procedure. Various surgical procedure have been described for the relief of pyloric stenosis. Pyloroplasty (Heinekemukulicz procedure) , Y-U pyloroplasty, gasttroduodenostomy, gastrojejunostomy , and the procedures including the various bypass operations. In the view of above the present study was designed to evaluate the efficacy of Biliroth I, Biliroth II, and Roux-en-y resection procedure as a treatment of pyloric stenosis in dogs with special emphasis on the effectiveness of procedures used in the terms of physical evaluation, healing of wound, radiography, and postmortem studies. Twenty four mongrel dogs were selected and divided into four groups comprising six animals each. In group A, group B, and group C, Biliroth I, biliroth II, and roux-en-y resection procedures were used respectively. Group D was kept as a control group. The operations were conducted in aseptic condition. The stomach was exposed through the celiotomy incision to proceed the three surgical procedures in dogs of group A, group B, and group C. The incision was closed in routine manner. Clinical evaluation was performed to evaluate the efficacy of the techniques and postmortem was performed to see the gross changes in the stomach and abdomen cavity of dog. The results of this study suggested that all the three procedures are effective roux-en-Y resection procedure was more effective and useful. Although Roux-en-Y resection procedure is more difficult procedure to perform but it is more effective than Biliroth - I and Billroth - II procedures regarding clinical evaluation (vomiting, diarrhea, healing of wound) delayed gastric emptying time and least postoperative complications and postmortem studies. Eimeria tenella is the most common cause of caecal coccidiosis but E. necatrix is also isolated rarely. In the present study, 400 caecal samples suspected for coccidiosis were collected from two districts (Lahore and Gujranwala). Ten samples from twenty farms of each district were collected and examined for coccidiosis and species identication. Prevalence of coccidiosis was found 68%. It was more prevalent in district Gujranwala (71.5%) as compared to Lahore (64.5%). Eimeria species were identified by conventional (Direct Microscopy, Sedimentation technique, Floatation Technique and Sporulation) and molecular technique (PCR). Polymerase chain reaction was found the most sensitive ands accurate technique for species identification as compared to all conventional techniques. The prevalence percentage of E.tenella and E,necatrix was found 68% and 12.5% respectively. E.tenella was more prevalent in district Gujranwala (71.5%) as compared to district Lahore (64.5%). E.necatrix was also more prevalent in district Gujranwala (13.5%) than district Lahore (11.5%). The difference in percent prevalence of coccidiosis and species of Eimeria may be due to difference in farm management, farming type, biosecurity measures. The infection is often asymptomatic, but chronic infection can lead to scarring of the liver and ultimately to cirrhosis, which is generally apparent after many years. In some cases, those with cirrhosis will go on to develop liver failure, liver cancer or life-threatening esophageal and gastric varices. The present project was carried out to study the prevalence of laboratory based confirmed patients of Hepatitis-C in various public, private hospitals and in high risk groups among the population of Lahore metropolitan and its distribution and pattern with respect to person, time and place. Second part of the project was designed to study the risk factors of Hepatitis-C patients from out patient departments of various public and private hospitals of Lahore. Individuals at high risk from different organizations and occupations across the city population of Lahore metropolitan were also included in the study. The third part of the project was designed to investigate the distribution of genotypes of Hepatitis-C virus among patients through RT-PCR and theireffect on viral load, various haematological and biochemical parameters. Project-I Study-1: To estimate the prevalence of hepatitis C in various public and private hospitals of Lahore Metropolitan among different groups a total of 1399 individuals were tested to estimate the hospital based prevalence of HCV. Out of these 233 individuals produced positive result for Hepatitis-C virus infection. The overall hospital based prevalence was estimated to be 16.66% during the year 2009. The current study revealed that the highest prevalence was estimated in Dialysis patients and Organ recipients (41.17%) followed by General Patients of age > 12 years (14.60%) and pregnant women (10.84%). It was further observed that the least affected group was the Children of age ? 12 years (3.85%). Study-2: The results of estimated prevalence of Hepatitis C virus infection in high risk groups from the population in and around Lahore revealed that the highest prevalence was estimated in patients with HIV/AIDS (36.36%) followed by injecting drug users (36.09%), blood donors (17.78%), long rout truck drivers (14.70%), house hold and direct contact personal (14.6%) and prisoners (14.28%). It was also find out that the less affected groups were police department (10.66%), staff nurses and other health care workers (9.87%) and barbers and beauticians (6.97%) while doctors and dental surgeons were least affected (1.32%) among the high risk groups. Study-3: To find out the pattern and distribution of HCV patients with respect to person place and time a total of 924 patients were selected from the registry of Provincial Hepatitis Control Cell Lahore through systematic random sampling. Out of these, 154 fulfilled the inclusion criteria. Among these, 90 were male and 64 were females. Average age of male and female patients was 35.88±10.49 and 37.78±9.12 years, respectively. The age difference between male and female patients was statistically non-significant (P-value>0.05). It was further observed that 147 patients were Punjabi and 7 were from other provinces. Moreover, It was found that the highest number of patients was observed during the month of December (n=18) followed by November, 2008 (n=15), March (n=15) and July, 2009 (n=14) while the least number of patients were observed during the months of September, 2008 and May, 2009 (n=10). Project-II To study the risk factors associated with HCV infection an analytical cross sectional study was conducted. Study-1: Lower socio economic class, place of birth (hospital), delivery assisted by whom and breast feeding were significantly associated with HCV infection in children of age ? 12 years. The mean age of reactive and non-reactive general patients was significantly associated (P=0.012) with anti-HCV status. Marital status (OR=2.042), socioeconomic status, blood donation (OR=2.15), prescription by doctor or non-doctor (OR=2.664), route of drug administration, relative having hepatitis and towel sharing (OR=1.987) were also significantly associated (P<0.05) risk factors for HCV infection. The mean age of reactive and non-reactive pregnant women was 27.55±3.43 and 25.37±4.24 years, respectively. Educational level (OR=3.093) and occupational status (OR=2.228) were the important risk factors associated with HCV infection. Tattoo on the body (OR=11.833), comb sharing (OR=20.86) and razor sharing (OR=4.786) were significantly associated (P<0.05) with HCV infection. Pregnant women who gave the history of dental procedures and tooth brush sharing were 3.15 and 4.12 times more prone to get HCV infection, respectively. In 205 patients having dialysis and organ recipients 41.17% patients were reactive for Anti-HCV. Blood transfusion, glass sharing and qualification of the patients were significant factors in this group. Study-2: In case of doctors/dental surgeons a significant association was observed with history of blood transfusion and duties in medical and surgical wards. The nurses who worked in surgical wards, visited beauty salons were significantly associated (P<0.05) with HCV infection. Among health care workers age, gender and other factors did not have any significant influence on the reaction of HCV. Among blood donors female to male ratio was 1:16.5. It was found that the occupational status (p=0.002), place of surgical treatment (p=0.035), history of blood transfusion (p=0.000), ever pricked by sharps (p=0.045), habit of injecting drugs (p=0.04) and glass sharing (p=0.017) were significantly associated with occurrence of hepatitis C in blood donors. In long route truck drivers geographical status, surgical procedure, dental treatment and family history were significantly associated (P<0.05). Among the injecting drug users, demographic factors like marital (P=0.007) and educational status (P=0.000) were found to be significantly associated with HCV infection. Furthermore, the behavioral factors; use of injectable drugs with reused syringes (P=0.003), sharing of syringes in groups (P=0.004), place of shaving (P=0.000), use of disinfected ustra (razor) (P=0.003) and razor sharing (P=0.000) were significantly associated with anti-HCV status for IDUs. Among HIV/Aids patients a statistically significant (P<0.05) difference was present among the ages of reactive and non reactive patients. Comb sharing has also a positive effect of HCV but all other factors were not contributing in this group. In Police personals odds ratio for married persons was higher (9.57) but statistically insignificant. The mean age for reactive persons was 39.75±8.24 years. A non-sexual contact with HCV patient and spoon sharing were significantly associated. In prison inmates skin infection and sexual involvement were significantly associated (P<0.000) with HCV infection. In the group of 43 barbers/beauticians age, working shift, tattoo on body (OR=19.5), injecting drugs (OR=19.5) and pre-testing for HCV (OR=19.5) were significantly associated with HCV infection. In house hold and direct contact group previous history of accidents and family history of HCV (OR=18.36) were significantly associated with HCV infection. Project-III A molecular epidemiological study was conducted in which the HCV reactive patients as tested by ELISA test were subjected to viral load and genotyping through RT-PCR. The positive cases of Project-I were included in this project. In the present study 558 patients were reactive for Anti-HCV. Out of these, 34 (6.09%) patients had Type-1 genotype, 67 (12%) patients were accounted for Type-2 and 410 (73.47%) patients were positive for Type-3. Multiple genotypes were seen in 19 (3.4%) patients, 9 (1.61%) patients had un-type able genotype whereas in 19 (3.4%) patients genotype could not be detected. According to the distribution of genotype-1, 1a was present in 30 (88.23%) while 1b was seen in 4 (11.76%) patients. In patients of Type-2 genotype, 2a and 2b were present in 54 (80.59%) and 13 (19.40%) patients, respectively. In patients having Type-3, 3a and 3b were identified in 353 (86.09%) and 57 (13.90%) patients, respectively. Furthermore, Bilirubin, ALT, AST, ALPT, viral load, Hb, TLC, DLC, Platelet and ESR were statistically same in all genotype. Therapeutic trials were conducting by using three herbal drugs Nigella sativa, Punica granatum, 77 Citrullus colocynthis and one conventional drug i.e. Mebendazole (Vermox). A total of 400 samples of cats were examined for intestinal nematodiosis of these 77.50 % were found positive for intestinal nematodiosis .The parasites found in order of frequencies were Toxocara cati (93.54%), Toxascaris leonine( 75.80%) and Ancylostoma tubaeforme (48.38 %). . Over all high prevalence was recorded in cats less than one year of age than above one year. There was no difference in prevalence of parasitism between males and females. Result of chemotherapeutic trials showed that among herbal drugs Nigella sativa was found to be very effictive 95.94%, followed by Citrullus colocynthis (93.63%) and then Punica granatum (85.90%). 15. Epidemological, Serological, Heamatological And Therapeutic Studies On Ovine Nematodiasis In Three Ecological Zones of Balochistan

Four main experiments were conducted to highlight the objectives of the present research study. First experiment was conducted to find out the prevalence of sheep major nematodes for one year (January-December 2011). For this purpose three sheep breeds i.e., Balochi, Babrik and Harnai (either sex and between 1-5 years age groups)were selected randomly from three sites i.e., Quetta, Ziarat and Loralai. Faecal analyses of these sheep showed overall higher (40.25%) nematodes prevalence at Loralai followed by Ziarat (29%) and Quetta (23.92%). Five nematodes infection were recorded at three experimental sites. Among these, H. contortus (5.58 to 10.42%)and was the higher prevalent followed by N. battus (6.92 to 9.33%), S. papillosus (4.42 to 9%), T. colubriformis (2.33 to 7.33%) and T. ovis (1.83 to 6.83%).The nematodes prevalence was higher in one and five years old sheep. The female-sheep were infected with higher nematode prevalence higher the than male once and sometimes non-significant difference. These five nematodes were prevalent almost throughout the year; however, a peak infection was recorded during August/September. The high temperature, rainfall and humidity during these months may be predisposing factor of higher prevalence. Second experiment was on diagnosis of sheep nematodiasis through Enzyme-linked immunosorbent assay (ELISA). For this purpose H. contortus and T. ovis positive samples (200) based on coprological examination were also indicated 100% positive sensitivity by the ELISA based on crude somatic antigen, while on excretory antigen based showed lower (92%) sensitivity. The sera (n=200) of non-infected sheep (based on coprological examination) showed marked difference results. Such as 168 (84%) and 166 (83%) samples were found positive with H. contortus and T. ovis, respectively. While, based on crude somatic antigen 158 (79%) and 144 (72%) samples were found positive with H. contortus and T. ovis, respectively. Third experiment was conducted to determine the hematological values and total serum protein indices in healthy and nematodes infected sheep. The statistically significant (P<0.05) difference in PCV, Hb, RBC, WBC, Eosinophil, ESR and Total serum protein values was observed among healthy and nematode infected sheep groups. While, there was no statistically significant (P<0.05) difference in TLC, Lymphocytes, Neutrophil, Monocytes and Basophils counts in healthy and nematodes infected sheep groups. Fourth experiment was conducted on assessing the comparative efficacy of synthetic (Oxfendazole and Ivermectin) and locally manufactured herbal medicine (Deedani, Kirmar and Atreefal Deedan) anthelmintics against sheep nematodes at AZRC/PARC Range-livestock Research Station Sanjavi district Ziarat. The present study results regarding the comparative efficacy showed that, Atreefal deedan among herbal products (Deedani and Kirmar) and Ivermectin than Oxfendazole was found effective against sheep nematodes. The sheep treated with Ivermectin showed highest (96%) FEC reduction, followed by Oxfendazole/Atreefal deedan (86%), Kirmar (60%) and Deedani (32%). It is caused by H. lineatum and H. bovis in cattle and P. silenus in goats and is commonly existed species in Pakistan which belong to order Diptera, family Oestridae. The larvae of these flies are obligatory parasites of their hosts and are economically very important causing severe damage to the skins/hides effecting leather industry. Goat warble fly infestation has severe economic impact on tanning industries and it also causes growth retardation, carcass depreciation and production losses of milk and meat. Keeping in view the importance of this infestation, studies were designed on serodiagnosis, epidemiology and economic importance of hypodermosis in goats in northern uplands of Balochistan, Pakistan, with the objectives to diagnose warble fly infestation at early stages through serology for better control and eradication strategies, to study different epidemiological factors (age, sex, breed, temperature, humidity and rain fall) contributing goat warble fly infestation in the study areas, to compare seroprevalance over the prevalence based on clinical examination and to study the economic importance of hypodermosis in Loralai, Ziarat and Pishin districts of Balochistan, Pakistan. The duration of the study was one year commenced from April 2011 to March 2012. The most prevalent breed of goat i.e. khurasani breed in uplands of Balochistan was involved in the study which was compared with the Non-descriptive breed of goats. It is expected that the result of this study will be helpful to plan better control and eradication measures of warble fly by providing the base line data for further improvement in livestock sector in Balochistan. Serological studies for an early diagnosis and seroprevalence of hypodermosis were carried out by commercial ELISA kit (IDEXX hypodermosis serum antibody test) method. A total of 1440 blood samples were collected during the study. Sera were separated and stored at - 20 °C until further use. The infestation was detected in May and June (summer) where as the infestation is usually detected clinically in the months of October until February (winter) when warbles appear on the back and flank regions of the animals. ELISA based seroprevalence showed the highest mean percentage (48.45 %) of goat warble fly infestation in all the three districts, while the mean percentage by clinical palpation method was 15.94 % in all the three districts. In the slaughter house and skin market studies the mean percentages of goat hypodermosis were 9.07 % and 7.16 %, respectively. In the study of epidemiology, a total of 8640 number (2880 number per district) of goats of different age, sex and breed were examined on monthly bases by hand palpation clinico-parasitological method for the presence and prevalence of hypoderma infestation in the field animals. The mean prevalence percentage was 15.94 % i.e. 10%, 21.25% and 16.59% in Loralai, Ziarat and Pishin districts, respectively. In the study conducted in slaughter houses of all the three districts, a total of 4320 number (1440 number per district, 120 animals per month per district) of goats of different age, sex and breed were examined and the prevalence was recorded. Three age groups were selected for both the studies i.e. < 1year, 1-2 year and > 2 year old. The larvae were collected from naturally infested animals for identification. The mean prevalence percentage of hypodermosis in the slaughtered animals was 9.07 % i.e. 4.86%, 12.56% and 9.79% in Loralai, Ziarat and Pishin districts, respectively. Analysis of data by Pearson Chi-square test (?2) revealed highly significant difference (P<0.05) in the overall prevalence of hypodermosis district-wise in the fields and slaughter houses. Results were processed using Microsoft Excel 2007. In comparison of the fields and slaughter houses, the prevalence was significantly higher in the field animals as compared with the slaughterhouse animals in all the districts. In the study based on examination of the infested skin in the skin markets, a total of 3600 numbers of skins of goats (1200 numbers per district, 100 skins per month regardless of age, sex and breed) were examined by making monthly visits to the skin markets of each district. The intensity of the infection was also recorded by counting the number of holes in each infected skin. The overall district-wise mean prevalence percentage was 7.16 % and the mean intensity was 16.19 holes per skin. Analysis of data of the skin market by Pearson Chi-square test, using SPSS version 16, revealed highly significant differences (P< 0.05) in the prevalence of hypodermosis district-wise. Month-wise prevalence in the field animals showed that the highest prevalence was during January and lowest during October in Loralai, Ziarat and Pishin districts. In the slaughter house study, highest prevalence was observed during October and the lowest during July in Loralai district, highest during January and the lowest during July in Ziarat district whereas the highest prevalence was during the month of November and the lowest during July in district Pishin. Month-wise analysis of data by Pearson Chi-square test, using SPSS version 16, revealed highly significant differences (P< 0.05) in the prevalence of hypodermosis both in the field as well as in the slaughter house studies. Age-wise prevalence in the field as well as in the slaughter house animals showed the highest prevalence in the age group of 1-2 years as compared to the age groups of < 1 year and > 2 years. Analysis of data by Pearson Chi-square test, using SPSS version 16, revealed highly significant differences (P< 0.05) in the prevalence of hypodermosis amongst age groups both in the field as well as in the slaughter house studies. No significant difference was observed sex-wise and breed-wise in both the studies of fields and slaughter houses of the study areas. The life cycle pattern in naturally infested goats with warble fly was studied in the fields and slaughter houses of the study areas from April 2011 to March 2012. All the observations regarding the life cycle stages were recorded. First instars larvae (L1) were observed in subcutaneous tissues from mid of May to mid of July in slaughter houses of the study areas. Second instars larvae (L2) were observed in the slaughter houses from mid of July to November. Third instars larvae (L3), as well developed warbles on the back and flank regions of the goats, were palpated in the field animals and observed in the slaughter houses from December until the end of February. The pupal period was observed in early spring in the months from March to mid of April. No larvae were observed in the slaughter houses and fields of the study areas during this period while the adult fly activity season (Oviposition period) was observed in early summer from mid of April to mid of May. The estimation of economic losses due to skin damages were determined by multiplying the estimated infested numbers of skins with the value of loss per skin. The overall losses due to warble fly infestation in the study areas were calculated as Pak. Rupees 7578625.49 (Pak. Rs 7.57 million = US$ 77530.69, US$ 1= Rs. 97.75) annually. Based on the early detection studies, it is concluded and recommended that the best time for the treatment of goat warble fly infestation in uplands of Balochistan is in the months of June and July when first larval instars are still in migratory stage and not yet have reached their final sites on the back and flank regions of goats to cause damage to the skin, where as the farmers usually treat their animals against warble fly in the winter season when they observe the warbles on the backs of the animals but at that stage the larvae have damaged the skins of the animals. Haemonchus contortus is the most significant parasite of small ruminants and cause heavy production losses by causing reduction in meat and wool production. The parasite directly affects the health of an animal and causes anemia, hemorrhages, anorexia, weight loss and death of affected animal. This study was designed to diagnose the Haemonchosis in sheep in and around Lahore. The accurate diagnoses of the parasite are important for its control and treatment. Direct smear, floatation technique and Polymerase Chain Reaction (PCR) were applied to check the sensitivity and specificity in diagnosing Haemonchosis in sheep. For this purpose 100 faecal samples were collected randomly from different areas of Lahore. Each faecal sample was examined by direct smear method and floatation technique. Out of 100 faecal samples 44 were microscopically positive. 30 by direct smear method and 44 by floatation technique. Specific primers were designed to diagnose Haemonchosis in sheep by using ITS-2 Region. 44 microscopically positive samples were confirmed by PCR. 29 (66%) samples were found to be PCR-positive and 15 (34%) were found to be PCR-negative. Several other species of parasites were also found during microscopic examination of faecal samples. Two samples wetre found positive for coccidial oocyst and seven samples were also positive for other nematodes along with Haemonchus spp. Flotation technique was found to be superior in diagnosis of Haemonchosis as compared to direct smear. Conclusion: From current study it is concluded that Haemonchosis is widely prevalent in ovines. Its accurate diagnosis is essential for the treatment of infectious diseases and control of this parasite. Molecular technique has the advantage over conventional diagnostic techniques because PCR is more specific than conventional methods of diagnosis. According to the present study by conventional method there is 34% error chances to diagnose other Trichstrongylid eggs as Haemochus spp. The main advantage of using PCR as diagnostic test, are an increased speed of diagnosting the disease and its capability to notice low worm burden in small volume of faeces from individual animals. Endo-parasites represent a major constraint to the production and growth of small ruminants (Babar et al. 2013). Pakistan has been reported to problem of helminths in sheep and goat (Raza et al. 2009) and cattle and buffalo (Athar et al. 2011). Mainly infection is generally controlled by allopathic drugs and vaccination (Behnke et al. 2008). Parasitic diseases are a major threat in efficiency, the most widespread helminth parasites being Haemonchus contortus, Trichostrongylus spp. and Oesophagostomum columbianum. Liver flukes and paramphistomes (Fasciola gigantica and Paramphistomum microbothrium) occasionally cause heavy mortality in animals grazing swampy areas (Akerejola et al. 1979). The adverse effects of nematode infections include: loss of weight, anorexia, anaemia, retarded growth, delayed sexual maturity, decrease in milk and meat production (Saddiqi et al. 2012). Indigenous knowledge of herbal medicine is a big source of the modern knowledge. Today, thousands of plants, traditionally used as medicines are being explored (Kakar, 2012). Chemical control of helminths coupled with improved management has been an important worm control approach throughout the World. However, increasing problems of development of resistance in helminths rise in price of drugs contributing factor for traditional plant used (Coles et al. 1997) against anthelmintics have led to the proposal of screening medicinal 1 Introduction plants for their anthelmintic activities. Reports of drug resistance have been made in every livestock host and to every anthelmintic class. In some regions of world, the extremely high prevalence of multi-drug resistance (MDR) in nematodes of sheep and goats threatens the viability of small-ruminant industries. Many parasitic nematodes of veterinary importance have genetic features that favor the development of anthelmintic resistance (Kaplan et al. 2004). The plants are known to provide a rich source of botanical anthelmintics (Lewis and Elvin Lewis 1977). There are many medicinal plants have been used to treat parasitic infections in man and animals (Iqbal et al. 2005). Various botanical plants have been possessed anthelmintic activity against helminth e.g Chenopodium album (Eguale & Giday, 2009). It has been expected that there are around 250,000 plant species present throughout the world. Plants, from ancient, have served human beings as sources of food, shelter, clothing and medicines. Before the advent of modern allopathic medicine and synthetic drugs, plants and to a certain extent, animals and minerals were used in various formulations for treatment of diseases by traditional medicinal practitioner (Rahmatullah et al. 2011). Plant medicine is very important from ancient to present daytime. The uses of biologically different plant assets for various ailments are the lifelong struggle of humankind (Hussain et al. 2008). In Indo-Pak subcontinent, Ayurvedic and Unani therapeutic systems are very popular and people have been using plants not only for the treatment of their own ailments but also for their domesticated animals. Chenopodium album (Chenopodiaceae) commonly known as “Bathu” is important medicinal plants in Pakistan and their different parts are utilized in the traditional system of medicine (Said et al. 1970). Chenopodium album usually recognized as “Bathua” is a familiar food as well as a medicinal plant. In traditional system of medicine, it is used as an anthelmintic, antimicrobial, antirheumatic, contraceptive, laxative, cardiotonic, antiscorbutic, 2 Introduction and blood purifier & also in management of hepatic disorder, spleen enlargement, intestinal ulcers, digestive, carminative, seminal weakness, pharyngopathy, splenopathy, hemorrhoids, cardiac disorder (Panigrahy et al. 2012). The seeds of the plant are known to possess anthelmintic activity against Haemonchus contortus. A compound C37- trihydroxy adjacent bistetrahydrofuran acetogenin, present in the seeds, is responsible for inhibition of the egg hatching of gastrointestinal nematodes (Saha et al. 2011). Chenopodium album (Chenopodiaceae) is a rapid rising fragile annual plant and it is found in Bangladesh. In English plant known as Lamb’s quarters and in Bengali as Buthiya shak. Both in vitro and in vivo activity of plant as anthelmintic has been reported (Jain and Singhai 2012). Chenopodium album was found to be one of the initial plant species colonizing a heavy metal-contaminated site, polluted by pyritic (sulphide-rich) waste from the Aznalcóllar Mine spill (South-western Spain). This shows its importance in the re-vegetation of this soil (Walker et al. 2004). In vitro experiments were arranged to conclude the possible anthelmintic efficacy of crude aqueous extracts and powder of the whole of Chenopodium album (Eguale et al. 2009). Commercial preparations of oil of chenopodium album and its active constituent, ascaridol, obtained by steam distillation, have been and continue to be, used with substantial success in mass treatment campaigns (Kliks et al. 1985). Various species of the genus Ascaris, Toxocara canis, Toxocara cati and Toxocara malaysiensis, are clinically important to cause diseases in mammals that are usually canids and felids (Fisher, 2003; Lee et al., 2010). Toxocara canis and T. cati are of zoonotic importance. (Fillaux et al., 2013). This parasite has a fecal-oral type of transmission. Dogs are its definitive host and humans are its accidental hosts. Its eggs are non-infective when eliminated with feces of dogs. Eggs then develop into infective embryonated stage. The duration of embryo development depends upon different environmental factors such as temperature, humidity and type of soil (Overgaauw et al., 2013). Soil of the parks, streets and other public places polluted with the feces of stray dogs and cats also play a great role in the transmission of toxocariosis to humans. Toxocara spp. have the ability to reproduce and survive for long time periods outside their host. In this way the lands of parks, streets and other public places remain infected for long times. This contaminated soil with Toxocara eggs poses a high risk of infection to humans. Specially children under 5 years of age are the key set affected because they play in playgrounds and by chance can swallow the eggs of these intestinal parasites from contaminated soil (Dado et al., 2012; Thomas et al., 2014). Infection commences both in man and dogs by ingestion of embryonated eggs, which are then hatched to liberate larvae. Humans get infected by the accidental ingestion of infective eggs with contaminated food or water. Eggs hatch in the stomach and larvae move to the small intestine. From small intestine these larvae migrate to visceral organs via blood causing a series of to retina via retinal artery and impair eye vision (Nicoletti, 2013). In dogs, these larvae mature within the small intestine. Then, mating takes place and new non-embryonated eggs are formed (Despommier, 2003). Humans can acquire infection by infective embryonated infections (Rubinsky-Elefant et al., 2010). Clinical signs of the infection varies from asymptomatic to a range of severe infections i-e. Visceral organ damage (Fillaux et al.) by the migration of larvae of T. canis and T. cati. These larvae cause Ocular larva migrans by the migration of larvae eggs from adulterate soil, dirty hands, un-cooked vegetables and by direct exposure to pets (Good et al., 2004; Wolfe et al., 2003). Nearly 100% of pups are infected in utero by reactivated somatic larvae from day 42 of the gestation period. This trans-placental path and intra uterine infection is the most important means of transmission in pups (Overgaauw et al., 2013). Zoonotic prevalence is higher in tropical region than in temperate regions. Rural populations consume more risks of infection as compared to urban population of same area (Rubinsky-Elefant et al., 2010). Worldwide surveys show that the prevalence of Toxocara infection ranges from 86% to 100% in pups and 3% to 81% in adult dogs. Infection rate is higher in pups between age of 1-6 months as compared to dogs more than 7 months old (Itoh et al., 2004). In Lahore, prevalence of Toxocara infection is 49% in stray dogs and 30% in pet dogs (Chattha et al., 2009). Various diagnostic methods are employed for diagnosis of Toxocara species. Coproscopical techniques are classical techniques which are extensively used to identify eggs and larvae in fecal samples based upon morphological characteristics, coproscopic techniques have been extensively used to diagnose larvae and eggs in fecal samples (Sweeny et al., 2011). But identification by these coproscopic techniques has limitation in the differentiation of different species of Toxocara (Li et al., 2007a). Immunodiagnostic approaches like ELISA have also been utilized by using excretory/secretory antigens of larvae (Fan et al., 2004). But it has drawback that there is chance of cross reaction between anti-toxocara serum and antigen from other Toxocara spp. (Lozano et al., 2004). Alternatively, the DNA based molecular approaches are being employed for the accurate diagnosis of toxocariosis at species level. This technique may overcome the drawbacks of traditional methodologies and help in molecular epidemiological screening because genetic variation can be studied. Molecular methods based on DNA are more sensitive and specific as compared to classical methods (Jacobs et al., 1997; Ndao, 2009). In Pakistan, DNA based diagnosis of Toxocara spp. has not been employed yet. The present study was designed to achieve the goal of accurate diagnosis of prevalent species of Toxocara in dogs by using effective DNA based diagnostic technique. Fleas are one of the most important ectoparasites with more than 2,000 species worldwide affecting mammals, birds, and reptiles (Hsu, 2003). In some locations, fleas represent over 50% of all the dermatological cases presented to small animal clinics. Most are limited to hosts with nests as this can provide conditions for the completion of their life cycle (Linardi & de Avelar, 2014). While fleas on pets are generally considered a nuisance that may cause some dermatologic problems, they are also responsible for the transmission of several important diseases in humans and animals (Dryden & Rust, 1994). They have been involved in transmission of cat scratch disease (Bartonella henselae) (Chomel et al., 2006; Comer et al., 2001), Rickettsia typhi (Murine thyphus), Rickettsia felis (Finkelstein et al., 2002; Rolain et al., 2005), and also serve as the intermediate host for the tapeworm Dipylidium caninum (Rust & Dryden, 1997) and several trypanosomatids (Coutinho & Linardi, 2007). The term ‘‘cat flea,’’ which is the approved common name for Ctenocephalides felis felis (C. f felis), can occasionally cause confusion. When it appears in print, it refers to the specific flea genus and species and not to fleas recovered from cats. There are four recognized subspecies of C. felis throughout the world: Ctenocephalides felis damarensis and C. felis strongylus occur primarily in East Africa, C felis orientis occurs in India and Australia, and the widespread C. f felis occurs in all continents except Antarctica and is the only subspecies that occurs in North America (Rust & Dryden, 1997). The cat flea, C. felis, is a clinically important parasite of domestic pets, being responsible for the production of allergic dermatitis, serving as the vector of Introduction 2 various bacterial pathogens, and being the intermediate host for filarid and cestode parasites. Flea allergy dermatitis is the most common dermatologic disease of dogs and a major cause of feline miliary dermatitis (Dryden & Rust, 1994; Rust & Dryden, 1997). Clinical features vary from asymptomatic to severe hypersensitivity reactions with restlessness, alopecia from scratching and biting resulting in a pruritic papular dermatitis. Vacuuming of carpets, furniture cushions, rugs, or other substrata, with a vacuum machine containing a ‘‘beater bar,’’ will remove many of the flea eggs and larvae. In addition, cocooned pupae at the upper levels of the carpet can also be affected. The vibration also stimulates adult fleas to emerge from their cocoons so that they can be collected in the vacuum machine. Therefore frequent vacuuming, during a flea infestation, can reduce the overall flea burden in the home. It should be ensured that vacuum bags are disposed of properly, to prevent recolonization of the home with flea stages previously removed by vacuuming. Because outdoor development of immature flea life stages is limited to shaded areas, altering outdoor environments to eliminate such habitats can effectively reduce flea populations. Because urban wildlife, such as opossums, raccoons, and foxes, are good hosts for cat fleas, pet owners should avoid encouraging visitations by wildlife, which will affect flea and tick control (see later discussion). Treatment of indoor and outdoor environments with insecticides requires knowledge of what to use and where to use it. For this reason, it is suggested that pet owners consult with a licensed pest control specialist for such applications (Angelbeck-Schulze et al., 2014; Perrins & Hendricks, 2007). In line with increasing urbanization over the last few decades, flea species that infest pets have become household pests. Thus, and for reasons of animal and human welfare, the control of fleas is of great importance worldwide. Despite the increase in the number of products available and Introduction 3 their use, flea infestation of cats and dogs is still widespread in Europe and on other continents, whereas resistance of these insects against many chemicals has been detected (El-Gazzar et al., 1986). Cat fleas are the most important ectoparasite of cats and dogs worldwide. During the past ten years, topical and oral applications of insecticides such as fipronil, imidacloprid, lufenuron and, most recently, selamectin have revolutionized cat-flea control. Recent studies show that these therapies eliminate the need to treat indoor and outdoor environments, and their use markedly reduces the severity and prevalence of flea allergic dermatitis. Surveys have yet to reveal the development of insecticide resistance to these chemical compounds. Extending the longevity of these effective host-targeted therapies should be a major goal of the veterinary community (Rust, 2005). 23. Prevalence And Chemotherapy Of Gastro- Intestinal Helminths In Camels Of Cholistan Area Of Bahawalpur

To study the gastrointestinal helminths, 384 camels of different age, sex and breed was examined coprologically. For this, five gram of fresh fecal sample was carefully collected into a sealed container from each camel and was transferred to Medicine lab, UVAS, Lahore in containers with ice packs. Fecal samples were scored 1-3 based on the consistency. The individual samples was triturated in saturated salt solution, sieved and examined for helminths eggs by using different techniques i.e. Direct Smear Method, Sedimentation technique and Floatation technique while the eggs count was performed by McMaster technique. Among the camel population, the current study indicates that in Marrecha breed the prevalence of Nematodes was Trichostrongylus 1.77%, Haemonchus 8.44%, Nematodirus 3.11% and Trichuris 7.11% respectively. Mixed infestation of all the nematodes was found 26.66% in infested camels, while the prevalence of Camelostrongylus, Trichostrongylus, Strongyloides, Haemonchus, Nematodirus and Trichuris in Barella breed was 4.40%, 1.88%, 2.51%, 6.91%, 2.51% and 7.54% respectively. Mixed infestation of all the nematodes was found 25.78% in infested camels. The prevalence of Fasciola hepatica was found higher in Marrecha breed as compared to Berrela breed. It was 18.66% in Marrecha breed and 14.46% in Barella breed while the overall prevalence of Fasciola hepatica in the camels irrespective of the breeds was 16.93%. While the prevalence of Fasciola gigantica was also found higher in Marrecha breed than Barella breed. It was 16% in Marrecha breed and 10.69% in Barella breed while the overall prevalence of Fasciola gigantic in the camels irrespective of the breeds was 13.80%. The overall prevalence of fascioliosis in camel is 30.31. The prevalence of Moniezia expansa was found higher in Marrecha breed as compared to Barella breed. It was 10.22% in Marrecha breed and 8.80% in Barella breed while the overall prevalence of Moniezia expansa in the camels irrespective of the breeds was 9.63%. The sex wise prevalence of gastrointestinal Nematodes in males was 39.28% which includes Camelostrongylus 8.03%, Trichostrongylus 3.57%, Strongyloides 6.25%, Haemonchus 11.60%, Nematodirus 4.46% and Trichuris 5.35% respectively while in the female it was 25.37% which includes Camelostrongylus 4.41%, Trichostrongylus 2.20%, Strongyloides 2.94%, Haemonchus 6.25%, Nematodirus 2.57% and Trichuris 6.98%. The overall prevalence of Nematodes in Male was found higher as compared to Female which was 39.28% and 25.37% respectively. Sex wise prevalence of GI Trematodes in Camels was determined through collected samples. The prevalence of Fasciola hepatica was found higher in Female as compared to Male. It was 16.91% in Female and 15.18 % in Male while the overall prevalence of Fasciola hepatica in the camels irrespective of the sex was 19.53%. The prevalence of Fasciola gigantica was also found higher in Female than Male. It was 12.13% in Female and 11.61% in Male while the overall prevalence of Fasciola gigantica in the camels irrespective of the sex was 11.98%. Sex wise prevalence of GI Cestodes in Camels was determined. The prevalence of Moniezia expansa was found higher in Female as compared to Male. It was 14.34% in Female and 13.39% in Male while the overall prevalence of Moniezia expansa in the camels irrespective of the sex was 14.06%. The overall prevalence of Nematodes in age group >10 yrs. was higher 30.25% as compared to age group 5-10 yrs. 20% which includes Camelostrongylus, Trichostrongylus, Strongyloides, Haemonchus, Nematodirus and Trichuris 3.36%, 2.52%, 6.72%, 5.88% 4.20% and 7.56 in age group >10 yrs and 1.88%, 1.51%, 1.88%, 4.90%, 2.64% and 7.16 respectively, in age group 5-10 yrs. The prevalence of Fasciola hepatica in age group 5-10 yrs was found higher as compared to age group >10 yrs. It was 30% in age group 5-10 yrs and 16.99% in age group >10 yrs while the overall prevalence of Fasciola hepatica in the camels irrespective of the age groups was 18.75%. The prevalence of Fasciola gigantica in age group 5-10 yrs was found higher as compared to age group >10 yrs. It was 9.33% in age group 5-10 yrs and 7.55% in age group >10 yrs while the overall prevalence of Fasciola gigantic in the camels irrespective of the age groups was 8.59%. The prevalence of Moniezia expansa in age group >10 yrs was found higher as compared to age group 5-10 yrs. It was 10.92% in age group >10 yrs and 10.18% in age group 5-10 yrs while the overall prevalence of in Moniezia expansa the camels irrespective of the age groups was 7.81%. To study the effect of gastrointestinal helminths on various blood parameters of camels, blood samples were collected from 5 infected and 5 healthy camels. Samples were analyzed using Hematology Analyzer and results were compared. For therapeutic trials, a total of 30 camels positive for helminths was taken and divided into three groups A-C each group comprising of 10 animals. A fourth group D was selected comprising of 10 uninfected camels. The camels of group A were treated with dry Neem (Azadirachta indica) leaf powder at a daily oral dose of 100 gm/camel for five days, group B was treated with Albenzole granules® (Albendazole) at a dose rate of 15mg/kg bd. wt. PO once. Group C (untreated) was designated as positive control, and the camels in Group D as negative control. Efficacy was determined on the reduction of eggs in feces at day 3, 7 and 14 (post-treatment). In the current study Neem leaves and Albendazole gave following results in the camels affected with Helminths. Neem leaves cured 20% of the animals on day 3rd after the drug administration while 40% and 60% of the animals cured on the day 7th and 14th respectively. Albendazole also gave good results against helminths affected camels. On the day 3rd of the treatment 30% animals cured while 60% animals were cured on the 7th day and on 14th day 80% animals recovered. Data regarding prevalence was measured by non-parametric, chi-square (χ2), while therapeutic trails were analyzed by repeated measures one way ANOVA, using SPSS (Statistical package for social sciences), P < 0.05 was considered significant. Blood samples were collected from 100 domestic cats at Pet Center, UVAS, Lahore,from their ear tips and cephalic/saphenous vein. The blood will be immediately transferred to EDTA coated vacutainers. Stained thin blood smears were observed for intra-erythrocytic bodies and 45 samples were selected after screening. Blood in EDTA were tested for PCR (already optimized) in the Molecular Parasitology laboratory at UVAS, Lahore, to screen for B. felis. ExtractedDNA of confirmed B. felis samples were further processed forLoop-Mediated Isothermal Amplification (LAMP). LAMP primers weredesigned recognizing four sections of the B. felis gene. LAMP reactions of 25µL were standardized at 60°C temperature for 1 hour time using DNA extracted from blood samples of cats found positive on PCR. Briefly, the concentration of FIP and BIP were varied from 0.8µM to 2.4µM, Mg2+from 2mM to 4mM, betaine from 0.2 to 0.8M and dNTPs from 1mM to 4mM.The LAMP reaction was optimized at the final concentration of 0.2µM F3 and B3, 2.0µM of each of the FIP & BIP, 2mM for each dNTPs, 0.8M betaine, 1X reaction buffer, 1µl bst polymerase and 2µl DNA templates at 60°C. Nicks of this problem also found in Pakistan, however the veterinarians are usually unable to properly diagnose this disease due to lack of knowledge as well as the ancillary diagnostic equipment availability for this disease. Present study was divided into two phases. Phase – 1 included clinico-epidemiological data. To this end, target of more than 502 domesticated client-owned cats of either sex, age, breed, etc showing signs of feline lower urinary tract disease (FLUTD) as per Buffington (1994) were examined accordingly from 3 different cities (Lahore, Faisalabad, and Islamabad) of Pakistan). All data collected was based on a predesigned proforma by using structured interview of the owners. Diagnosis was solely based on serum-cortisol levels, urinalysis, radiography and ultrasonography. Phase II involved experimental trial. The data obtained from whole of the study was then presented in tabulated form as frequencies and percentages. Treatment and outcome of the disease were also analyzed accordingly. According to the present study conducted it is proved that iFLUTD is present among the cats in Pakistan. Its proper cognizance among the Pakistani veterinarians is still non-existent and is misdiagnosed as colic or constipation issues in cats. The present study was undertaken to bring iFLUTD into the reportive of small animal practitioners working in Pakistan. The present study debunked various previous notions like iFLUTD is associated with commercial diets and canned foods only if we talk about this region majority of cases were noticed that had home-cooked food given by the owner. Moreover, cases in Siamese breed are larger than Persian breed. It has been strongly associated with Indoor housing management. Additional work is still needed to explore untouched areas of epidemiology including factors other than those being studied in the previous literature. Academicians in veterinary pathology and veterinary medicine of Pakistani universities should embrace this malady in the Doctor of Veterinary Medicine (DVM) curricula. According to the present study results it is concluded that two factors like stress and pain accelerate the sympathetic nervous system outflow compared to normal felines leading to the inflammatory response. Thus the stress factor must be reduced in the form of making hiding places for cats at home to reduced down the fear factor along with enhancing the feeling of owes for that particular place. Moreover, some more practices should be performed by the owner to reduce down the stress factor like playing with the pet, giving full attention, placing toys and other attractive things like yarn balls at the feline places (where they live/placed). There was no significant difference found between the groups based on the food with health score along with the therapeutic judgment. Hence, it is recommended that more experiments should be performed on larger scale to assess GAG therapy on increased number of felines and need of hour is to conduct more veterinary studies to get information and authenticity for its use against iFLUTD. From this study conducted, I recommend to the owners that the cats must be provided with the indoor hiding places and play with their pets in order to reduce the stress factor that increases the risk of idiopathic lower urinary tract disease. In order to find the harmful effect of the theileriosis, blood hematology and biochemistry values were also be determined in both healthy and infected cattle. The samples from theileriosis positive animals were used for nucleic acid extraction. The extracted nucleic acid was amplified using Species specific primers.Agarose gel electrophoresis was used to visualize the amplified DNA. Amplicons of nucleic acid was treated with different restriction enzymes and pattern was visualized to find differences among different species of Theileria. Suitable statistical tool was used to analyze the data collected. Chi was applied on data by using statistical software SPSS version 20.0. It is difficult to diagnose the Dermatophytosis from other skin infections by routine tests in most of the cases especially subclinical. Polymerase Chain Reaction (PCR) is advanced and the most reliable technique to detect genome of Dermatophytes even in minute quantities specifically and can efficiently detect the presence of any Dermatophyte specie on the skin of dog. The current study was planned to develop and validate a diagnostic assay which could be able to detect and distinguish tree important dermatophytes species including Microsporum, Trichophyton and Epidermophytonby a uniplex PCR reaction. Analysis of involvement of certain predisposing factors in dermatophytosis was second goal to be worked on in this study. Samples of suspected pet dogs (n=50) were collected by scraping the skin at affected areas over skin. DNA was extracted from the skin scraping samples by organic Phenol Chloroform Isoamyle Alcohol method. Primers, specific to the 18-S ribosomal RNA region of genomes of the Dermatophytes, were designed after alignment of available sequences of Microsporum,Trichophyton and Epidermophyton at NCBI. Annealing temperature and recipe of PCR reaction was optimized by gradient PCR in BIO-Rad thermal cycler. Amplification reaction of all samples collected was carried out as per optimized reaction conditions, afterwards. Amplified products obtained were subjected to genotyping by agarose gel electrophoresis for size based separation of the amplified products. The specific amplified bands of desired genomic region of dermatophytes were seen in UV light transilluminator. The data of results of predisposing factors involved in dermatophytosis wasanalysedby using Pearson’s chi squared test with the help of Statistical Package for the Social Science (SPSS) Program. Genome specific product sizes of Microsporum and Trichophyton i.e. 366 bp and 351 bp in respective positive samples were observed. Out of 50 suspected samples 46 samples were positive for dermatophytosis out of which 38 samples (82.6%) were positive for Microsporum, 6 samples (13%) for Trichophyton and 2 samples (4.4%) were positive for both Microsporumand Trichophyton. This study will help to validate a diagnostic technique for Dermatophytosis with greater efficacy and reliability. A total of 49 stations were sampled for bats where total 1982 bats were recorded. A total of 21 species of bats belonging to six families, fourteen genera were recorded. M. formosus was common to all the three districts while B. leucomelas and P. pipistrellus were captured only from Dir district. The Hodgson‟s bat (M. formosus) and the Schreiber‟s long-fingered bat (M. fuliginosus) were captured from Malakand and Swat districts. The skeleton of C. sphinx was recorded only from adjacent area of Malakand district. The Indian flying fox (Pteropus giganteus) was not previously recorded from Khyber Pakhtunkhwa while it has been reported from Punjab and Sindh province of the country. Bat surveys. This is the first extensive exploration of that small portion of the Khyber Pakhtunkhwa which comprises of only three districts of Malakand Division i.e. Malakand, Dir and Swat. Although more focus remained towards Malakand district, six families, fourteen genera, twenty one species were identified. Moreover, two new country Summary 185 records (Myotis formosus and Miniopterus fuliginosis) were also explored. Further bat surveys in poorly surveyed parts of the country especially in KPK and Baluchistan may result in identification of some other new bat taxa. More bat surveys involving greater field efforts may also confirm the presence or absence of those already described from the country. 2. Distribution ranges and species specific habitat analysis. Presence of thirteen new locality records (Pteropus giganteus, Cynopterus sphinx, Rhinopoma hardwickii, Megaderma lyra, Rhinolophus Lepidus, Hipposideros fulvus, Barbastella leucomelas, Scotophilus heathii, Scotophilus kuhlii, Eptesicus serotinus, Pipistrellus javanicus, Pipistrellus dormeri and Scotoecus pallidus) and two new country records (Myotis formosus and Miniopterus fuliginosis) gives credence to the idea that distribution ranges of most of the bat species has change over the past sixty years. Thus serious scientific studies are needed to redefine distribution ranges and identify species specific habitats using global positioning system and radio-telemetric studies. 3. Reconfirmation of bat taxonomy. Genetic analysis of none of the bat species of the country has been made using molecular markers thus leaving behind a chance to doubt identification of cryptic bat species. Thus molecular genetic studies of all the bat species of the country is highly recommended which may also lead to the discovery of such bat taxa which are new to science. 4. Bat call library. The only bat detector (Patterson D 1000X) present in the country fell down from my hand in a water body and became out of order. So none of the bat could be recorded. Bat call analysis has boosted bat identification throughout the world but the Summary 186 lack of such sophisticated equipment in the country has become a major bottle neck in the establishment of a bat call library. 5. Awareness campaigns. Majority of the countrymen are unaware of the ecological services rendered by bats. Khyber Pakhtunkhwa is the major fruit growing region of the country. Based on misperceptions, the locals consider all bats as vermin and kill them ruthlessly. Conservation education to highlight the significance of bats must be included in the curriculum of children at primary school level so that they may adopt a pro-conservation attitude in the first few years of their personality building. It retards growth, decrease body condition, and lowers the clutch size which imparts huge economic losses to breeders and birds owner. Over dosage of frequently used anthelmintic increase the risk of adverse effects and stress on birds. So there arises a need to adapt alternative therapeutic approaches. The present study was designed to compare the anthelmintic activity of frequently used synthetic drugs levamisole and piperazine and herbal drug pineapple leave extracts against ascariasis in java sparrows.To study the comparative anthelmintic efficacy of levamisole, piperazine and pineapple leave extract against Ascaridia galli in captive java sparrows, fecal sample of (n=200) java sparrows captive for breeding purpose were screened for the Ascaridiagalli by direct fecal smear method. Among the all the infected birds 30 infected and 10 healthy birds were selected to conduct the study and divided into 4 groups and included in therapeutic trial.Birds were treated with levamisole, with piperazine and with pineapple leaves extract according to standard dose rate and route of administration in Group A, B & C respectively, Fourth group D comprising of 10 birds waskept as negative control. To evaluate the comparative efficacy of drugs, EPG will be calculated according to McMaster Technique post treatment at day 0 and after treatment on 7th, 14th and 21s. Results revealed that levimosle was the very effective against Ascaradia galli in java sparrows. Levaimsole decrease the average EPG of treated group to 50 from 600 and show 100 percent efficacy against Ascaradia galli and recovered all the infected birds of the group A. Piperazine was less effective as compare to the levamisole but show more effectiveness as compare to the pineapple leave extract. Piperazine reduced the average EPG of treated group B to 150 from 650 and show 70 percent and recovered 7 infected from 10. Pineapple leave extract showed least effectiveness and reduced the EPG of treated group C to 350. Pineapple showed 40 percent efficacy against Ascaradia galli and recovered only 4 infected birds of group C from Ascariasis.Pineapple leaves extract were effective against ascariasis in captive Java sparrows but least effective as compare to levamisole and piperazine having no adverse effects.
