2. Antimicrobial Efficacy Of Different Drugs Against Experimentally Induced Salmonella Pullorum Infection In Broilers

Material type: book Book; Format: print ; Literary form: not fiction Publisher: 1996Dissertation note: A total of 120 broiler chicks were randomly selected and divided into six groups viz A, B, C, D, E and F, consisting of 20 birds each. At 4 week of age, A to E groups were experimentally infected with Salmonella pullorum inoculum. Group E (infected, non-medicated) and F (non-infected and non-medicated) were kept as control. The groups A, B, C and D were treated with Ampicillin 20%, Oxy-N-50, Chioricol10 and Trimodin forte, respectively, post infection. All the groups were kept under close observation to record signs and symptoms of disease, mortality, body weight gain and Feed Conversion Ratio. Gross lesions in visceral organs were also noted during this study. Some blood parameters (TEC, TLC and Hb content) were also estimated, pre and post infection. A mortality percentage in groups A, B, C and D were recorded as, 25%, 35%, 15% and 10%, respectively, while 70% mortality was recorded in control group F. According to this trial Trimodin forte (Trimethoprim Plus Sulphadiazine) . afforded maximum protection against infection and proved best in relation to weight gain and F.C.R. Chloricol-10 (Chioramphenicol) stood second in the list, while Ampicillin-20% (Ampicillin Trihydrate) was the third drug in furnishing protection against the infection. Whereas, Oxy-N-50 (Oxytetracycline plus neomycin sulphate) was the least effective drug in respect of protection against pullorum disease. The signs of ill health appeared 3 to 4 days post infection were listlessness, ruffled feathers, droopy wings, loss of appetite, poor growth, depression, increased thirst and severe diarrhoea of chalky white colour. The postmortem findings were enlarged and congested liver streaked with haemorrhages, 2-3 times enlarged spleen (spleenomegaly), enlarged heart and pericarditis, congested and distended kidneys, area of gray hepatization in lungs, thickened and inflammed intestinal walls and presence of necrotic foci in cardiac muscles, liver, spleen, lungs, caeca, large intestine and muscles of gizzard. The Total Erythrocytic count and Haemoglobin contents were decreased and Total Leukocytic Count, were increased 3 days post infection. The findings of the present study will further elucidate the disease and help in diagnosis and treatment of this malady on large scale. 3. Effects Of Supplementation Of Selenium And Zing Alone And In Combination On The Immune Response Of Broiler Chicks

Material type: book Book; Format: print Publisher: 2002Dissertation note: The present project was designed to investigate whether different levels of selenium and zinc alone and in combination in diet had any effect on the immune response of broiler chickens. For this purpose two hundred and twenty five day-old broiler chicks were taken and equally divided into three different trials i.e. A, B and C. Trial A was supplemented with selenium (Se) at four different rates 0.18, 0.5, 1.0 and 0.0 mg/kg in groups Al, A2, A3 and A4, respectively. Trial B was supplemented with zinc (Zn) at four different rates 40, 80, 120 an zero mg/kg in groups Bi, B2, B3 and B4, respectively. Trial C was supplemented with Se + Zn at rates of 0.18±40, 0.5±80, l.0±120 and 0.0±0 in groups Cl, C2, C3 and C4, respectively. Immunomodulatory effects o Se and Zn were evaluated by conducting hurnoral immune response against Newcastle disease vaccine (ND vaccine), delayed hypersensitivity reaction, weights of immune organs, growth traits and economy. Highest humoral immune response was observed in groups A3 (Se 1.0 mg/kg), B3 (Zn = 120 mg/kg), C3 (Se = 1.0 mg/kg + Zn = 120 mg/kg). rp1.e groups A4, B4 and 04 which were kept without Se and Zn supplementation had the lowest Immoral immune response. Delayed hypersensitivity index was highest in groups A3, B3 and C3, while lowest in control groups A4, B4 and C4. There was positive effect of Se and Zn on the histopathology of bursa of Fabricius, thyrnus and spleen. Body weight gains were also highest in A3, B3 and C3 and FCR was best in groups A3, B3 and C3. Weights of bursa of Fabricius, thymus and spleen were also highest in groups A3, B3 and C3, while lowest in the control groups A4, B4 and C4. On the whole immunomodulatory effects of supplementation of Se and Zn ir combination were better than individual supplementation of Se and Zn. 4. Effect Of Various Extenders On Semen Characteristics Of Beetal Buck (Capra Hircus)

Material type: book Book; Format: print Publisher: 2003Dissertation note: The artificial use of germ cells of genetcally superior bucks can enhance the growth potential of goat population to meet the shortage of mutton meat in Pakistan for daily slaughtering, sacrificial events, skin, hair and goat milk too. Beneficial use of superior germ cells can be made only when there is extension of life of germ cells for long periods, maintenance of motility of spermatozoa and increase in number of doses of ejaculate, for which an extender of choice has to be developed. Semen characteristics of forty ejaculates of bucks were evaluated. Pooled samples of ejaculates having motility estimates of at least 60% were used for evaluation. After washing of seminal plasma with physiological normal saline (20% ringer solution) and centrifugation at l000xg for 10 minutes to remove the sperm toxic factor Lecithinase-A. Pooled semen samples were extended in Tris yolk fructose citric acid (TYFCA), milk yolk (MY) and egg yolk citrate (EYC) extenders. Samples were extended using one step extension at a ratio of 1:60 in such a way that each milliliter of semen contained 30x106 progressively motile spermatozoa. Finally extended semen samples were placed at 5°C and 37°C for evaluation of motility percentage after every 24 hours interval and livability (hours) and absolute index of livability o spermatozoa, respectively. Mean±S.E. values of ejaculates of bucks for volume, pH, mass motility, individual motility percentage, sperm cell concentration, live and dead percentage, sperm abnormalities was recorded and post-extension motility percentage at 5oC, livability (hours) and absolute index of livability of of' spermatozoa of pooled semen at 37°C was recorded. Significant differences were observed (P<0.01) in post extension motility percentage at 5°C, at all intervals except at 120 hours interval where deference vas non-significant (P>0.05) between milk yolk and egg yolk citrate extenders. Post extension motility percentage at 5°C was highest in TYFCA than MY and EYC e\tenders. Post extension livability (hours) at 37°C was significantly different among three extenders at a level of probability (P<0.05) but non-significantly different among three extenders at a level of' probability (P>0.01). Absolute index of' livability at 37°C shows significant differences (P<0.01) for all extenders under statistical analysis. Livability (hours) and absolute index of' livability was higher in TYFCA than MY and EYC extenders. Based on these results and effect of extenders on semen characteristics of Beetal Buck, Tris- Yolk-Fructose-Citric Acid was developed as an extender of choice for short term preservation of' semen of' Beetal Buck.
