1. Clinico-Therapeutical And Haematological Studies On Babesiosis In And Around The Livestock Experimental Station Qadirabad in Lohi Sheep

The disease results into death due to fever, excessive destruction of RBC's, anemia, haemoglobinuria and icterus. The present study was conducted to find out the prevalence of babesiosis in Lohi sheep in and around the Livestock Experimental Station, Qadirabad and to study the effect of 2 anti-protozoan (babesicidal) drugs i.e.Diminazene diaceturate (Diminazene; star lab.) and Imidocarb dipropionate (Imizol; ICI) on the recovery and health of animal. The effect of hernosporidian parasites on various blood parameters was also studied. The parameters included: Haemoglobin (Hb) estimation and Packed Cell Volume (PCV). For this purpose 310 Lohi sheep were examined for the presence of babesiosis. Out of 310 Lohi sheep, 30 were found positive showed 9.67% prevalence. For treatment protocol, thirty naturally infected and ten noninfected sheep were selected and divided into four groups having ten sheep in each group i.e. A, B, C and D. Diagnosis was confirmed on the basis of blood smear method, which revealed the presence of Babesia. The parasite appeared as piriform bodies within the light pink erythrocytes. The animals of group A were treated with diminazene (Diminazene diaceturate) at a dose rate of 3.5mg/kg body weight at zero day. Three out of ten animals recovered at 3rd day of first injection, three animals recovered at 7th day of first injection, two recovered at 10th day of first injection, while two animals did not show any improvement and might be chronically infected. This drug showed 80% efficacy at 10th day while the animals of group B were treated with Imizol (Imidocarb dipropionate) at a dose rate of 2mg /kg body weight at zero day. Six out of ten animals recovered at 3rd day of first injection, three recovered at 7th day of first injection and remaining one recovered at 10th day of first injection. This drug showed 100% efficacy at 10th day. Animals of group C were served as infected untreated control while animals of group D were served as healthy control. Efficacy of drugs was detected by negative blood smear. Imizol gave best results where as Diminazene was second in efficacy against babesiosis. Haematological examination revealed a significant decrease in .Hb and PCV in group A and C on 3rd, 7th and 10th day post medication as compared to group B. From the results of present study, it has been concluded that Imidocarb dipropionate (Imizol) was the drug of choice for the treatment of ovine babesiosis in Lohi sheep when given at the dose rate of 2mg/kg body weight by intramuscular route. 2. Comparative Therapeutic Trials Against Coccidiosis In Dogs

Material type: book Book; Format: print ; Literary form: not fiction Publisher: 2005Dissertation note: The present project was designed to find out the prevalence of coccidiosis (Isosporiosis), to evaluate the comparative efficacy of Co-trimoxazole &Furazolidone and to study the effect of Isosporiosis on blood parameters. For this purpose the faecal samples of two hundred dogs were examined in the Medicine laboratory, University of Veterinary and Animal Science, Lahore by direct smear method, and also concentration technique for identification of Isospora oocyst. Out of two hundred dogs thirty-six were found positive for the coccidial infection, showing the prevalence of Isosporiosis as 18%. For chemotherapeutic trials, thirty naturally infected and ten non-infected dogs were selected and divided randomly into four groups having ten dogs in each groups viz A, B, C and D. Group A was treated with Co-trirnoxazole at the dose rate of 15mg/kg (orally) and group B was treated with Furazolidone at the dose rate of 8mg/kg (orally). Group C was kept as infected non-medicated control. Group D was kept as non-infected and non-medicated control. Faecal samples of experimental dogs were examined for OPG counts on day zero (pre-medication), 3rd, 7th and 10th day of medication and percentage reduction of oocyst was calculated. Cotrirnoxazole showed 63%, 90%and 97% efficacy whereas Furazolidone showed 63%, 89.8%and 95% efficacy on day 3, 7 and 10, respectively. Statistical analysis of data revealed non-significant difference (P>0.05) between the groups A, B and D (-ye control) on day 7 and 10 of treatment. Based on criteria of percentage reduction in the number of oocyst in faeces, it was observed that Co-trimoxazole was the most effective (97%) followed by Furazolidone (95%) against Isosporiosis in dogs. Haematological study revealed that the values of haemoglobin and packed cell volume were lower in groups A, B, and C as compared to non-infected control group. Whereas as a result of treatment in-group A and B the values of haemoglobin and packed cell volume gradually increased on day 3, 7 and 10 post-medication. It has been observed during the present study that Cotrimoxazole and Furazolidone showed their effect on blood parameters as a result of curing Isosporiosis. 3. Incidence, Chemotherapy And Haematology Of Strongylosis In Horses Of District Layyah

Material type: book Book; Format: print Publisher: 2007Dissertation note: The present project was designed to find out the infection rate of strongylosis, to test the anthelmintic efficacy of Endoectiven, Vermox, Neem (Melia Azadarch linn) and Oxfendazole, to study the effect of strongyloids on blood parameters in horses of district Layyah. Horses were examined for the presence of strongylosis coprologically, till 50 horses naturally infected with strongyle spp. become available. Fifty horses were found positive for strongylosis out of eighty seven horses, so the infection rate of strongylosis was 57.47%. Fifty positive animals were randomly divided into five groups A, B, C, D and E each comprising of 10 animals. Another group F comprising of healthy horses was made. Each animal of group A was treated with Endoectiven (8 mg/kg body weight.), group B was treated with Vermox (10 mg/kg body weight), group C was treated with Neem (375 mg/kg body weight), group D was treated with Systamex (10mg/kg body weight) while group E was kept as positive control (untreated) and group F was kept as negative control (uninfected untreated). The efficacy of anthelmintics was calculated on the basis of reduction of eggs per gram (EPG) of faeces on day 7 and 14 post treatment. Efficacy of Endoectiven (Closantel) was 0 % and 0 % on day 7 and 14 respectively. Efficacy of Vermox (Mebendazole) was 91.93% and 95.16% on day 7and 14 respectively. Efficacy of Neem (Melia Azadarch linn) was 6.89% and 8.62% on day 7 and 14 respectively. Efficacy of Systamex was 96.82% and 98.4 1% on day 7 and 14 respectively. It was concluded from the above results that Oxfendazole was found most effective than Mebendazole, Neem (Melia Azadarch linn) and Closantel, while Mebendazole showed better results than Neem (Melia Azadarch linn) and Closantel. Neem (Melia Azadarch linn) showed better results than Closantel. Haematological parameters showed significant reduction in total erythrocyte count (TEC), haemoglobin (Hb) level and packed cell volume (PCV). 4. Disgnosis Of Theileriosis In Buffalo Through Blood Smear Examination And Pcr In District Lahore

Material type: book Book; Format: print ; Nature of contents: biography; Literary form: Publisher: 2008Dissertation note: The present study was designed to determine diagnosis and infection percentage of Theileriosis in buffalo in and around District Lahorë, and to design the primer for Theleria parasite with Microscopic Examination. For this purpose blood samples were collected from 300 buffaloes randomly from 20 villages, during the month of May, June, July and August of 2007 in and around District Lahore. Screening was done by blood smears, stained by Giemsa’s staining technique and later the blood sample from same animals was also processed by PCR. The blood smear showed Theileria, piroplasms, including cocci, rod, and signet-ring, with diameter of 0.5-1.5 micrometer. The blood parameter i-e PCV, Hb concentration and TEC showed presence of microlytic hypochromic anemia in diseased animal. On the basis of microscopic examination overall 44.66% (134/300) prevalence was recorded. On the basis of PCR test prevalence of Theleria parasite 5 9.66% (179/300) was recorded show the presence of carrier animal in buffalo population in district Lahore. The result had shown high efficacy of PCR as compare to microscopic examination. It is anticipated that present study was proved helpful in diagnosis of Theileria in infected as well as in carrier animals in District Lahore and will be beneficial for further study. 5. Detection And Chemotherapy Of Sub Clinical Mastitis In Dairy Catlle And Buffaloes

Material type: book Book; Format: print Publisher: 2008Dissertation note: The aim of this study was to detect subclinical mastitis in bovine and to find out the most appropriate antibiotic for its treatment. For this purpose, Milk samples were taken aseptically from 500 apparently healthy animals (n= 250 cattle and n= 250 buffaloes) for screening tests. Two different tests, Surf Mastitis Test (SMT) and Somatic Cell Count (SCC) were used. Milk samples declared positive by both of the above mentioned tests, were subjected to culture sensitivity test. Six different antibiotics were evaluated i.e. Enrofloxacin, Norfioxacin, Amoxicillin, Oxytetracycline, Gentamicin and penicillin G. On the basis of sensitivity test; two topmost drugs were selected and be given to two equal groups of animals. Each group was comprised of equal number of cattle and buffaloes. Each antibiotic was given for 5 days as intramuscular (IM) injection. A positive and a negative control groups were also kept. To detect the in-vivo efficacy of antibiotic, again the milk samples of all the groups were examined by SMT and SCC on day 10 and day 20 of first injection. The results obtained were the following; Sub clinical mastitis was found 42.8% in cattle and 37.6% in buffaloes. Out of the total of 201 mastitis positive milk samples cultured, bacterial growth occurred in 98 (48.75%) of milk samples. The various bacterial species isolated from milk samples of cattle and buffaloes were E. coli Staphylococcus, Streptococcus, Proteus, Kiebsiella, Pseudomonas, and Pasteurella with overall percentage of 50 %, 17.34 %, 3.06 %, 12.24%, 10.20%, 2.04% and 5.10 % respectively. The in vitro efficacy of Enrofloxacin was found to be the best one i.e. 77.55 % followed by Norfioxacin (67.34 %), Gentamicin (53.06 %), Oxytetracyclin (30.61 %), Amoxicillin (22.44 %) and Penicillin G (4.08 %). After chemotherapy, there was significant difference between treatment groups and control groups (P<0.05). The difference in the efficacy of enrofloxacin and norfioxacin was statistically non significant (P>0.05), however mathematically, the recovery rate by enrofloxacin was greater (93.75%) then norfioxacin (87.50%). 6. Clinico -Therapeutical Trials Of Lactic Acidosis In Small Ruminants

Material type: book Book; Format: print ; Literary form: not fiction Publisher: 2009Dissertation note: During the present study clinical examination of 200 small ruminants (sheep and goats) any breed, age and sex with the history of diarrhea was performed at the Outdoor Hospital, University of Veterinary and Animal sciences Lahore and adjacent areas. A total of 38 animals were diagnosed positive for the disease. Temperature, pulse rate, respiration rate, rumen pH, serum pH, ruminal movements and ruminal protozoa of affected animals were studied before and after treatment with two drugs magnesium hydroxide and sodium bicarbonate. It was found that rectal temperature, rumen pH, blood pH, ruminal movements decreased significantly in lactic acidotic sheep and goats before treatment as compared to control group. While increased pulse and respiration rate were observed in lactic acidotic group before treatment as compared to control group. Thirty diseased and ten healthy animals were divided into four groups i.e. A, B, C and D each having ten animals. Group 'A" was provided with magnesium hydroxide at a dose rate of 1.11 g/ Kg body weight orally mixed in water at a rate of 450 g/ 3.5 liter once, group "B" was treated with sodium bi carbonate at a rate of lg/ kg body weight orally once, group "C" was kept as diseased untreated control and group "D" acted as healthy control. Mean values obtained before treatment were rectal temperature 99.74 F° + 0.67153, lOO.59F° 1 0.75580, 101.25 F° ± 0.49448 and 102.86 F° ± 0.30155 for group A, B, C and D respectively. Rectal temperature values after treatment were 102.47 F° ± 0.2 1294, 101.9 + 0.27849, 101.19 F°±0.41965 and 102.95 F° ± 0.16142 for group A, B, C and D respectively. Mean pulse rate values for group A, B, C and D before treatment were 100.6/mm ± 3.40008, 97.2/mm ± 4.24745, 103.6/min ± 2.5636 and 86.3/min ± 1.85626 respectively while after treatment were 88.55/min ± 1.77768, 92/min ± 1.75123, 94.3/min ± 1.90938, 86/min ± 1.06460 respectively. Mean respiration rate values before treatment were 24.1min ± 0.86218, 28.2/min ± 2.52020, 24/min ± 0.84329 and 19.8/min ± 0.59256 for group A, B, C and D respectively while after treatment were 20.55/min ± 0.78953, 22.8/min ± 0.80001, 24.5/min ± 0.79234 and 20.7/min ± 0.66751 respectively. Mean rumen pH values obtained before treatment were 4.43 ± 0.14533, 4.49 ± 0.13287, 4.76 + 0.17075 and 6.39 ± 0.13699 while after treatment were 5.66 ± 0.17125, 5.41 ± 0.13506, 4.94 ± 0.18683 and 6.42 ± 0.08035 for group A, B, C and D respectively. Mean d pH values obtained before treatment were 7.18 ± 0.04667, 7.1 ± 0.05375, 7.21 ± 0.05044 and 7.33 ± 0 .02808 while after treatment were 7.37 ± 0.02806, 7.25 + 0.02390, 7.18 ± 0.03837 and 7.41 + 0.0383 7 for group A, B, C and D respectively. Ruminal protozoa were completely absent in ruminal fluid of lactic acidotic group. Ruminal stasis was also observed in lactic acidotic group. 7. Comparative Efficacy Of Guar (Gyamopsis Tetragonoloba) And Salinomycine Sodium Against Coccidiosis in Broiler Chicken

One hundred and twenty (120) day old broiler chicks free from coccidial infection were purchased from local market. The broiler chicks were divided into four groups viz. A, B, and I) each comprising of 30 birds. Each group was further divided into three replicates comprising 10 chicks each. All groups except group D was induced 10,000 sporocyst of Eimeria species orally on day 14th. Guts from the broiler chicks suspected with coccidiosis were collected from the field and local poultry farms. The coccidial oocysts in faeces were isolated and sporulated. Oocysts were collected from the gut samples. The counting of oocysts in samples was performed by McMaster technique. The experimental birds were vaccinated against various diseases i.e., Gumboro (IBI)) and Newcastle Disease. Salinomycin and guar was used against coccidiosis as per medication protocol the dose rate of 5 gm/kg in feed and Sgm/95 gm of feed given to group A and B respectively. While group C consist of positive control and group I) was negative control. OPG, weight gain and FCR was also being recorded as well as postmortem was performed in case of mortality. For statistical analysis one way ANOVA technique was used. It is concluded that Salinomycin 5 gm/kg in feed had shown best results in terms of maximum weight gain. FCR as compared to Guar 5gm/95 gm of feed which shown reduced oocyst count and mortality. It was not clear which specific compounds in guar contributes to the anticoccidial or growth inhibitory effects. Further research is needed to investigate whether the anticoccidiosis effect of guar was due to residual guar gum (galactomannans), saponins, or some unknown component of guar. It is anticipated that the results of this study will help the poultry farmers and other concerned peoples to choose best possible drug to eradicate coccidiosis and take maximum production. 8. A Study Of Giardiasis And Its Chemotherapy With Metronidazole And Albendazole In Dogs

The objective of the study was to determine the percentage of giardiasis in dogs, its treatment and blood examination before and after treatment. The study was conducted in pet center of university of veterinary and animal sciences (UVAS) and private clinics in Lahore; collected sample were processed in medicine laboratory, department of clinical medicine and surgery (UVAS), Lahore. Faecal samples of five hundred dogs suspected with giardiasis were collected and assessment of giardia cyst were seen using zinc sulphate floatation technique and sucrose gradient centrifugation method, then the positive dogs were separated and divided into four groups viz A, B, C, groups D was negative control, group A was treated with Metronidazole and its efficacy wasll.3 %,64 % & 85 % on day 31,5th & 7th day post treatment while group B was treated with Albendazole & its efficacy was 16.3 % ,44.2 % & 74 % on day 31,5th & 7th day post treatment the results showed that the Metronidazole was better in efficacy then Albendazole, C group was serve as positive control & all these results were compared with negative control (healthy dogs) that is group D.The efficacy of the drugs was determined by the disappearance of giardia cysts from feceas. The blood examination total leukocyte count & Differential Leukocyte Count was conducted before & after the treatment with drugs & it showed that there was mild increase in TLC but not more increase in DLC & no adverse effect of drugs on blood parameters and values were almost normal and close to the values of negative control(healthy dogs). It has been observed during the present study that metronidazole & albendazole showed their mild effect on blood parameters as a result of curing giardiasis. Statistical analysis of data revealed non significant difference (p>O.O5) between groups, A, B, & D (-ye control) on day 3 & 7 of treatment. it was observed that metronidazole was the most effective 85% followed by albendazole 74% against giardiasis in dogs. 9. The Efficacy Of Chemotherapeutic Agents Against Paramphistomum In Sheep

Material type: book Book; Format: print Publisher: 2011Dissertation note: The present project was carried out to diagnose the paramphistomum infection in sheep and to evaluate the efficacy of oxyclozanide and albendazole and to observe the effect of paramphistomum on various blood parameters, haemoglobin estimation, erythrocytes sedimentation rate and packed cell volume. For this purpose twenty four Sheep were selected and divided into 4 groups. Each group comprised of 6 animals. Faecal samples were collected from Out Reach centre Sitar wala and its adjacent areas and out door clinic UVAS Lahore for the diagnosis of paramphistomum. The faecal and blood samples were collected on day zero pre-medication, and day 7 and 10 post-medication. The drug efficacy was checked at those animals which were positive for the paramphistomum. The animals in Group A were treated with Oxyclozanide (Zanil) at a dose rate of 15 mg/kg body weight. The animals in Group B were treated with Albendazole (Albensel) at a dose rate of 10 mg/kg body weight. The animals in Group C were kept as positive control (infected-untreated) and Group D were kept as negative control (uninfected-untreated). Eggs per gram (EPG) of the faeces were noted on day 0 day before medication. The efficacy of these drugs was calculated on the basis of reduction of eggs per gram (EPG) of faeces after treatment. Zanil showed a progressive decrease in the faecal egg counts. The efficacy of Oxyclozanide (Zanil) was 58.33% on day 7 and 83.33 % on day 10. The efficacy of Albendazole (Albensel) was 36.36% on day 7 and 56.36 % on day 10. It was observed that Oxyclozanide (Zanil) showed better efficacy than Albendazole. Haematological parameters showed different results. A marked decrease in haemoglobin was observed after the study of infected sheep and the haemoglobin value was in normal range in healthy sheep on day zero. Packed cell volume was reduced on day zero in all infected groups, but was normal in the group of healthy sheep. After the treatment these values increased towards normal range in group A and B. The erythrocyte sedimentation rate was increased after 24 hours on day zero in all infected groups A, B, and C but was normal in the group D of healthy sheep. The peak incidence of subclinical ketosis occurs during the fourth week of lactation. There are indications that cows with the highest milk yield directly after calving are at greatest risk for developing ketosis. Subclinical ketosis causes delayed reproductive functions, increased intervals from calving to first and last service, and an increased frequency of ovarian cysts. The study was designed to diagnose the Subclinical ketosis on the basis of Ross Rothra's method. This method was easy to conduct and can be able to perform at the farm. The purple color indicated the presence of ketone bodies in the sample. After diagnosis the treatment trails was done on three different treatment methods comprises of Dextrose, Dexamethasone and by increasing the diet. Total 200 cow were studied for the diagnosis of subclinical Ketosis. Total positive samples of subclinical ketosis were 62 and the overall Incidence of Subclinical Ketosis in was 31 % and in sheikhupura and Raiwind areas it was 35% & 27 respectively. The results indicates that the use of dexamethasone was very effective and it cured 84%,dextrose cured 64% ,and by the diet it was 25%.Although, dexamethasone was very effective but it also reduced the milk production in very next day after the administration of drug while, the dextrose shows no any reduction in milk production. The animals who received only green fodder as ration showed very positive results for subclinical ketosis while, the animals who received a concentrated diet with green fodder they showed very little presence of subclinical ketosis. Prevention of subclinical ketosis can be done by providing extra ration to pregnant animals and during pregnancy give them pregnancy allowance and by effective managemental practices the effects of subclinical ketosis can be minimized. In the present study a total of 100 feacal samples were collected from deer and were examined in the Clinical Medicine Laboratory, department of CMS, University of Veterinary and Animal Sciences, Lahore. Out of 100 samples thirty eight were positive and among these thirty eight, 12 deer were infected with cestodes, 6 with nematodes, 12 with trematodes, and 8 with the mixed infection of parasites. So, the prevalence of the cestodes, trematodes, nematode and mixed infection was 12%, 12%, 6% and 8% respectively. For the chemotherapeutic trails thirty positive animals and 10 healthy animals were divided into four groups A, B, C and D with 10 animals in each. The animals of group A was treated with albendazole @ 7.5mg/kg orally. Those of group B was treated with the ivermectin @ 1mg/ 50kg orally. The group C animals were kept as positive control and group D were kept as negative control. The feacal samples were collected on the 0 day (pre-treatment), 7th, 14th and 21st day (post-treatment). The efficacy of the drugs were calculated on the basis of reduction of the eggs count from the feacal samples on the respective days. The efficacy of albendazole 42.59 %, 71.3% and 87.96% against cestodes, 33.33%, 60.3% and 82.88% against nematodes and 40.18%, 68.22% and 89.72% against trematodes at day 7, 14 and 21 respectively.. The efficacies of ivermectin were 42.59 %, 73.15% and 88.88% against cestodes, 51.78%, 76% and 91.07% against nematode while 42.85%, 64.28% and 85.71% against trematodes at day 7, 14 and 21 respectively. It is thus concluded that the ivermectin is more effective drug than albendazole as it caused more reduction of the egg count than albendazole. The present study was conducted on cryptosporidiosis, and to evaluate the comparative efficacy of paromomycin and Kalvanji therapy in lambs in and around Lahore Pakistan. In this plan of research work 400 lambs, less than 1 month of age suspected for cryptosporidium infection on the basis of clinical signs were included. The fecal sample were collected from sheep presented at medicine outdoor hospital of University of Veterinary and animal sciences, various veterinary hospitals in and around Lahore and was processed in medicine laboratory UVAS ,Lahore for observing the oocyst of cryptosporidium by using Ziehl-neelsen technique. The prevalence was found to be 24.25%., The 40 sheep were divided into four groups=designated as A, B, C & D. Each group was comprised of 10 sheep. In group A the infected sheep was treated with paromomycin@ 100 mg/kg orally for 5 days, Group B infected sheep was treated with Kalvanji@1/2 tea spoon for 5 days. Group C infected sheep kept for positive control, whereas healthy sheep group D were kept in negative control. After the 5 days treatment The effect of drugs were checked, by reduction of oocyst, at 0, 7 and 14 day. Oocysts of cryptosporidium were counted by using Mc Master Technique. The therapeutic results of this study concluded that paromomycin was much effective for cryptosporidiosis. we also done hematological studies on 40 sheep(n=30 sheep with cryptosporidiosis and n=10 healthy sheep) was included to see the effect of drugs on various blood parameters i.e PCV,TLC, The 5 ml blood were collected from each lamb directly from jugular vein, and were shifted into a sterilized plastic bottle coated with EDTA@1mg/ml were further analyzed through hematological analyzer . In disease condition the TLC and PCV was increased. The results showed the decrease number of TLC and PCV. One way ANOVA was used to analyze the data. The outcome of this study would helped to control cryptosporidiosis in lambs and reduce their substantial losses rendered by cryptosporidium. The physical condition and production in animals is severely affected by tick and mite infestation. It is one of the most common problem of sheep characterized by anemia, poor physical condition, decreased milk quality and poor meat production.. The present study was conducted on ticks and mites to study their prevalence, their effect on various blood parameters and to evaluate the efficacy of Ivermectin and Azadirchta indica (neem oil) on sheep in and around Lahore, Pakistan. In this study 300 sheep suspected for tick and mite infestation on the basis of clinical signs were collected from outdoor hospital of University of Veterinary and Animal Sciences Lahore, Outreach hospital Sitar wala and Civil Veterinary Hospital Thokar Niaz Biag. The tick sample were collected with the help of foreceps and mites sample through skin scraping. Out of this 300, 198 samples were collected from outdoor hospital UVAS Lahore, from which 84 were positive with prevalence of 42.42% for tick and mite infestation, and 62 samples were collected from outreach hospital Sitarwala from which 24 were positive giving prevalence of 38.70% and 40 samples were collected from CVH Thokar Niaz Biag, out of which 17 samples were positive with prevalence of 42.5%. All the collected samples were processed at medicine laboratory, UVAS, Lhr. Whereas in the third part therapeutic trials on 20 sheep (n=15 tick and mite infested and n=5 healthy sheep) was done and divided into four groups A, B, C&D. Each group was comprised of 5 sheep .In group A, the infested sheep were treated with two doses of ivermectin 0.2mg/kgS/C 7 days apart, on 7th day its efficacy was 40%and on 14th day it gave 100% efficacy. In group B the infested sheep were treated with neem oil (Azadirachta indica) @1 table spoon, its efficacy was 20% on 7th day and on 14th its gave 40% efficacy, in group C infested sheep were kept as positive control, whereas healthy sheep were included in group D. In the second part of present project haematological studies on 10 sheep (n=5 tick and mite infested sheep, n=5 healthy sheep) were performed to see the effect of tick and mite infestation on various blood parameters (Hb, PCV,TLC) and it was fsound that Hb and PCV values were decreased whereas TLC was increased in infested sheep. The therapeutic trials of this study concluded that the Ivermectin was most effective drug against ticks and mites in sheep. In Pakistan parasitic infestation is quite prevalent and the loss about 26.5 million rupees annually to the livestock industry. Although no exact figure of economic losses is available but it is fact that millions of rupees are being lost due to reduced milk yield, rejection of meat and edible offal, depreciation of hides, delay age of maturity and death particularly in calves and high production cost due to use of drugs, they also decrease feed intake efficiency. Present study was conducted to check the burden of endoparasites in different live animal Markets of Lahore, For this purpose 800 samples of large ruminants (cattle, buffalo) and small ruminants (sheep, goat) were collected and transported to lab for further qualitative fecal examination through macroscopic and microscopic examination. In six live animal markets overall prevalence of endoparasites found 21.37%. Out of 800 samples 171 were found positive. Prevalence of large and small ruminants was found almost similar i.e: 21.5%, 21.25% respectively. However much parasitic burden was observed 43.1% in cattle to that of 28.1% in buffalo.Prevalence in sheep was high as compared to goat whereas kacchi breed of sheep and Desi breed of goat showed higherprevalence that as 14.5%, 16% respectively.Prevalence in Nilli ravi was 28.0% in non-exotic cattle it was 31% however in exotic jersey breed prevalence of endoparasites was 11%. Endoparasites found more prevalent in Nilli Ravi. In ruminants prevalence found high in male animals as compared to female animals. Overall prevalence in ruminants was 21.0% while in male and female animals prevalence was 21.8%, and 20% respectively. prevalence found more in less than 1 year aged animal than above this age i.e, in less than 1 year of age was 22.4% prevalence while above 1 year aged animals it was 20.0% (P<0.05 ). In large and small ruminants nematodes were more prevalent followed by trematode, cestode, protozoa, and mixed infection. Among nematodes haemonchus were found more prevalent gastrointestinal parasites. Out of 171 positive samples 46 were of haemonchus (5.75%) followed by ostertagia 18 (2.25), trichostrongyloids 17(2.12), cooperia 8(1.0%), bunostomum 6(0.75%), trchuris 11(1.37%), oesophagostomumm 5(0.5%), in large ruminants. Small ruminants also found chabertia ovine 1(0.0125%) and strongyloid papillosus 1(0.0125%) infestation. During this study overall prevalence of Nematode, trematode, cestode, protozoa, and mixed infection found 15.0%, 3.5%, 1.875%, 0.625%, and 0.25% respectively. Mixed infection only found in live animal market 1 at Multan road. In live animal Market 2 at Sagyan Bridge only nematode and trematode were present. In live animal Market 3 at shahpur kanjran no mixed infection was found. And protozoa were only found in large ruminants. In live animal Market no protozoa and mixed infection was found. In live animal Market 4, 5, 6 no protozoa and cestode found. The data obtained was analyzed through chi -square test (SPSS) and probability level of <0.05 was considered as significant different. As results show ( p< 0.05) data was considered as significantly different. Fascioliasis, parasitic gastro-enteritis, coccidiosis, theleriosis, and babesiosis are major parasitic problem in ruminants in order to priority. There is urgent need to carry out research on applied aspects leading towards control of parasites. This parasitic research has been focused on a limited part of country. Tick-borne diseases include babesiosis, theileriosis, anaplasmosis, lymedisease . Among these tick-borne diseases anaplasmosis is one of the vital diseases which is responsible for the substantial economic losses in term of high morbidity, mortality and production losses that are decreased in milk, meat and other important livestock productions. Throughout the world, the “tick-borne diseases” are extensivelyspread in sub-tropic and tropicareasand Pakistan also includes in these areas(Khan et al. 2004). Moreover, anaplsamosis is non-contagious disease but also known to be infectious, transmission of which occur through mechanical means that includes the flies or tick bites. As well as different equipment that are using during tattooing or castration, in dehorning and needles also causes transmission (Aubry & Geale. 2011). Anaplasma marginale (A. marginale) is the major cause of Bovine anaplasmosis and this speciesbelongs to the genus Anaplasma (“Rickettsiales, Anaplasmataceae”). The most important 20 ticks species which cause transmission are Argaspersicus, Boophiludecoloratus, B. microplus, B. annulatus, Dermacentoroccidentalis, D. andersoni, D. variabilis, D.albipictus, Hyalommaexcavatum, Ixodesricinus, Ornithodoroslahorensis, Rhipicephalussimus, R. saniguineus, and R. bursa(Marchette & Stiller. 1982), on the other hand, Boophilusmicroplus found as a major contributor in cause of anaplasmosis (Tick Fever Research Centre, 1996). The family anaplasmatacease was classified in 1957 of the order Rickettsiale. In the genera Anaplasma, Aegyptianella, Haemobartonella and Eperythrozoon, this family was rationalized (Ristic&Kreier. 1984), and projected a combination based on analysis of genes sequences for protein on surface and ribosomal RNA (16S)(Dumler et al. 2001; Walker & Dumler. 1996). Anaplasma is a rickettsial, obligate intracellular bacteria found in red blood cells. Disease is characterized by fever, anemia and jaundice. Infection is transmitted by ticks or mechanically by biting insects or contaminated hypodermic needles or surgical instruments (Tylor et al. 2007).The most significant parasite is Anaplasma speciesthat is transmitted by at least twenty different species of ticks. But among those all mostly Bmicroplus causes Anaplasmosis (Rajput et al.2005). A. marginale is etiological agent of bovine anaplasmosis (Bram, 1957; Dumler et al., 2001; Kocan et al., 2000). A. marginale persistently infect both cattle and ticks and help as infection reservoirs(Kocan et al. 2003). Bovine erythrocyte membrane bound parasitophorous vacuoles is the site for replication of this obligate intracellular organisms. Dogs, humans, wild and domesticated ruminants are the species effected by A. marginale. This anaplasma genus is an obligate intracellular parasite of vertebrate hosts. In cattle, theA. marginal, A. phagocytophilum, A. central, and A. bovisare major pathogenic species producing diseases (Inokuma et al. 2007). Researchers suggestedthat in cattle, some unidentified Ehrlichia or Anaplasma species are present (Awadia et al. 2006). For this pathogen, which cause the persistent infection, the reservoirs are generally present in different host like tick or mammalian (Kocan et al. 2004). The wide range of different ruminant can be affected by Anaplasmabecause the species of anaplasma are not considered as strictly specific for any particular host specific(Kuttler. 1984). The A. marginalecan only be developed inside the RBCs of bovine (Richey et al. 1981). After invading the erythrocyte the tick multiplies and form almost eight initial bodies, they then get enlarge in the outer membrane and give it an appearance of a large dot. The organism gets mature in the outer membrane of RBC and force the infected RBC to get rupture due to infection. By rupturing the outer membrane where it previously multiplied come into the blood stream to infect the other RBCs. On the progression of progresses, more number of erythrocytes is infected by this organism and afterward destruction of erythrocytes occur (Stewart et al. 1981). Upon the clinical signs and symptoms are appear when the 15 percent of the RBCsare affected by the parasite(Radostits et al. 2006). The symptoms of the disease are pyrexia, anemia, icterus, hematuria, anorexia, muscular tremors, dyspnea, depression, lethargy, constipation and yellow colour of mucous membrane (Bram et al. 1983), low milk production, miscarriage and sometime mortality (Alderink and Dietrich. 1981). The mature RBCs are the site for multiplication after the invading by A.marginale. In the acute case of Anaplasmosis the higher number of RBCs are affected by the parasite for example more the 109 red blood cells per ml and after that the disease is categorized by death, abortion, loss of weight and anemia, and the survival cattle that are infected act as reservoirs for transmission of the disease in the herd level(De Echaide et al. 1998).The haematological profile can be used as useful diagnostic tool in anaplasma infection(Van Wyk et al. 2013).There is significant decrease in packed cell volume (PCV), Hemoglobin (Hb) and erythrocytes count.Serum biochemical analysis showed increased total protein,bilirubin and alanine aminotransferase enzyme (Sharma et al. 2013). A diagnosis ofcattle anaplasmosis may be made tentatively based on geographical location, seasonal variation and presenting clinical signs or necropsy finding in infected animals (Kocan et al. 2010). A.marginale,can affect throughout the life of cattle but disease severity is dependent on the age of the cattle. The clinical disease is less occurs in the calves and disease is very rare under the age of 6 month. The mild type of the disease can occur in the animal, which are between the ages of six month to one year. The acute type of the disease occurs in the animals between the ages ofone to twoyears but this disease is never fatal for the animal between these age groups.Whereas if the acute type of the disease occur in the adult animal that are more than two year aged can face the serious consequences and this disease can cause the death of the cattle with higher percentage (29 to 49%) of mortality (Kocan et al. 2003; Richey. 1991). Beside the animal age at the infection time, onceA. marginale infect animals, the animals become carrier for the infection throughout of animal life, with or without the clinical signs of the infection (Richey et al. 1991). The animal with the strong immune system are recovered from the acute type of anaplasmosis (Palmer et al. 1989). The regular cycle of ten to fourteen days are present in the carrier animals and in this cycle there are more or less number of RBCs are infected with the pathogen(Kocan et al. 2003;Kieser et al. 1990; Viseshakul et al. 2000). For the proper diagnosis of the disease and quality control in livestock, the most important aspect is the preciseor correct detection of pathogen that is spread by the tick.In 1990, with the beginning of the diagnosis with the techniques on the molecular bases the researcher produced more accurate and sensitive techniquesfor pathogenic species determination and these techniques are still used till to date (Ahantarig et al. 2008). The Giemsa staining method is used for the diagnosis of anaplasmosis in bovinesthat can be achievedby finding ofA.marginalein smear of blood from animals that are affected clinically. For the carrier animals or pre symptomatic animals thistechnique is not reliable. In these types of conditions, the serological tests are used for general diagnosis of infection by detection of antibodies. This is confirmed by molecular detection methods. A.marginaleappears as “dark staining blue purple bodies” measuring about 0.3 to 1 µm in length. A. centrale inclusion bodies are well differentiated by their location. Serological diagnosis can be done for the identification of infected cattle, DNA based test can be used for the molecular diagnosis(Stuen et al. 2011). The conventional parasitological technique like Giemsa staining always remained gold standard for diagnosis of Bovine anaplasmosis. Light microscopy of thin blood smears stained with giemsa stain may facilitate demonstration of A. marginale organisms in the erythrocytes (Kocan et al. 2004) .As giemsa staining method is not applicable for the detection of subclinical infection. Therefore PCR can also be used as a diagnostic tool for the detection of A.marginale in this technique primer pairs are used which resulted in the amplification of only their target DNA i.e. A.marginale(Munderloh et al. 2004). In the United states, for the treatment of anaplasmosis, the specific approved compound is the tetracyclines that include chlortetracycline or oxytetracycline(Kuttler. 1980). Oxytetracycline, Chlortetracycline and Tetracycline can be used to treat anaplasma infection in cattle .Among these Oxytetracycline is the most used drug at the dose rate of 10mg/kg body weight. While single dose of imidocarb dipropionte can be used at the dose rate 3mg/kg body weight (De Waal 2000).Imidocarb dipropionate with the dose of 3 milligram/kilogram body weight repidly treat A. marginale(anaplsmosis), on the other hand, same drug with double dose (6 milligram/kilogram body weight) and each dose has gape of two weeks,fail to cure A. marginalecarrier heifers(McHardy & Simpson. 1974). The animals recovered completely with imidocarb dipropionate (Akhter et al. 2010).The E elephantina and Aloemarlothiileaf extracts demonstrated good activity against rickettsia. These 2 plants along with Usanguineaand Rtridentata in certain concoctions are believed to be effective against anaplasmosis (Naidoo 2004). Dairy industry of Pakistan contributes up to 46.8% in the agriculture and about 10.8% of the GDP in the form of milk, milk products, meat, hides, skin and bone meal (Ghaffar et al. 2007). Molds are fungi consisting of filaments that are present in the feed of animals. Molds can cause many infections in dairy cattle, especially during period of stress when their immune system is depressed. They cause a special disorder that is termed as Mycosis. Specific mycotoxins are produced by these fungi and when these animals eat such type of feed then they also engulf such dangerous mycotoxins. Mycotoxins are produced by a wide range of various molds and can be classified as secondary metabolites meaning that their function is not essential to the mold’s existence (Whitlow and Hagler 2005). In the European countries, sheep milk is mainly produced in the Spain, and it is about 403000 tons per year. The main purpose of this milk is to produce cheese. Manchego cheese is considered as the best quality cheese in the world. This is the only technique by which we can produce the high quality of cheese (Rubio et al. 2009). These Aflatoxins are the fungal main dangerous constituents that can contaminate the feed of animals very easily. If AFB1 contaminated ration is consumed by animals then metabolism takes place in these toxins and excrete AFM1 in milk which further reduces milk production. More long-lasting aflatoxin poisoning produces very dissimilar signs that may not be clinically superficial and decreased rate of production in young animals. Severe aflatoxicosis causes hepatitis, hemorrhage, and death. Decreased growth rate is the most delicate clinical symbol of chronic aflatoxicosis, and it may be the only readily obvious abnormality. Natural toxins considerably show the greater danger to human and animal health. One large group of natural toxins that are universally Introduction 2 documented as poisons of food and feed are the mycotoxins. The toxins occur naturally in various animal feeds, including corn and cottonseed. To control AFM1 in milk, it is essential to decrease the feed contamination by AFB1 (Prandini et al. 2009). The livestock is playing major role in enhancing agricultural productivity and it has also major contribution to decrease the rate of poverty in the rural areas of Pakistan. Most farmers have domesticated sheep in their home which are their major sources of income (Mahmood et al. 2009). Aflatoxins adulterated rations prompted important reduction in daily feed consumption and the means of body weight, body weight increment and feed transformation rates were radically affected during the exposure stage to aflatoxins. Furthermore, serum ingredients and ruminal measurements showed lessened liver function and digestive turbulences in sheep fed aflatoxin (Jouany and Diaz 2005) Aflatoxins are included in that group of toxins which are considered as highly toxic toxins. Aspergillus parasiticus, Aspergillus flavus and Aspergillus nomius are those types of fungi which can produce very strong type of toxins. Aflatoxin B1 (AFB1) is considered as the most dangerous type of aflatoxin and it is very strong hepato-toxin (damaging the liver as a whole). It can also cause cancer, can produce teratogens, and may also cause mutation in the both animals and in human beings. AFB1 being very toxic, can be activated by the special metabolism of liver and through cytochrome P450. The cancer causing ability of AFBI is ten times more than the AFM1. The conversion of AFB1 to AFM1 in the milk producing animals is in the range of half percent to about six percent. In milk producing animals the level of aflatoxin M1 is greatly altered by the contamination rate of aflatoxin B1 (Bognanno et al. 2006). It is observed that the excretion pattern of aflatoxin M1 in the milk of sheep is lower than the excretion pattern of aflatoxin M1 in the milk of cows. Due to presence of aflatoxin B1 in the feed of animals and hence appearance of aflatoxin M1 in the milk of animals shows that Introduction 3 milk of such animals becomes more toxic and more un-hygienic. The young ones having milk as their main feed ingredient may have much more chances of getting various infections. (Var and Kabak 2009). To check the level of aflatoxin M1 in the milk of sheep, various experimental studies have been performed in the whole world (Battacone et al. 2005). In Pakistan, there are many favorable environmental conditions in which various types of fungi can easily prompted (Iqbal et al. 2011). The contamination level of aflatoxin M1 in the milk of dairy animals and in the dairy products has been found by (Hussain et al. 2008). Aflatoxins can be produced from fungal metabolites. Aflatoxin M1 is produced as the complete metabolism of aflatoxin B1 has taken place. Aflatoxin B1, when converted into aflatoxin M1 is appeared in the milk of that animals and the same aflatoxin M1 can also be appeared in the dairy products which can contaminate the feed of human too. Transformation of aflatoxin B1 into the aflatoxin M1 is around one to two percent. This transformation is highly dependent upon the feed of animals, transformation from one affected animal to other healthy animal, transformation from the milking of affected animal to the milking of healthy animal and from one day to another day also. It is also noted that as the level of aflatoxin B1 in the feed of animal is decreased down then the level of M1 in the milk of animals is also dropped down significantly but it takes about 72 hours for this dropped down (Nilchian and Rahimi 2012). AFM1 had a resistant to thermal inactivation used during food processing procedure such as pasteurization and autoclaving. Storage of various dairy products was not effective in the reduction of this toxin (Ozdemir 2007). Aflatoxin M1 level in the milk of animals can also be found in 12 to 24 hours after the ingestion of aflatoxin B1. After this, its level can be raised within the few days. The estimation of conversion rate of aflatoxin B1 in the contaminated feed of animals into aflatoxin M1 in the milk that animal is elaborated as 1 to 3 percent in a specific range (Ozdemir 2007). Introduction 4 Aflatoxin B 1 being very much dangerous mycotoxin, it is very much important to know about such methods by which the level of aflatoxin B1 in the feed of animals can be controlled. For this purpose we should first come to know the basic source of feed contamination of animals, and after this we should adapt such easy method to determine the level of aflatoxin B1 in the animal’s feed and such methods should be very cost effective. The best accepted and recognized method to determine aflatoxin M1 level in the milk of infected animals is HPLC high-performance liquid chromatography and TLC thin-layer chromatography (Thirumala-Devi et al. 2002). Aflatoxin M1 can be found in the milk of infected animals and can also be found in the products that are prepared from that contaminated milk. 21. Comparative Efficacy Of Various Immunopotentiating Agents In Subclinical Mastitis In Dairy Cows

Mastitis, the inflammation of the udder regardless of the cause, is the most costly disease of dairy cows. It affects the milk quality and production of cow along with changes in the composition of milk and the extent to which various compositional changes occur depends on the inflammatory response. There is also fluctuation in complete blood count (CBC). The study was conducted in dairy cow at different government and private dairy farms in and around Lahore Total n=30 cows were included in study trials and for treatment trials n=24 cows suffering from subclinical mastitis were taken and divided into 5 groups, 1st group was Vit E + Se 2nd group was Garlic, 3rd group was Black pepper and 4th group was Lemon having n=6 cows in each group randomly. A 5th group was formed comprising of n=6 healthy cows served as negative control. The drug trials showed that it Vit E+ Se and Garlic were the most effective drugs by boosting immune system by increasing the hematological parameters and improving the milk compositional contents and after that Lemon showed the good results while treating subclinical mastitis. The Black Pepper showed significant role by enhancing immunity nut didn’t cure as much of subclinical mastitis. Statistical Design: The data on chemotherapy, hemogram and milk composition and milk sodium chloride level was analyzed by one way analysis of variance (ANOVA) and paired T test using SPSS 20.0. 22. Development Of A Cost-Effective Serodiagnostic Assay For Peste Des Petits Ruminants (PPR)

Currently the poor small ruminant’s farmers in Pakistan are facing huge economic losses due to PPR virus. In Pakistan PPR causes economic losses of Rs. 20.5 billion annually. The objectives of present study were to develop a cost effective sero-diagnostic assay for PPR (active haemagglutination inhibition and passive haemagglutination inhibition) and determination of comparative efficacy of active and passive haemagglutination inhibition assay (HI and PHA respectively) for detection of PPR virus infection. In the present study, n= 300 sera samples were collected from sheep and goats during the (15 Februry 2016 to 2 January 2017).The serum samples were collected from kotli AJK :20(8 goats and 12 sheep),from gilgit:30 serum (20 goats and 10 sheep),from mansehra:22 serum (13 goats and 9 sheep),from mithi:112 (60 goats and 52 sheep) and 116 serum samples (88goats and 28 sheep) from Dhera ghazi khan.None of the animal was known to have been vaccinated against PPR previously or at the time of sampling. These samples were collected from animals showed symptoms of PPR suggestive of PPR disease as well as from healthy animals. The sera were transferred into sterile tubes and were preserved on ice packs while shifting to the laboratory. PPR virus isolate was originally isolated from an outbreak in Taxila village, district Rawalpindi, the isolate was attenuated serially onto the Vero cell lines up to 20 passages. After, which antigen was titrated using a micotiter haemagglutination (HA) test with chicken RBCs and stored at -70◦c until use as a PPR antigen in a HI test. In this study Active haemagglutination inhibition (HI) and passive haemagglutination inhibition were developed. The Haemagglutination Assay was standardized by different factors i.e. diluents, Temperature of incubation, Time of Incubation and concentration of Chicken R.B.C̓s. An additional test passive haemagglutination inhibition was performed to check the comparative efficacy of Active and Passive haemagglutination inhibition. In passive haemagglutination inhibition tanned sensitized cells remains effective due to their long effective life when stored at 4̊c and its makes an ideal test for diagnosis of PPR. Newly developed assays were compared against cELISA for PPR using kappa statistics and diagnostic sensitivity and specificity were determined. The results of both assays were compared with results of competitive enzyme linked immunosorbent assay. In this study cELISA was considered as golden standard. The relative sensitivity and specificity of Active haemagglutination inhibition is 94.9% and 97.9% respectively. (Kappa 0.9264). However the sensitivity and specificity of Passive haemagglutination inhibition is 91.1% and 95.0% respectively.(kappa 0.8595). This study describes the serological detection of PPR virus by Active haemagglutination and passive haemagglutination inhibition (HI and PHA respectively). It was also concluded the comparative efficacy of (PHA and HI) that Active haemagglutination inhibition is more reliable technique than passive haemagglutination inhibition assay for the diagnosis of PPR disease in small ruminants (sheep and goats).
