1. Clinico-Epidemiological And Therapeutical Studies On Coccidiosis ( Isospora ) In Dogs In And Around Lahore;

Material type: book Book; Format: print Publisher: 2012Dissertation note: The present project was designed to find out the prevalence of coccidiosis (Isosporiosis), to evaluate the comparative efficacy of Sulfadimethoxine and Furazolidone and to study the effect of Coccidiosis on blood parameters. For this purpose, the faecal samples of three hundred dogs were examined in the Medicine laboratory, University of Veterinary and Animal Sciences, Lahore by direct smear method and concentration technique. Out of three hundred dogs, forty nine were found positive for coccidiosis, showing the prevalence of 16.33%. For the chemotherapeutical trials, forty five naturally infected and fifteen non-infected were selected and divided randomly into four groups having fifteen dogs in each group wise A, B, C and D. Group A was treated with Sulfadimethoxine at the dose rate of 60 mg/kg (orally). Group B was treated with Furazolidone at the dose rate of 20 mg/kg (orally). Group C was kept as infected non-medicated control. Group D was kept as non-medicated and non-infected control. The faecal samples of experimental dogs were examined for OPG (oocyst per gram) counts on zero day (pre-medication), 3rd day, th day, and 14th day medication and percentage reduction of oocysts was' calculated. Sulfadimethoxine showed 66.69%,90.89% and 98.29% efficacy and Furazolidone showed 63.95%, 88.78% and 96.64% efficacy on 3rd, th and 14th day, respectively. Haematological study showed that the values of haemoglobin, packed cell volume and total erythrocyte count were lower in groups A, Band C as compared to non-infected control group. The values of total leucocyte count were higher in group A, B and C' as compared to non-infected group. As a result of treatment in group A and B, the values of haemoglobin, packed cell volume and total erythrocyte gradually increased while that of total leucocyte count gradually decrease on day 3rd ,7111 and 14th post-medication. It has been observed during the present study that sulfadimethoxine and Furazolidone showed their effect on blood parameters as a result of coccidiosis. 2. Molecular Diagnosis And Therapeutic Trials On Fasciolosis In Sheep And Goats

Material type: book Book; Format: print Publisher: 2012Dissertation note: The fasciolosis in sheep and goat is an important and infectious disease of ruminants characterized by dullness, weakness, lack of appetite, pallor and edema of mucosa and death of animal. The accurate diagnosis of Fasciolosis remained a difficult task for the field practitioners and the right choice of medicine. For this purpose 600 sheep and goats was examined for fasciolosis. The fecal samples were collected directly from rectum while bile samples were collected from various private and public slaughter houses of districts Okara, Sahiwal and Lahore. Fasciola egg was identified by standard text while molecular conformation of Fasciola spp. was done by using PCR. For therapeutic trials, sixty animals (n=30 sheep; n=30 goats) positive for Fasciolosis was equally divided into A, B and C groups and each group comprising of 20 animals (n=10 sheep; n=10 goats). The animals of group A was given Zanil (Oxyclozanide) orally @ 1ml per 3kg body weight; the animals of group B was given Albendazole orally @ 1ml per 3kg body weight and the animals of group C was given the grounded powder of Neem leaves (Azadirachta indica) @ 5-6 teaspoon (25-30gm) orally. Efficacy of drugs was measured on the basis of disappearance of Fasciola eggs at 3, 7 and 14 day of post-medication. Data on the prevalence of fasciolosis was analyzed by Pearson's chi-square test while therapeutic trials data was analyzed with one way ANOVA using Statistical Package for Social Sciences (SPSS); P < 0.05 was considered significant. To study fasciolosis in sheep and goats, samples were collected from Lahore, Okara and Sahiwal. The collected samples were processed at the Medicine laboratory at UVAS, Lahore and LPRI laboratories of Bahadurnagar, Okara. In this study first of all the fecal and bile samples were examined through microscope then these samples were processed by PCR test. No study was conducted previously on the molecular diagnosis of fasciolosis in sheep and goats in Pakistan, this is the first report for the molecular diagnosis of fasciolosis. In all F. hepatica positive samples approximate 300-bp fragment was generated on gel tested with DSJF/DSJ3 primers for sheep and goats. All positive samples by microscope were also positive by PCR, but out of the 92 samples positive (43 in sheep; 49 in goats) by PCR test, out of these 14 (7 in sheep; 7 in goats) were negative by microscopy. This study provides the base line data for the molecular diagnosis of fasciolosis in sheep and goats in Pakistan. Overall prevalence of fasciolosis in sheep in both fecal and bile samples in various districts of Punjab was calculated. Out of 300 samples (150 fecal; 150 bile) of sheep, F. hepatica prevalence rate was 12% (36 samples) by microscopic examination while 14.33% (43 samples) by PCR test. On the basis of microscopic examinations districts wise prevalence rate was 11, 12 and 13% in Lahore, Okara and Sahiwal respectively and by PCR test prevalence rate was 13, 14 and 16% in Lahore, Okara and Sahiwal respectively. Prevalence of F. hepatica was more in Sahiwal than Okara and Lahore in sheep in both fecal and bile samples on the basis of microscopic examination and PCR. Overall prevalence of fasciolosis was also calculated in goats on the basis of microscopic examination and PCR in both fecal and bile samples. Among 300 samples (150 fecal; 150 bile) of goats F. hepatica prevalence was 14% (42 samples) by microscopic examination while 16.33% (49 samples) by PCR. On the basis of microscopic examinations the prevalence was 13, 15 and 14% in Lahore, Okara and Sahiwal districts respectively while on basis of PCR test prevalence was 15, 18 and 16% in Lahore, Okara and Sahiwal districts respectively. Prevalence of F. hepatica was more in Okara than Sahiwal and Lahore in goats in both fecal and bile samples on the basis of microscopic examination and PCR. The eggs per gram (EPG) values and efficacy of Oxyclozanide (Zanil), Albendazole and Neem leaves against fasciolosis in 30 positives sheep was recorded at 3, 7 and 14 days after the treatment. As group A was treated with oxyclozanide (Zanil), its efficacy was 50, 75 and 90% at 3rd, 7th and 14th day respectively after treatment. The efficacy of albendazole was 46, 65 and 81% at 3rd, 7th and 14th day respectively while the efficacy of Neem leaves was lowest that was 15, 30 and 42% at day 3rd, 7th and 14th day respectively. The efficacy of oxyclozanide and albendazole was significantly higher (p= 0.000) than Neem leaves. In sheep against the fasciolosis Oxyclozanide significant effect on EPG followed by albendazole and Neem leaves. The EPG and efficacy of Oxyclozanide (Zanil), Albendazole and Neem leaves were determined in 30 goats against fasciolosis at 3, 7 and 14th day of post treatment. The efficacy of oxyclozanide was 57, 76 and 90% at 3, 7 and 14th day of post treatment, respectively. The efficacy of oxyclozanide was higher than Albendazole against the fasciolosis. The efficacy of albendazole was 55, 68 and 79% at day 3, 7 and 14th, respectively. Efficacy of Neem leaves against fasciolosis was lower as compared to other two drugs, it was 15, 23 and 40% at day 3, 7 and 14th, respectively. When the efficacy was compared at different days, Oxyclozanide reduced the EPG significantly, followed by Albendazole and Neem leaves. From this study it was concluded that PCR is more reliable technique than microscopic examination for the diagnosis of fasciolosis in sheep and goats. Microscopic technique provides the base line data for the diagnosis of fasciolosis in sheep and goats but PCR provides help to conduct further molecular diagnosis on fasciolosis as well as others parasitic diseases. Secondly fasciolosis which is an important parasitic problem of ruminants is significantly prevalent in sheep and goats in Lahore, Okara and Sahiwal districts of Punjab. Oxyclozanide is the most effective drug while albendazole and neem leaves (Azadirachta indica) are relatively less effective against fasciolosis in sheep and goats. It is due to the insufficient production of insulin by the pancreas. The multi-factors such as obesity, feeding, exposure to toxic drugs etc. are involved for the onset of this chronic disease. The basic energy fuel of the body is glucose. In the absence of insulin the glucose level in the blood is increases which can be noticed as hyperglycemia (high blood glucose level). There are the three types of diabetes mellitus which includes insulin-dependent diabetes mellitus (type I or IDDM), noninsulin-dependent diabetes mellitus (type II or NIDDM) and secondary diabetes mellitus. The dogs play a vital role in our society. The present study was conducted on diabetes mellitus to study its prevalence, its effect on various blood parameters and to evaluate the efficacy of insulin therapy in pet dogs in Lahore, Pakistan. The 500 blood samples were collected from the dogs of age ?5 years, presented at Pet centre of the University of Veterinary and Animal Sciences (UVAS) Lahore and various private clinics in Lahore, and was processed in Medicine Laboratory of UVAS, Lahore. The blood glucose level was measured with the aid of glucometer. Out of 500, 16 (n=5 male & n=11 females) were positive for diabetes mellitus. The 3.2% prevalence of diabetes mellitus was recorded in pet dogs in Lahore. The study concluded that the prevalence of diabetes mellitus in female dogs (68.75%) was significantly (p<0.05) higher than that of male dogs (31.25%). The results of this study showed that the prevalence of diabetes mellitus was higher in old age dogs (>9 years) as compared with other age groups. Diabetes mellitus was found to be more prevalent in winter season same as in human beings. The clinical signs like Polyuria (81.25%), Polydipsia (68.75%), Hyperglycemia (100%), Dehydration (43.75%), Anemia (56.25%) and cataract (18.75%) were commonly observed in diabetic dogs. In the present study the pet dog breed like Yorkshire terrier, Samoyed, Pomuranian and Cross bred were prone to have diabetes mellitus whereas German shepherd, Bull dogs, Pugs and Labrador retriever were at low risk for this malady. In the present project, the haematological study was conducted on 28 dogs (n=14 diabetic and n=14 healthy dogs). The five ml blood sample were collected from each dog directly from the cephalic vein and were shifted into a sterilized plastic bottle coated with EDTA@1mg/ml and were further analyzed through haemtological analyzer. The results showed the decreased number of RBCs (4.63±0.42) and an increased in PCV values (62.91±4.18). The results also showed dehydration and anemia in diabetic dogs. In the present study the 21 dogs (n= 14 diabetic dogs, n= 7 healthy dogs) were selected for therapeutic trials. The dogs were divided into further three groups; A, B and C. Each study group was comprised of 7 animals. Group A (comprised of diabetic dogs) was treated with Humulin-N (Insulin) @ 0.5 U/Kg, q12h, SC for 7 days. The group B (comprised of diabetic dogs) was kept as positive control while the healthy dogs were kept in group C for negative control. After the seven days treatment of group A, the blood samples were collected every 2 hr interval for 12 hrs for measuring the glucose value. The process was repeated once in a week for one month. The four blood glucose curves were made. Each blood glucose curve has its own nadir value. The time of glucose nadir value was indicated the peak action of insulin. The blood glucose curves were made to evaluate the effectiveness of insulin. The therapeutic results of this study concluded that the Humulin-N (insulin) was much effective for management of diabetic dogs. Considering the significance and utilization of equines in our country and the substantial losses rendered by theileriosis, the present study was designed to study prevalence and chemotherapy of theileriosis in equines in and around Lahore. For this purpose 300 equines (Horses=100; mules=100; donkeys=100) were examined for theileria infection.The blood was collected by ear tip puncture and from Jugular venipuncture. Then thin blood smear slide was prepared and dried up in air and stained with Geimsa's staining method and examined under microscope. Theileriaequiwere identified by their morphological characteristics as described by Rashid et al. (2009) and Guimaraeset al. (1997).Over all prevalence of Theileriaequiin equines was estimated as 58%. There was also a mixed infection of Theileriaequi and Babesiacaballiin some cases. Individually there was 54%, 64% and 56% prevalence of Theileriaequi found in horses, mules and donkeys respectively. The effect of theileriosis on various blood parameters (Hb, ESR, TEC, TLC, DLC and PCV) was determined in 30 equines (Horses= 10; mules= 10; donkeys= 10). The remarkable decrease in hemoglobin, TEC, PCV and lymphocytes was observed. The value of ESR increased while there were no remarkable changes in TLC, monocytes, basophils, neutrophils and eosinophil. For chemotherapy, 36 equines (Horses=12; mules=12; donkeys=12) positive for theileriosiswere divided into three groups (A, B, and C). Each group was further subdivided into three subgroups comprising of 4 animals in each subgroup. The animals of group A (Horses=4; mules=4; donkeys=4) were treated with Imidocarbdipropionate (Imizol®, ICI, Pakistan) @ 2 mg/kg BW I/M, and the efficacy of drug was found 91.7% in equines against theileriosis.The group B (Horses=4; mules=4; donkeys=4) was treated with Buparvaquone (Butalex®, ICI, Pakistan) @ 2 mg/kg BW I/M alone and was observed 66.7% effective. While the group C (Horses=4; mules=4; donkeys=4) was treated with Auk(Calotropisprocera)@ 0.3mg/kg BW, 8 doses at alternate days (local treatment), which was found 58.3% effective. The efficacy of drugs was measured on the basis of disappearance of clinical signs and blood smear examination at day 2, 4, 6 and 10 of post-medication. The efficacy of each drug was measured through recovery rate of the animals. Finally, the data on prevalence of theileriosis was estimated by Pearson's chi-square test for significance whereas data on hematology was analyzed by Student's T-test using statistical software package STATA 9.1 (College Station T×77845, USA), P < 0.05 was considered significant. Considering the significance and utilization of equine in our country and the substantial losses rendered by theileriosis, the present project was designed to check prevalence and chemotherapy of theileriosis in equines in and around Lahore. The disease is worldwide in distribution and is caused by many species of Eimeria which invade cells of intestinal epithelium. To treat and control coccidiosis in sheep and goats, numerous drugs were found to be effective including Amprolium, Toltrazuril, Decoquinate, Diclazuril and Sulphadimidine. The present study was conducted to determine the prevalence of coccidiosis in Dera Ghazi Khan, for this 752 fecal samples (376 from sheep and 376 from goats) were collected and examined through direct smear method and salt flotation method. Overall prevalence of coccidialinfection was 48.67%, with prevalence of 52.92% for sheep and 44.41% for goats. The prevalence of coccidiosis in sheep was significantly higher (P<0.05) compared to goats. Eleven Eimeria species were identified 6 from sheepand 5 from goats. In sheep the common Eimeria species wereE. ovinoidalis(56.78%), E. ahsata(47.23%), E. parva(35.67%), E. intricate (30.15%), E. faurei(26.63%) andE. pallid (19.09%). In goats the prevalent species wereE. ninakohlyakimovae(68.86%), E. alijevi(59.88%), E. arloingi(53.29%), E. caprina(46.70%) andE. hirci(22.15%). Eimeria infection was significantly high (P<0.05) in female (54.57%; 310/568) compared to male (30.43%; 56/184).The prevalence of coccidiosis was significantly higher (P<0.05) in ?6 month as compared to above 1 year age.A significant difference was found in three feeding systems (i-e) in stall feeding, grazing and both (grazing + stall) feeding system.Coccidiosis was significantly high (P<0.05) in animals with poor and week body condition compared to medium, good and fat animals. A strong association (P<0.05) was observed between fecal score and prevalence of Eimeria infection. Peak prevalence was observed in month of August (68.08% and 60.63%) while low in October (38.29% and 30.85%) in sheep and goats, respectively. In present study threeaniticoccidial drugs were used and the percent efficacy of Furazolidon, Sulphadimidine and Amproliun was 96.34, 95.40 and 98.08 percent, respectively at day 7. Whereas 98.58, 98.03 and 99.55 percent,respectively at day 21.It was concluded from the results that there was no significant difference (P>0.05) among the percentage efficacy of Furazolidone, Sulphadimidine and Amproliun used in these trials. Hematological analysis was performed using Automated Hematological Analyzer. Hematological analysis showed a significant reduction (P<0.05) in total erythrocyte count (TEC) and hemoglobin (HB) content due tococcidiosis. Results show a significantincrease (P<0.05) in TLC, MCV and PCV of infected animals as compared to control negative. Mean lymphocytes, Monocytes, MCHC and MCH counts were also decreased in infected animals. Hematological values of treated animals become normal at day 21stafter treatment. Considering the significance and utilization of equines in our country and the substantial losses rendered by trypanosomiasis, the present study was designed to study prevalence and chemotherapy of trypanosomiasis in equines of Gujranwala and Sialkot districts. For this purpose, 300 equines (horses=100; mules=100; donkeys=100) were examined for Trypanosoma infection. The blood was collected by ear-tip puncture and from Jugular venipuncture. T. evansi was identified by their morphological characteristics as described by Smyth, (1996). Overall prevalence of T. evansi in equines was estimated as 13%. For chemotherapy, 36 equines (horses=12; mules=12; donkeys=12) positive for trypanosomiasis, were divided into three groups (A, B, and C). Each group was further subdivided into three subgroups, comprising of 4 animals in each subgroup. The animals of group A (horses=4; mules=4; donkeys=4) were treated with diaminazine aceturate (Fatrybanil®, Fatro, Pakistan) @ 3.5 mg/kg BW I/M, and the efficacy of drug was found 83.3% in equines against trypanosomiasis. The group B (horses=4; mules=4; donkeys=4) was treated with isometamedium chloride (Trypamidium Sumarin®, Merial, Pakistan) @ 0.25 mg/kg BW I/M alone, and was observed 91.6% effective. While the group C (horses=4; mules=4; donkeys=4) was treated with Auk (Calotropis procera) @ 0.3mg/kg BW, 8 doses at alternate days (local treatment), which was found 41.6% effective. The efficacy of drugs was measured on the basis of disappearance of clinical signs and recovery rate of the animals, and blood smear examination at day 2, 4, 6 and 10 of post-medication. Finally, the data on prevalence of trypanosomiasis was estimated by Pearson's chi-square test for significance whereas data on haematology was analyzed by Student's T-test using statistical software package STATA 9.1 (College Station T×77845, USA), P < 0.05 was considered significant. Sheeps and goats are playing a leading role in the national economy by producing wool, skin, meat, milk and farm yard manure. Fasciolosis, parasitic gastrioenteritis, hydatidosis, coccidiosis, ectoparasitism, theleriosis and babesiosis are the major parasitic problems of small ruminants .There was an urgent need to carry out research on applied aspect leading control of parasites. Study Objectives: The main objectives of the present study were; (i) To have an overview on the overall prevalence of ectoparasites and endoparasites in small ruminants throughout the district by means of fecal tests. (ii) To observe the affect of ecto and endoparasites on blood profile of the infected individuals. Hypothesis: It was hypothesized that ¢ Parasitic diseases are highly prevalent in sheep and goat in District Bannu.. ¢ External and internal parasites have significant effect on blood parameters. Methodology and Results During this study period 500 sheep and 500 goats were examined for endo and ectoparasites while using various fecal tests such as using direct microscopic examination, simple test tube flotation method and sedimentation method for endoparasites. Out of 500 goats and 500 sheep, 47 goats and 63 sheep were found positive for ectoparasites and percentage ratio was 9.4% in goats and 12.6% in sheep. Mostly, three types of ectoparasites were observed in both of cases i.e. goats and sheep (ticks, lice and mites). The total no. of positive cases found for ectoparasites in goats were 244 (ticks 10.2%, lice 30.4% and mites 8.2% in district Bannu). This result indicates a significant load of ectoparasites in this area of the country. Out of 149 positive cases of ecto-parasites in sheep; tick infection was 7.8% (39/500), lice 15.2% (76/500) and mite infection was 6.8% (34/500). Fecal samples of all 1000 animals were analyzed for the presence of endoparasites, out of which 90.6% goats and 87.4% sheep were found positive for endoparasites. Overall goats were more likely to be positive for Haemonchus Spp. and almost 50.2% (251/500) animals were found positive. The prevalence of other endo-parasites like Trichurus 5.6% (28/500), Strongyloid 4% (20/500), Coccidia 8.6% (43/500), Fasciola 12% (60/500) and Trichostrongyloide were 10.2% (51/500) while in case of sheep Haemonchus Spp. positivity was 33.2% (166/500). The prevalence of other Endo-parasites like Trichurus 6.8% (34/500), Strongyloid 2.4% (12/500), Coccidia 6.8% (34/500), Fasciola 10.6% (53/500) and Trichostrongyloide were 13.6% (68/500). The hematological profile of infected animals indicates high TLC and low hemoglobin level. Conclusion It is concluded that high prevalence was due to poor management and lack of knowledge about feeding, vaccination, hygienic conditions and lack of proper deworming programme. To overcome the problem it was necessary to educate the farmers about deworming programme and proper use of insecticides. In Pakistan the high prevalence of 49% is in the rainy season (Farooqet al. 2007) Clinically the signs of Hemorrhagic septicemia includes profuse salivation,pyrexia, respiratory distress, swelling in throat, discharge through nostrils, protrusion of tongue, edema in the brisket area as well as in forelegs. Hemorrhagic septicemia can be diagnosed on the bases of clinical signs and laboratory conformation by Gram staining and. Serum agglutination test, Counter immune-electrophoresis and ELISA techniques are also used.Enrofloxacine was found to be effective treatment in case Hemorrhagic septicemia. Best protection measure vaccination by alum precipitated vaccine (Boudewijn et al. 2008). During the present study blood samples were collected from the calves at different intervals of times. Serum was separated from that blood and was analyzed by using Indirect Haemagglutination(IHA) Test to observe the antibodies titer in the blood.The data was analyzed to calculate geometric mean titer (GMT) of the antibodies. Immunity status is much important in the defense of disease especially in the newly born claves which are more at risk of infections.The result of present study showed geometric mean titer 16 in colostrum of vaccinated dams and their newly born claves showed 0, 3. 4, 7 and 3 at 0 hour, 72 hour, 7, 15 and 30 days of their age. Colostrum of non-vaccinated dams showed GMT 8 and their newly born claves showed 0, 1. 3, 3 and 1 at 0 hour, 72 hour, 7, 15 and 30 days of their age.The maternal antibodies which are produce in the body of mother are transferred to their calves.The calves given birth by the vaccinated dams receive much higher level of antibodies from their mother than the calves born from non-vaccinated dams. This antibodies production in non-vaccinated dams is because of the carrier status of hemorrhagic septicemia.The non-vaccinated dams which are not expose to the any type of HS infection or not expose to the vaccines, they do not produce antibodies against Pasturellamultocidaand also not transfer to their young babies after birth. The present study showed that claves of vaccinated dams are much protected in their early period of life against hemorrhagic septicemia. Buffalo calves need quick and intense care at early age and they need quick vaccinations. The high maternal antibodies level against Pasturellamultocidain vaccinated dams then in non-vaccinated dams antibodies levels in newly born buffalo calves remain effective for longer period. This study suggested for the vaccination of dams not only for the protection of themselves but also for the protection of their newly born claves at the crucial period of their young age. Liver flukes and paramphistomes (Fasciola gigantica and Paramphistomum microbothrium) occasionally cause heavy mortality in animals grazing swampy areas (Akerejola et al. 1979). The adverse effects of nematode infections include: loss of weight, anorexia, anaemia, retarded growth, delayed sexual maturity, decrease in milk and meat production (Saddiqi et al. 2012). Indigenous knowledge of herbal medicine is a big source of the modern knowledge. Today, thousands of plants, traditionally used as medicines are being explored (Kakar, 2012). Chemical control of helminths coupled with improved management has been an important worm control approach throughout the World. However, increasing problems of development of resistance in helminths rise in price of drugs contributing factor for traditional plant used (Coles et al. 1997) against anthelmintics have led to the proposal of screening medicinal 1 Introduction plants for their anthelmintic activities. Reports of drug resistance have been made in every livestock host and to every anthelmintic class. In some regions of world, the extremely high prevalence of multi-drug resistance (MDR) in nematodes of sheep and goats threatens the viability of small-ruminant industries. Many parasitic nematodes of veterinary importance have genetic features that favor the development of anthelmintic resistance (Kaplan et al. 2004). The plants are known to provide a rich source of botanical anthelmintics (Lewis and Elvin Lewis 1977). There are many medicinal plants have been used to treat parasitic infections in man and animals (Iqbal et al. 2005). Various botanical plants have been possessed anthelmintic activity against helminth e.g Chenopodium album (Eguale & Giday, 2009). It has been expected that there are around 250,000 plant species present throughout the world. Plants, from ancient, have served human beings as sources of food, shelter, clothing and medicines. Before the advent of modern allopathic medicine and synthetic drugs, plants and to a certain extent, animals and minerals were used in various formulations for treatment of diseases by traditional medicinal practitioner (Rahmatullah et al. 2011). Plant medicine is very important from ancient to present daytime. The uses of biologically different plant assets for various ailments are the lifelong struggle of humankind (Hussain et al. 2008). In Indo-Pak subcontinent, Ayurvedic and Unani therapeutic systems are very popular and people have been using plants not only for the treatment of their own ailments but also for their domesticated animals. Chenopodium album (Chenopodiaceae) commonly known as “Bathu” is important medicinal plants in Pakistan and their different parts are utilized in the traditional system of medicine (Said et al. 1970). Chenopodium album usually recognized as “Bathua” is a familiar food as well as a medicinal plant. The seeds of the plant are known to possess anthelmintic activity against Haemonchus contortus. A compound C37- trihydroxy adjacent bistetrahydrofuran acetogenin, present in the seeds, is responsible for inhibition of the egg hatching of gastrointestinal nematodes (Saha et al. 2011). Chenopodium album (Chenopodiaceae) is a rapid rising fragile annual plant and it is found in Bangladesh. In English plant known as Lamb’s quarters and in Bengali as Buthiya shak. Both in vitro and in vivo activity of plant as anthelmintic has been reported (Jain and Singhai 2012). Chenopodium album was found to be one of the initial plant species colonizing a heavy metal-contaminated site, polluted by pyritic (sulphide-rich) waste from the Aznalcóllar Mine spill (South-western Spain). This shows its importance in the re-vegetation of this soil (Walker et al. 2004). In vitro experiments were arranged to conclude the possible anthelmintic efficacy of crude aqueous extracts and powder of the whole of Chenopodium album (Eguale et al. 2009). Coccidiosis with the protozoan parasite Eimeria as the infectious agent causes enormous economic losses, particularly in poultry farms (Dkhil et al. 2011). Eimeria are common parasites in the digestive tract of the hosts causing diarrhea and fluid loss. Infections begin with oral uptake of Eimerian oocysts which release sporozoites in the intestine. These in turn invade enterocytes in which they multiply, and finally, oocysts are released again with the faeces (Renaux et al. 2001). Medicinal plants as natural feed additives are recently used in poultry diet to enhance the performance and immune response of chicken (Tanweer et al. 2012). Coccidiosis of chickens is an enteric parasitic disease caused by multiple species of the protozoan parasite genus Eimeria (Apicomplexa: Eucoccidia: Eimeriidae) and is one of the commonest and economically most important diseases of poultry world-wide (Shirley et al. 2005). It causes production losses, and high morbidity (due to acute, bloody enteritis) and mortality rates. While the control of coccidiosis has relied mainly on the preventive use of anticoccidial drugs together with the induction of species-specific natural immunity in chicken flocks (Shirley et al. 2005), this widely used approach is costly and has led to serious problems with drug resistance in Eimeria populations (Williams, 2006). Levine discovered that sulphanilamide would cure coccidiosis in chickens, various anticoccidial feed additives have been developed and added (Levine 1939). Efforts have always been made to add coccidiostats on regular basis in chicken’s diet. Consistent use of coccidiostats has not only narrowed the gap between cost of production and returns, it could be a potential threat to human being as a result Introduction 2 of their residual effects. The increasing resistance of avian coccidiosis to anti-coccidial drugs currently used by poultry industry together with the requirement for drug and antibiotic free production systems has meant that it is now important to look for new ways to control the disease. So, research initiated for possible use of herbal (natural) products against avian coccidiosis. Herbal products in past have been effectively used for the control and treatment of several ailments in poultry and human beings. Allium sativum a medicinal plant could be an effective substitute for coccidiostats because of its chemical nature and antimicrobial activities. Allium sativum (garlic) have been widely used for their broad range of pharmacological activities, including antiparasitic activities. Avian coccidiosis is one of the most costly and widespread parasitic diseases in the poultry industry, and has been mainly controlled by the use of different chemotherapeutic agents. Due to the emergence of drug-resistant strains, alternative control strategies are needed (Elbanna, 2012). Hence, further investigations are required to understand the exact mechanism underlying the effects of these feed additives, which are still in use in broiler nutrition as performance enhancers and as agents for controlling coccidiosis. Garlic (Allium sativum ) has been known as an herbal remedy to prevent and treat a variety of heart diseases and metabolic diseases, such as atherosclerosis, thrombosis, hypertension, dementia, cancer, and diabetes (Elbanna, 2012). Garlic has rich organosulfur compounds and precursors (allicin, diallyl sulfide, and diallyl trisulfide). The enzyme allinase that is responsible for converting alliin (S-allyl cysteine sulfoxide) to allicin is inactive. When garlic is chopped or crushed, the allinase enzyme present in garlic is activated and acts on alliin (present in whole garlic) to produce allicin (Tanweer et al. 2012). Many studies indicate that allicin is the potentially active component of garlic. These compounds provide garlic its characteristic odor flavor as well as most of its biological properties and have been identified as having the Introduction 3 hypocholesterolemic effect in human and animal products (Silagy and Neil, 1994; Konjufca et al., 1997; Chowdhury et al., 2002). Species of Eimeria in poultry are usually identified by the size and morphology of different stages of the life cycle of the parasite and the nature and location of lesions in the intestines. Differentiation of species in mixed infections, however, can be difficult because some species have similar characteristics (Long and Reid, 1982). Different workers have investigated the prevalence of eimeriosis in different classes of poultry birds and in different areas of Pakistan (Dar and Anwar, 1981; Anjum, 1990; Ayaz et al., 2003). Each Eimeria species has a particular predilection site in the chicken digestive tract; for example, E. tenella attacks the cecum. However, the most common and pathogenic species that affects the poultry industry in Pakistan is E. tenella, which results in high mortality (Ayaz et al., 2003; Abbas et al., 2008). The increasing resistance of avian coccidiosis to anti-coccidial drugs currently used by poultry industry together with the requirement for drug and a production systems which should be antibiotic free, it’s now very much necessary to go for the new and advanced methods to prevent this disease. Therefore, scientists started to work on medicinal use of herbal products to control this eimeriosis. In past for treatment of various human and poultry ailments the natural herbal products have been effectively used several times. Because of the anticoccidial nature and antibacterial effect, garlic could be served as a valuable alternative for coccidiostats as a medicinal plant. Pakistan has population of about 64.9 millions of goat, 38.3 millions of cattle, 33.7 millions of buffaloes and 28.8 millionsheep population according to the economic survey estimate. Mutton production is about 643 million tons and beef production is about 1,829 tons out of total 3,379 million tons of meat production. Livestock produces approximately 49,512 million tons of milk out of which buffalo produces 62.0% while cattle produces 35% and sheep-goat produces 1.69% (Economic Survey, 2012-2013). Pakistan is the 3rd largest milk producing country in the world (Afzal, 2010). Diarrhea can be defined as an increased in volume and quantity of fecal excretion. The diarrhea may contain mucous, blood, poor odor and variation in color then the normal feces.On the basis of time period, diarrhea has different types likeacute, constant and chronic. Time Period of acute diarrhea is not more than 14 days while in case of persistent/constant form, it continued for more than 14 days and in chronic case the diarrhea remain for a time period of 28 days(Bazeley, 2003) Diarrhea is a leading factor towards the death of calves. Neonatal calf diarrhea is among the very important diseases of the calves worldwide. It is animportant contribution to the major economic losses in cattle industry. The cause of calve diarrhea may be infectious or non-infectious (Elhassan et al. 2011). Bad hygiene, overpopulation, overfeeding, hot/cold temperature, feeding the calves artificially and the colostrum’s deprivation are all predisposing factor in the complex etiology of the disease (Hemashenpagam et al. 2009). Infectious agent and the co-infection of different multiple pathogens are common in calves having diarrhea. Rotavirus, Cryptosporidium, corona virus and enterotoxigenicE. coli collectively are associated with 75-95% of the infections in calf diarrhea around the world. Clinically, Neonatal calf diarrhea is characterized by profuse acute watery diarrhea, progressive dehydration, acidosis and death within a few days. The degree of dehydration is estimated by the physical examination of calf and has been introduced as a reliable method (Radostits et al. 2009). Experimentally the intestinal potassium and sodium combination can be bothered by different causes and various types of factors. However, electrolytes that are built up in the gastrointestinal tract have a function to reduce the water assimilation. Due to this, the organism losses water and the electrolyte commonly (Smith, 2009) To evaluate the serum electrolytes as well as the acid-base imbalance, there is a need of laboratory tests which cannot be measured regularly in field condition.To check the degree of dehydration is a uniform protocol and is anticipated by body condition score of calves (Naylor 1989). The electrolytes deficiency in neonatal diarrheic calves might be the cause of death in some cases. Althoughit is understood that serum concentrations of potassium (K) and sodium (Na) are very much necessary with reference to the fluids composition used for treatment purpose (Rucker et al. 2008). Trace elements have also important part inthe reproduction, growth and productivity of the householdanimals. The imbalance and Difference in ratio of minerals leads towards both production and economic losses throughout the world. The important symptoms of trace element deficiencies that can be clinically shown in animals are anemia, diarrhea, loss of hair, pigmentation loss, growth disorders in bones, difficulty during walking, a flaky deposit in skin, bending, hyperkeratosis, Para keratosis, lack of appetite, decrease in reproductivity and fertility, young animal growth disorders, Tetany, reduction in protein synthesis, immunity compromised calves, abortion related to non-infectious factors and pica. However, reduction in the concentration of trace elements like cobalt, copper, iron, iodine, manganese, selenium or zinc leads to lower reproduction rates and lactation. Untreated changes in the serum concentrations of Ca, Mg and some trace elements may be the root of cause of some death or post diarrhea complications such growth retardation (Tajik and Nazifi, 2012). The serum values of calcium (Ca) and magnesium (Mg) that is abnormal persists for a significantly long duration after the treatment of neonatal diarrheic calves. The normal serum concentration of Ca in cow has been reported as 2.43-3.1 mmol/L while the mean serum Ca in diarrheic calves reported was 1.297mmol/L. The normal serum concentration of Mg in cow has been reported as 0.74-0.95 mmol/L while the mean serum Mg was reported to be 0.8145mmol/L (Kaneko et al. 2008). There are many advantages of using the hypertonic saline solution (7.5% NaCl; 4ml/kg BW) in diarrheic buffaloes and cattle to resuscitate them. Infusion of Hypertonic saline solution resulted in significant increase in sodium and chloride level in serum. The important feature for the successful and complete resuscitation of hypovolemia along with endotoxaemia in diarrheic buffalo calves is the total amount of sodium. The infusion of Hypertonic saline solution is quite safer and it does cause hypernatremia but not for a prolonged period. So, hypertonic saline solution has advantage of resuscitating the calves from acidemia and hypovolemia. It was also helpful in the improvement of oxygenation and cardiovascular performance (Zafar et al. 2012). The neonatal calf diarrhea can be cured by using the electrolyte therapy like ringer lactate, non-steroidal anti-inflammatory medicines and antibiotics in case of infectious diarrhea and through better farm management depending upon the root of cause (Radostits et al. 2009). Besides, livestock is considered a source of employment generation at rural level. It plays an important role in poverty mitigation and keeps in elevating the socio-economic situation of our rural commonalities. Livestock share approximately 55.4 percent to the agricultural value added and 11.9 percent to national GDP during 2013-14. Anonymous (2013-14). Parasitism is one of the major problems lowering livestock productivity round the earth Vercruysse and Claerebout et al. (2001). The significance of helminthes infections is increased manifold in developing countries like Pakistan where 65.2% population is rural (Population Census Organization (2007) depending upon livestock for their incomes and where parasitism acts as a serious trouble for livestock economy Chaudhry et al. (1984). Among helminthes infections, fasciolosis commonly called as liver fluke disease is of vital importance due to its wider spectrum of ultimate hosts Rondelaud et al. (2001) causing acute and chronic infections Sampaio-Silva et al. (1996). The disease is primarily caused by Fasciola hepatica and Fasciola gigantica Soulsby et al. (1987). Fasciolosis is a parasitic liver infection of wild and domestic ruminants caused by genus Fasciola, which has worldwide distribution Soulsby et al. (1986). Fasciolosis causes economic losses as a result of mortalities, abortions, retard growth, reduced milk and meat production, condemnation of infected liver and emaciated carcasses and cost of animal treatment Gracey and Collins et al. (1992). Fasciolosis caused by F. hepatica is a cause of important economic losses in ruminants. The disease is widely spread throughout in Africa, Asia and the Pacific. Although mostly a problem of young stock, the disease is also found in older animals where it adversely affects health, growth rate and development. Apart from its great veterinary importance throughout the world, fasciolosis caused by both F. hepatica and F. gigantica has recently been shown to be a re-emerging and wide spread zoonosis affecting a number of human populations Mas-Coma and Bargues et al. (1997); Esteban et al. (2003). Prevalence of fasciolosis infestation in buffaloes is 49.01%. The infestation of fasciolosis was optimal from January to September, while least during October to December. Fecal samples indicated F. hepatica prevalence 65%. Whereas prevalence of F. gigantica was observed higher in adult cattle as compared to cattle calves by fecal egg counts Khalil et al. (2009). 4% Buffalo calves infested with trematodes. Availability of intermediate host and the grazing habits of the final host determines the epidemiology and seasonal pattern of infection with trematodes Pfukenyi et al. (2005); Bhutto et al. (2002). The life cycle of F. hepatica includes many stages which develop in the environment or the intermediate host. Metacercariae which is the infective stage encysted and ingested by grazing herbivores. Temperature and rainfall are the important factors affecting the life cycle and the prevalence of F. hepatica infection McCann et al. (2010). Many studies revealed that F. hepatica is endemic in buffaloes, cattle, goats, sheep and humans in Pakistan Ijaz et al. (2009); Iqbal et al. (2007); Khan et al. (2010) and Qureshi et al. (2005). Fasciolosis is the disease by which about 250 million sheep and 300 million cattle are potentially affected worldwide Boray et al. (2005), and more significantly infects all ruminants Iqbal et al. (2007). F. hepatica and F. gigantica are the two liver flukes commonly reported to cause fasciolosis in ruminants Walker et al. (2008). Infected cattle can exhibit Introduction 3 poor weight gain and dairy cattle have lower milk yield, and possibly metabolic diseases Mason et al. (2004). Losses due to fasciolosis in the United Kingdom and Ireland alone are more than £18 million a year Mulcahy and Dalton et al. (2001); €52 million a year or €299 per infected animal in Sweden Schweitzer et al. (2005); 0.26 million USD annual due to fasciolosis associated liver condemnations in cattle slaughtered in Kenya Kithuka et al. (2002). In Pakistan, 35697 USD (3141360 PKR) are reported in only one tehsil of Punjab province, the Sargodha T.U. Rehman et al. (2013). While Fasciolosis is prevalent in whole Punjab province of Pakistan but the marshy areas such as regions of Gujranwala, Lahore, Multan, Faisalabad, Jhang, Muzaffargarh, Sheikhupura, and Sargodha are of major concern. Fascioliasis in ruminants causes substantial economic losses, estimated at US$ 2 billion per annum worldwide, to rural agricultural communities and commercial animal producers due to death of infected animals, condemnation of affected livers and production losses associated with reduced feed conversion efficiency Urquhart et al. (1996); Spithill and Dalton et al. (1998). In tropical countries, fascioliasis is prevalent up to 90% and is considered the most important helminthes infection of cattle Spithill and Dalton et al. (1998). Most important, human can also become infected with Fasciola spp. and the disease is referred to as human fasciolosis. It is estimated that 2.4 million people in more than 60 countries are infected and the number of people at risk is more than 180 million throughout the world Haseeb et al. (2002); Mas-Coma et al. (1999); Ishii et al. (2002). Khalil et al. (2009) recorded 49.01% prevalence of fasciolosis infestation in buffaloes. He also observed that optimum infestation was seen from January to September, while minimum during October to December. Whereas Bhutto et al. (2002) recorded 4% buffalo calves infested with trematodes. Introduction 4 Morphological identification of Fasciola species requires significant parasitological skills and is not an accurate method of characterization, especially for the ‘intermediate’ form Kendall et al. (1965); Lin et al. (2007); Le et al. (2008). Hence, different molecular tools have been developed during the last decade for the accurate identification of Fasciola spp. Marcilla et al. (2002); Velusamy et al. (2004); Cucher et al. (2006); Magalhaes et al. (2008); Ai et al. (2010). Conventional techniques like fecal examination used for its diagnosis are not comprehensive because of Species similarity in distribution in many countries of East and North Africa, and Southeast and Central Asia, and are similar in egg morphology. Coprological methods continue to be the most widely used approach for the detection and quantification of Fasciola and other helminth eggs in laboratory animals, domestic and sylvatic animals and humans Cringoli et al. (2004, 2010); Bergquist et al. (2009), with a more recently developed method that is currently undergoing broad-scale validation the FLOTAC techniques Cringoli et al. (2010) for the detection and quantification of F. hepatica eggs in feces obtained from experimentally-infected rats before and after drug administration. FLOTAC has been used successfully in the diagnosis of F. hepatica infections in naturallyinfected sheep, which underwent treatment with standard Cringoli et al. (2006). However PCR based diagnosis using specie-specific primers has been proved more accurate and successful in diagnosis and distinction of Fasciola species Le TH et al. (2012). The molecular technique such as PCR is used for the recognition of parasite. These molecular techniques are largely accepted all over the world. These are more exact methods than traditional in epidemiological studies Heckeroth and Tenter et al. (1999). Herbal drugs have been used since ancient times to cure diseases and medicinal plants have been used to treat fasciolosis among these Nigella sativa and Allium sativum have been used to treat the worm infestation. No report of resistance development to plant anthelmintic although many have been used in veterinary medicine for many years Ashaal et al. (2010); Introduction 5 Waller et al. (1995); Koko et al.(2000); Thienpont et al. (1979); Keiser et al. (2011). Herbal (indigenous) drugs have been used since ancient times to cure diseases, and several medicinal plants have been used to treat fasciolosis Satyavati et al. (1987). Among these, Nigella sativa has been used to treat worm infestation Nadkrani et al. (1954; Said et al. (1969); Akhtar et al. (1988). N. sativa can be used as a therapeutic agent against helminthes Khan et al. (2013). Neem leaf powder can be used as therapeutic agent against gastrointestinal parasites of camel, equine, cattle, buffalo and small ruminants Mehmood et al. (2013). (Palacio J et al., 2003).Animal bites, particularly dog bites, are a major public health problem throughout the world (Morgan and Palmer, 2007). Animal bites and scratches, even when they are minor, can become infected and spread bacteria to other parts of the body. Dog attacks, by street or domestic dogs, with injuries from very minor to significant and severe to fatal, are not uncommon. According to WHO report, ten million people are bitten by animals (especially Dogs) around the world, considered for prophylaxis and treatment against rabies and almost (55,000) people die from this disease annually (Mohdjunaid et al., Oct 2012). 6.2 Hypothesis: The incidence of dog bite injuries is high in densely populated towns of Lahore when compared to less populated towns of Lahore. 6.3 Methodology: General information about patients and possible causes was collected on a structured questionnaire. All the dog bite victims that were come to emergency ward during three months period were included in the study. Questionnaire include closed questions about the demographics of the victims, circumstances of bite incidents, body parts injured and the degree of injury, type of dog, history of previous bites, the level of knowledge about rabies, post bite home treatment (washing of bite wound etc.) prior to visited the hospital for medical treatment and post exposure treatment at hospital. The densely populated area (ravi town, data ganj bakhsh, shalimar town, samanabad etc) and less populated area (gulberg, azizbhati town, wagah, Allama iqbal town, & nishtar) was be compared. 6.4 Statistical Design Cumulative incidence was calculated as described by Leon Gordis (2008). Chi square test was applied on the data by using SPSS (version 19.0). 6.5 Results In this study, cumulative incidence is 4.653 per 1000 popluation, most of the dog bite victims were male 79.9 % as compare to female 20%.5.7% cases visited Mayo Hospital’s emergency department during the study period, 4.5% cases were reported in Services Hospital and 89.9% cases from IPH Lahore. In this the higher percentage was the patients who were illiterate (47%) and very few patients were those who had intermediate and above education (6%). The occupation of the patients who were attended Hospitals, were farmers (31.56%). The 24% patients were those who had bitten once before in previous years. In those 24% patients 19.2 % those who were bitten by dog and 5% were bitten by others (cats, horse, donkeys, etc). Most of the victims were bitten in rural area (65%) and most of the injuries were from stray dogs (65.9%). Most of the victims had provoked bite (74%) and the high frequency were recorded that the cases had type II wound category (deep scratch but no bleeding) 82%. The cases received treatment in different hospitals were RIG (18%), Tetanus toxoid (32%), Antibiotic (78%) and Suturing (7.9%). Most of the patients were from Ravi town Lahore that is one of the highly densely populated areas of Lahore (38%) and very few cases reported from Nishtar town Lahore (0.5%). The demographic location and the hospitals had no association as the results were insignificant (p-value>0.05). Most of the cases who were bitten by dog and they already bitten by dog in previous years, significant in relation as p-value <0.05. The association between dog status and biting animal was significant, as mostly biting dogs were stray dogs and most of the bite was provoked, there was significant association between biting animal and biting type (p-value<0.05). 25. Comparative Efficacy Of Anthelmintics Against Gastrointestinal Nematodes In African Lion (Panthera Leo)

A survey was carried out to estimate the prevalence of gastrointestinal nematodes in captive African lion and it was found that toxocara canis, toxocara mystax and toxascara leonina are the most commonly prevailing gastrointestinal nematodes in captive African lions in above mentioned places. The overall incidence rate of toxocara canis was 82.5 %, of toxocara mystax was 60% and of toxocara leonina was 50% respectively. The Anthelmintics trials were conducted on 14 animals which were divided into 2 groups A and B each having 7 animals. Group A was treated with Zental (Albandazole), group B was treated with Vormox (mebendazole), The percentage efficacy of each drug was determined on the basis of reduction on number of eggs per gram of discharged in faeces at day 7, 10, 13, 16 and 22 after deworming. In group A, decrease in EPG of toxocara canis in group A was 21.1% at day 7, 37.2% at day 10, 54.7% at day 13, 74.6% at day 16 and 88.8% at day 22. Decrease in EPG of toxocara mystax was 15.9% at day 7, 38.1% at day 10, 56.6% at day 13, 70.6% at day 16 and 82.7% at day 22.similarly decrease in EPG on toxocaris leonina was 15.6% at day 7, 29.2% at day 10, 45.6% at day 13, 61.1% at day 16 and 83.1% at day 22 respectively In group B, decrease in EPG of toxocara canis in group A was 7.24% at day 7, 14.57% at day 10, 23.26% at day 13, 31.27% at day 16 and 38.08% at day 22. Decrease in Summary 25 EPG of toxocara mystax was 16.38% at day 7, 22.4% at day 10, 29.6% at day 13, 34.3% at day 16 and 41.2% at day 22.similarly decrease in EPG on toxocaris leonina was 13.6% at day 7, 21.29% at day 10, 30.8% at day 13, 39.1% at day 16 and 47% at day 22 respectively. To study the gastrointestinal helminths, 384 camels of different age, sex and breed was examined coprologically. For this, five gram of fresh fecal sample was carefully collected into a sealed container from each camel and was transferred to Medicine lab, UVAS, Lahore in containers with ice packs. Fecal samples were scored 1-3 based on the consistency. The individual samples was triturated in saturated salt solution, sieved and examined for helminths eggs by using different techniques i.e. Direct Smear Method, Sedimentation technique and Floatation technique while the eggs count was performed by McMaster technique. Among the camel population, the current study indicates that in Marrecha breed the prevalence of Nematodes was Trichostrongylus 1.77%, Haemonchus 8.44%, Nematodirus 3.11% and Trichuris 7.11% respectively. Mixed infestation of all the nematodes was found 26.66% in infested camels, while the prevalence of Camelostrongylus, Trichostrongylus, Strongyloides, Haemonchus, Nematodirus and Trichuris in Barella breed was 4.40%, 1.88%, 2.51%, 6.91%, 2.51% and 7.54% respectively. Mixed infestation of all the nematodes was found 25.78% in infested camels. The prevalence of Fasciola hepatica was found higher in Marrecha breed as compared to Berrela breed. It was 18.66% in Marrecha breed and 14.46% in Barella breed while the overall prevalence of Fasciola hepatica in the camels irrespective of the breeds was 16.93%. While the prevalence of Fasciola gigantica was also found higher in Marrecha breed than Barella breed. It was 16% in Marrecha breed and 10.69% in Barella breed while the overall prevalence of Fasciola gigantic in the camels irrespective of the breeds was 13.80%. The overall prevalence of fascioliosis in camel is 30.31. The prevalence of Moniezia expansa was found higher in Marrecha breed as compared to Barella breed. It was 10.22% in Marrecha breed and 8.80% in Barella breed while the overall prevalence of Moniezia expansa in the camels irrespective of the breeds was 9.63%. The sex wise prevalence of gastrointestinal Nematodes in males was 39.28% which includes Camelostrongylus 8.03%, Trichostrongylus 3.57%, Strongyloides 6.25%, Haemonchus 11.60%, Nematodirus 4.46% and Trichuris 5.35% respectively while in the female it was 25.37% which includes Camelostrongylus 4.41%, Trichostrongylus 2.20%, Strongyloides 2.94%, Haemonchus 6.25%, Nematodirus 2.57% and Trichuris 6.98%. The overall prevalence of Nematodes in Male was found higher as compared to Female which was 39.28% and 25.37% respectively. Sex wise prevalence of GI Trematodes in Camels was determined through collected samples. The prevalence of Fasciola hepatica was found higher in Female as compared to Male. It was 16.91% in Female and 15.18 % in Male while the overall prevalence of Fasciola hepatica in the camels irrespective of the sex was 19.53%. The prevalence of Fasciola gigantica was also found higher in Female than Male. It was 12.13% in Female and 11.61% in Male while the overall prevalence of Fasciola gigantica in the camels irrespective of the sex was 11.98%. Sex wise prevalence of GI Cestodes in Camels was determined. The prevalence of Moniezia expansa was found higher in Female as compared to Male. It was 14.34% in Female and 13.39% in Male while the overall prevalence of Moniezia expansa in the camels irrespective of the sex was 14.06%. The overall prevalence of Nematodes in age group >10 yrs. was higher 30.25% as compared to age group 5-10 yrs. 20% which includes Camelostrongylus, Trichostrongylus, Strongyloides, Haemonchus, Nematodirus and Trichuris 3.36%, 2.52%, 6.72%, 5.88% 4.20% and 7.56 in age group >10 yrs and 1.88%, 1.51%, 1.88%, 4.90%, 2.64% and 7.16 respectively, in age group 5-10 yrs. The prevalence of Fasciola hepatica in age group 5-10 yrs was found higher as compared to age group >10 yrs. It was 30% in age group 5-10 yrs and 16.99% in age group >10 yrs while the overall prevalence of Fasciola hepatica in the camels irrespective of the age groups was 18.75%. The prevalence of Fasciola gigantica in age group 5-10 yrs was found higher as compared to age group >10 yrs. It was 9.33% in age group 5-10 yrs and 7.55% in age group >10 yrs while the overall prevalence of Fasciola gigantic in the camels irrespective of the age groups was 8.59%. The prevalence of Moniezia expansa in age group >10 yrs was found higher as compared to age group 5-10 yrs. It was 10.92% in age group >10 yrs and 10.18% in age group 5-10 yrs while the overall prevalence of in Moniezia expansa the camels irrespective of the age groups was 7.81%. To study the effect of gastrointestinal helminths on various blood parameters of camels, blood samples were collected from 5 infected and 5 healthy camels. Samples were analyzed using Hematology Analyzer and results were compared. For therapeutic trials, a total of 30 camels positive for helminths was taken and divided into three groups A-C each group comprising of 10 animals. A fourth group D was selected comprising of 10 uninfected camels. The camels of group A were treated with dry Neem (Azadirachta indica) leaf powder at a daily oral dose of 100 gm/camel for five days, group B was treated with Albenzole granules® (Albendazole) at a dose rate of 15mg/kg bd. wt. PO once. Group C (untreated) was designated as positive control, and the camels in Group D as negative control. Efficacy was determined on the reduction of eggs in feces at day 3, 7 and 14 (post-treatment). In the current study Neem leaves and Albendazole gave following results in the camels affected with Helminths. Neem leaves cured 20% of the animals on day 3rd after the drug administration while 40% and 60% of the animals cured on the day 7th and 14th respectively. Albendazole also gave good results against helminths affected camels. On the day 3rd of the treatment 30% animals cured while 60% animals were cured on the 7th day and on 14th day 80% animals recovered. Data regarding prevalence was measured by non-parametric, chi-square (χ2), while therapeutic trails were analyzed by repeated measures one way ANOVA, using SPSS (Statistical package for social sciences), P < 0.05 was considered significant. Tick infestation is a common problem in dogs and its transmitted infections usually tick borne hemoparasitic diseases are emerging as a worldwide problem not just for animals but also for humans. Due to this phenomenon, ticks are now recognized as a major cause of various diseases in of urban environments. The current research has been conducted to study the prevalence of tick infestation, associated risk factors and chemotherapeutic trials of topical acaricides were used in order to study comparative efficacy, were conducted in dogs. Furthermore, prevalence of tick borne hemoparasitic diseases was also observed by performing microscopic blood smear examination in dogs of Lahore district. A total of 300 dogs (n= 200 positive for tick infestation; n= 100 negative for tick infestation) having tick infestation were included in the present study. Dogs of different sex, age and breed were selected for tick collection. The ticks were collected with the help of forceps from the target sites of each dog, while adopting all necessary measures to avoid damage to the mouth parts of the ticks and skin of the host. The collected ticks were preserved in plastic containers containing 70% ethanol, which was separate for each specimen. Ticks were identified under stereoscope by observing morphological characteristics of each tick, according to the key. Blood sample for tick borne hemoparasitic diseases was drawn from cephalic vein of each dog using disposable syringe and was poured into EDTA vacutainer. Thin blood smear slide was then prepared from the blood of the animal, dried in air and was stained using Geimsa’s staining method. The hemoparasites observed in the blood of the dogs were then characterized and Summary 51 identified by their morphological characteristics. For chemotherapy, a total of 30 tick infested dogs of different breeds viz. Rottweiler, German shepherd and Labrador was used in therapeutic trials. Each breed of dog was divided into 2 subgroups which were then treated with topical ivermectin and fipronil 1500μg/kg b.w and 15mg/kg b.w respectively. Efficacy of the drugs was then measured on the basis of disappearance of clinical signs and reversal of tick infestation at day 2, 4 and 6 of post-medication. Data regarding the prevalence of tick borne hemoparasitic diseases and tick infestation was analyzed by using Non-parametric, Chi-square test whereas data on therapeutic trials was analyzed by Z-test, using statistical package for social science (SPSS). P < 0.05 was considered significant. The results obtained showed us that the study area is at a risk of tick infestation and tick borne hemoparasitic diseases. In dogs, Rhipicephalus sanguineus (brown dog tick) species was observed and identified as a major tick infesting dogs of the study area during the course of study. Various determinants including tick density and its genus, host age, sex, predilection site host breed and season significantly affected the tick infestation in the study area. The prevalence for tick infestation was found highest in Pastoral dogs as compared to other breed types found in dogs. Among breeds German shepherd showed highest prevalence for tick infestation. Adolescence age group among dogs was more prone to tick infestation. The number of males affected by ticks was highest as compared to females in dog population. Among the dog population, majority of the animals infested by ticks were categorized under mild infestation. The most favorable predilection site for tick infestation in dogs was ear pinnae, followed by head and then neck. Monsoon was the most favorable season for tick infestation in dogs. September was found to be highest favorable month for tick infestation in dogs. Summary 52 In tick borne hemoparasitic diseases, babesiosis was the highest prevalent disease in dogs (63.33%), followed by trypanosomiasis (0.66%) and mixed infection (0.33%). Age group among dogs showed non-significant factor affecting prevalence for TBHD’s. The number of affected dogs for babesiosis was greater in adolescence age group as compared to other age groups. On the other hand, trypanosomiasis and mixed infection adult age group was found highly susceptible. Males of canine group were slightly more susceptible than females in case of babesiosis. Similarly for trypanosoniasis and mixed infection males were more prone than females. For babesiosis, monsoon was the most favorable season in dogs. Whereas for trypanosomiasis and mixed infection, post monsoon was found to be favorable season in dogs. In breed type of dogs, prevalence for babesiosis was found highest in Pastoral dogs. In dogs the prevalence for babesiosis was found highest in German shepherd breed, whereas for trypanosomiasis and mixed infection Cross breed was the most susceptible breed. For trypanosomiasis and mixed infection Cross breed was the most susceptible breed type. The prevalence for babesiosis was found highest in the month of September. For trypanosomiasis and mixed infection, the month of October was found to be highest susceptible month for dogs. Keeping in view the importance of foal diarrhea and its impact on the equine industry the current study was designed to determine serum concentration of electrolytes (Ca, K, Na) and trace elements (Li, Cu, Fe) along with the associated risk factors in diarrheic foals. The present study was designed to examine diarrhea in foals at various equine stud farms, private and public veterinary hospitals. For this purpose a total of (n=100 foals) suffering from clinical diarrhea irrespective of the cause of diarrhea selected randomly after clinical examination and (n=10 foals) negative for diarrhea as control were included. Data of each diarrheic foal was collected separately in a data capture form. Blood and serum sample were collected for hematological and serum biochemical studies, respectively. Concentrations of serum electrolytes (Ca, Na, and K) were analyzed by Flame Photometry and trace elements (Cu, Li, and Fe) were estimated through Atomic Absorption spectrophotometer. PCV was determined by micro hematocrit method. Risk factors including species and age were analyzed for association. Foals which were suffered from diarrhea showed increase in Pack cell volume. Serum concentration of Sodium, potassium, calcium, iron, copper and lithium were measured in diarrheic foals by atomic absorption and flame photometer method and were compared with healthy group of foals.Foals having diarrhea showed an acid baseimbalance as a result decrease in serum electrolytes and trace elements. The blood acid-base balance is precisely controlled because even a minor deviation from the normal range can severely affect many organs. The body uses different mechanisms to control the blood's acid-base balance.Sodium and potassium and calcium are important electrolytes imbalance in diarrhea. Potassium level was increased in foals having diarrhea. Calcium concentration in ppm significantly decreased from its normal value. In some diarrheic foals increased in level of calcium was also observed because these foals have blood in their feces that result in the calcium to come in blood as a clotting factor. Trace elements such as Cu, Fe and Li are needed in a very little concentration for many physiological functions. The serum iron was measured in foals having diarrhea.The iron concentration was less than normal value.The deficiency of iron lead to anemia. Clinical signs of anemia because of irondeficiency are rarely seen in foals. Therefore, mineral imbalances influence iron metabolism and may cause iron deficiency in foals. The serum copper concentration was significantly decreased in the diarrheic foals. Copper concentration is also related to the packed cell volume as in anemia copper deficiency is a significant feature. Copper is important element and its deficiency lead to decrease in serum concentration of iron. This decreased level might be because most of the circulating copper in plasma is attached to the serum glycoprotein, ceruloplasmin. Ceruloplasmin has ferroxidase activity and may be required to deliver iron into the circulation Therefore it was observed that low iron concentrations might have resulted from copper deficiency.The concentration of Lithium was also decreased in diarrheic foals when compared to healthy foals. Exuberant granulation typically is an irregular and unhealthy appearing tissue with many grooves and clefts. Histologically exuberant granulation tissue has remains of fibrin deposits, which have not been cleared by the acute inflammatory response. Horses are predisposed to trauma wounds that can be labour intensive and expensive to manage. Wounds localized at the trunk region are less problematic and heal faster than wounds located at the distal aspect of the limb. Complications such as wound infection, formation of exuberant granulation tissue (EGT) and hypertrophic scarring are frequent. The EGT or “Proud Flesh” is considered similar to the keloid of the human skin and together with wound contamination/infection, is currently the most complicated aspect of wound management in equines. The present project was designed to evaluate comparative efficacy of surgical and chemical debridement of such kind of wounds in horses. The clinical study was conducted on 20 already traumatized horses of either sex. The animals were divided into four groups (I, II, III and IV) comprising five animals each. These animals were kept in S.P.C.A, University of Veterinary and Animal Sciences, Lahore. In a group I, Surgical excision was carried out to remove excessive growth from wounded area. Local anaesthesia with two percent lignocaine was in filtered for removal. Antiseptics were applied and over-granulation was smoothly removed from one margin of skin to other. Group II animals were treated with ten percent copper sulphate ointment, Group III animals were treated with two percent silver nitrate ointment and Group IV animals were treated with white lotion. Each technique had its own advantages and disadvantages. The results proved that surgical removal was a best choice for management of hyper-granulated wounds. It also had good scar formation SUMMARY 72 and faster recovery as compared to other chemical agents. It allows the closure of large wounds and faster epithelization to margins. The results of present study clearly indicated that surgical excision of hyper-granulated wound on early stages yields good recovery and proper union. Although silver nitrate has better penetrating power in wounds but it has no ability to epithelize the wound which terminates in slow recovery. Moreover silver is an expensive and less affordable to a common man. Copper sulphate has good recovery but it causes irritation while application and animal feels pain. A successful vaccination campaign depends on the serotype identification and specific vaccination against the prevalent serotype of the virus. The present study was designed to evaluate comparative potency of two anti-FMDV vaccines (UVAS-FMD, Deccivac Intervet) used in cattle around areas of Lahore Pakistan. Blood samples were taken from vaccinated animals on day 0, 30, 60 and 90 post priming. Antibody titer was evaluated with different route of administration and various adjuvant based vaccines. Four animal groups were made each containing 5 animals; in group 1 UVAS vaccine was used by Sub/ Cut route (gel based) at priming dose, followed by Intra Muscular (oil) on 30th day. In group 2 UVAS vaccine was given I/M (oil) as priming dose and booster (oil) I/M. In group 3 Deccivac (oil) vaccine was used I/M for priming and boosting. In group 4 Deccivac (oil) vaccine was used as Sub/ Cut for priming and boosting. The results revealed non-significant difference (p>0.05) among the four different groups administered with FMDV vaccines when evaluated at day 30 and significant difference (p<0.05) at day 60 and 90 post vaccination. Analysis of variance showed significant difference (p<0.05) in antibodies between groups and with in groups at day 60 and 90. Gel based vaccine gave quick antibody response which later maintained with oil based booster dose. The difference in antibody titers obtained in the present study was found non-significant (P>0.05) between the antibody titers of FMD trivalent vaccine of UVAS and Deccivac at 90th day of inoculation. There was significant difference (p<0.05) between the adjuvants of vaccine. Animals inoculated with priming dose of gel based vaccine followed by oil based boosting showed significantly high anti FMD antibody titer than animals inoculated with oil for both priming and boosting. There was significant Summary 54 difference (p<0.05) between the groups vaccinated with various routes of administration. The animals inoculated with priming dose through s/c followed by boosting dose i/m showed significantly high anti FMD antibody titer at 90th day of inoculation compared with those inoculated intramuscularly for both priming and boosting. The animals inoculated with oil based vaccine for both priming and boosting through S/c showed marked significant decreased in anti FMD antibody titer. The route of administration revealed significant difference (p<0.05) in antibody response within groups and between groups at day 30, 60 and 90. In all three readings the mean for sub/cut priming and IM boosting were found significantly high (p<0.05) as compared to other routes. A total of 300 nail samples were collected from the fingernails of the school children of different age and sex. The nails were swabbed into a clean sterile container containing normal saline. Finger nail clippings were collected from both hands of each subject using sterile nail clippers and were placed in labeled containers containing normal saline. Samples were examined by direct microscopy using Lugol’s iodine and Eosin preparations. Sediments from the nail clippings were examined after digestion with 10% Potassium hydroxide and centrifugation at 2500rpm for 5minutes.The parasites were isolated from the fingernails of the primary school children of both sexes and prevalence was recorded. The collected data was analyzed using SPSS version 20. The prevalence of parasites was presented as descriptive statistics, while the relationship between several variables (age, gender,educational level) and presence of parasites was determined by the chi-square test. The level of significance used was P<0.05. This study will help to create awareness among society especially school children about parasites and parasitic infections. This study has been designed to check anthelmintic resistance against haemonchosis of sheep by an in vivo method. The current study was carried out at Parasitology laboratory (Toxovacc lab), Department of Parasitology, University of Veterinary and Animal Sciences, Lahore. 100 faecal samples were collected from sheep at Livestock Research and Development Station, Paharpur, D.I. Khan. Animals were drenched with anthelmintic (Albashell containing Albendazole 2.5%, administered @10 mg/kg of body weight) orally after 1st sampling, at 0 day. The faecal samples were examined microscopically, micrometery was exploited and EPG analysis was performed by using McMaster technique. After 14 days, the second sampling was done. The fecal samples were brought and stored at 4°C in Parasitology laboratory (Toxovacc lab). Pre-trial & Post trial EPG were compared and positive samples were taken (tag#1057 Damani sheep male, tag#13 Balkhi sheep male, tag#1096 Damani female, tag#06 Balkhi female, tag#20 Balkhi) for egg isolation (Module, 2004) for egg DNA extraction through classical method of Phenol-Chloroform-Iso-Amyl Alcohol extraction. DNA samples were subjected to polymerase chain reactions (PCR) targeting β tubulin gene for detection of benzimidazole resistance at genetic level. Fecal egg count reduction percentage of 74.57% at day 14 post treatment clearly shows the presence of benzimidazole drug resistance in parasites infecting Balkhi and Damani sheep at Livestock Research and Development Station, Paharpur, D.I. This study was done to evaluate the effect of different anthelmintics on nematode control in different parity cattle at calving and their effect on milk yield. In this study, we screened cattle of 1st and 2nd parity for nematodes. Animals were randomly selected in each group. In first parity animals, we found 23 positive cattle which were divided into 3 different groups while in second parity animals, we found 20 positive cattle which were divided into 3 groups. We used Albendazole (Velbazine) @ 10mg/kg body weight and levamisole (Nilverm) @ 7.5mg/kg bodyweight for treatment. In this study both drugs were found effective in controlling nematodes infections in cattle. Percentage reduction of epg by albendazole was 48.20%, 85.34%, 93.90% and 51.54%, 81.43%, 91.74% on day 7, 14 and 21 in 1st and 2nd parity animals respectively. Percentage reduction of epg by levamisole was 44.45%, 76.92%, 88.03% and 46.60%, 73.78%, 85.43% on day 7, 14 and 21 in 1st and 2nd parity animals respectively. The increase in milk production in current study was 0.39 liter per day and 0.92 liter per day in 1st and 2nd parity cattle respectively. Albendazole increased the milk fat by 0.07 %, while levamisole decreased 0.02 % in 1st parity cattle. In 2nd parity cattle, there was sufficient increase of 0.1% of milk fat by albendazole while levamisole has decreased 0.05 %. We conclude from our studies that anthelmintic treatment of recently calved cattle have significant effect on milk production due to the nematode control. Milk production increased significantly in 2nd parity friesian crossed cattle following anthelmintic treatment as compared to Summary 70 1st parity animals. Levamisole has negative effect on fat concentration in friesian crossed cattle while albendazole treated cattle has positive effect. Albendazole has been found more efficient in reducing EPG of helminths in both parity animals as compared to levamisole treated animals. It is recommended that cattle should be checked for GIT parasites by faecal examination on regular basis so that cattle should be dewormed when needed. The segmented, negative strand RNA viruses that form the family Orthomyxoviridae are divided into three types of influenza virus, A, B and C. Only influenza A viruses have been reported to cause natural infections of birds. The outbreak can be mitigated by applying biosecurity measures, controlling poultry movement, using inactivated vaccines and initiating an AI surveillance network throughout the country. A targeted survey was conducted for a period of 3 months in order to determine prevalence of Avian Influenza H5, H7 and H9 in the villages of Kasur district. Two stage cluster sampling without replacement was adopted in this study. Thirty clusters were selected and in each cluster, seven elementary units (chicken) were sampled, i.e. a total of 210 birds. Tracheal swabs were collected from live and apparently healthy backyard birds then swabs were stored properly at 4°C (24-48hours) until processed. Data were collected from the owner in a face to face interview. A detail predesigned questionnaire was filled after taking written consent from the owner. The sample was collected during the survey of backyard poultry in villages of Kasur district was processed for laboratory analysis. Polymerase chain reaction for Avian Influenza virus isolation was conducted to diagnose sample for AIV. The weighted proportion estimate with 95% Cl (confidence intervals) of the overall prevalence was computed by using “R” software. Multivariable logistic regression was conducted to estimate the effect of each study variable on the outcome. Variables with significant univariate relationship at p <0.25 was selected for inclusion in the final model. Odds ratios (ORs) and corresponding 95% Cl (confidence intervals) were calculated (Hosmer and Lemeshow, 2000). CHAPTER 6 SUMMARY Discussion 53 The management practice risk scores provide a means to quantify the level of risk to avian influenza across villages. However, the utility of such a measure cannot be assessed until these are related to AI status form clinical tests. Nevertheless it is useful to assess what factors are associated with these scores. In particular, specific profiles of farms can be identified with potential high risk and control resources allocate accordingly. For example, this study has found amongst other things that farms with mixed poultry breeds, which have reported diarrhea, or reported high mortality have high risk scores, and these might be targeted for intervention. Prevalence estimate of Avian Influenza was generated. Potential risk factors associated with this prevalence was identified and will be communicated to concerned persons through publication. The poultry birds reared completely outdoor have more chance to contact with wild birds which increase the occurrence of AIV. A strong association between source of water and AIV was found. Water source can play a vital role in transmission of AIV. Another strong association was found in unethical disposal of dead birds and waste poultry farm near to houses with increase the chances of infection. Presence of live bird market stall and visiting of poultry farm vehicle to villages can increase the chances of AIV infection. Elimination or reduction of above mentioned risk factors, use of disinfectant for cleaning and regular vaccination against AI could significantly improve backyard poultry production system in villages. 37. Prevalence And Chemotherapy Of Bovine Anaplasmosis In District Mirpur Azad Jammu And Kashmir

Piroplasms belongs to this complex and affects both large and small ruminants with high morbidity and mortality rates resulting in heavy economic losses and thus poses a serious risk to livestock production. A total of 200 blood samples of bovine, cattle (n=100) and buffalo (n=100) showing the signs of fever, progressive anemia, a marked decline in body weight, depression and debility from district Mirpur AJK were included in the study. The diagnosis was made through thin blood smear examination. The overall prevalence was found 15.00% in both species of animals. The prevalence in cattle and buffaloes revealed 22% and 08% respectively. The results showed significant difference (P<0.05) in prevalence between cattle and buffaloes. The gender wise prevalence of the disease revealed 12.12% in male and 26.87% in female cattle whereas; these values were 6.45% in male and 8.70% in female buffaloes. Chi-square analysis showed significant difference (P<0.05) between male and female animals in the area. The data on breed wise prevalence of anaplasmosis showed highest prevalence in exotic breeds (28.00%) followed by cross breed cattle (24.44%) and native breed (16.67%) of AJK. The prevalence was 5.71% in Kunddi breed of buffalo and 9.23% in Nili Ravi buffaloes. Chi-square analysis showed significant difference (P<0.05) between breeds of animals. Three different age groups of cattle and buffaloes were analyzed for the prevalence percentage of anaplasmosis in the area. The data showed highest prevalence (35.48%) in 1-3 year age group of animals followed by 18.92% in 3-5 year and 12.50% in age group 5-7 year in case of cattle and 14.29%, 6.67% and 5.88% in buffaloes respectively. the analysis of the data revealed a significant difference (P<0.05) among different age groups. The values of hemoglobin percent, packed cell volume and total Summery 40 erythrocyte count were found increased significantly (P<0.05) in cattle and buffaloes infected with anaplasmosis whereas; total leukocyte count was decreased significantly. The parameters were tested through student’s T-test. The analysis showed significant difference of values of all parameters in normal and infected animals. The chemotherapeutic trials were conducted with two drugs against bovine anaplasmosis in clinically diagnosed cases. Twelve positive cases of each cattle and buffaloes were divided into two main groups A and B comprising of 06 animals in each group. Each group was further divided into two sub groups comprising of 03 animals in each sub groups. The group A was treated with Oxytetracycline @ 20 mg/kg B.W. I/M the efficacy of the drug was evaluated on the basis of disappearance of Anaplasma in the blood smear. The efficacy percentage of Oxytetracycline was 33.3, 33.3, 66.7, and 100 at 2nd, 4th, 6th and 8th day respectively post treatment in cattle whereas; 0.00, 33.3, 33.3 and 66.7 respectively in buffaloes. The group B was treated with Calotropis procera (Aak) at the dose rate of 0.3 mg/kg body weight orally. The efficacy percentage of Calotropis procera (Aak) was 0.00, 33.3, 66.7, and 66.7 at 2nd, 4th, 6th and 8th day respectively post treatment in cattle whereas; 0.00, 0.00, 0.00 and 33.3 respectively in buffaloes. The efficacy of Oxytetracycline against bovine anaplasmosis on day 08 was found 83.33% whereas; of Calotropis procera was 66.66%. Malondialdehyde (MDA) is a byproduct of lipid peroxidation and causes irreversible damage to sperm structure and function. In buffalo, blood plasma MDA level increases with age. Therefore, we hypothesized that MDA level in buffalo bull semen will increase with age and will affect the semen quality. The objective of the study was to compare MDA level and quality of fresh and frozen-thawed semen in aged vs. young Nili-Ravi buffalo bulls. Single ejaculate was collected on weekly basis for four weeks from aged (13.6±1.0 years; n=3) and young (3.4±0.3 years; n=3) Nili-Ravi buffalo bulls. MDA level was estimated through thiobarbituric acid assay (TBA) in fresh and frozen-thawed semen. The quality of fresh and frozen-thawed semen was estimated through sperm motility, viability, DNA and acrosome integrity. MDA level (nmol/ml) did not differ (P>0.05) between aged vs. young bulls in fresh (2.3±0.2 vs. 2.9±0.7) and frozen-thawed (53.1±2.8 vs. 48.4±2.6) semen, respectively. In fresh semen, sperm motility and concentration did not differ (P>0.05) in aged vs. young bulls; however, the volume of fresh semen increased (P<0.05), while sperm viability and DNA integrity decreased (P<0.05) in aged vs. young bulls. In frozen-thawed semen, sperm motility, viability, and DNA integrity decreased (P<0.05) in aged vs. young bulls. In frozen-thawed vs. fresh semen, MDA level (nmol/ml) increased within young (48.4±2.6 vs. 2.3±0.2) and aged bulls (53.1±2.8 vs. 2.9±0.7), while motility and viability decreased (P<0.05) within the age groups. In conclusion, 1) lipid peroxidation (MDA) does not increase due to age in buffalo bull semen, and 2) freezing causes increase in lipid peroxidation irrespective of age and deteriorates semen quality of Nili-Ravi bulls. As diarrhoea has different levels of dehydration recognized by their specific signs can lead to abnormal level of serum electrolytes and trace elements (minerals). Serum concentration of electrolytes and minerals varies during gastroenteritis in dogs and its values change dramatically at different dehydration levels. Which could cause death in dogs. The present study was designed to check the effect of diarrhea on different electrolyte and mineral. Total 40 dogs was included in this study from different private pet clinics and Pet Centre, UVAS, Lahore suffering from clinical diarrhea and vomiting irrespective of cause, These dogs were divided into four groups A, B, C and Control, comprising of 10 dogs in each group. Group A, B and C will be categorized according to dehydration state i.e. 0 - 5%, 5 -8% and 8 - 10% respectively. These groups were made on basis of clinical signs and Packed Cell Volume, (PCV), whereas Group D was be kept as a control, comprising of normal and healthy dogs. These were further subdivided on the basis of age. 5 dogs were included having less than 1 year age and 5 dogs were included having more than 1 year age with the same dehydration level. PCV was checked to diagnose the level of dehydration. The PCV value, which comes in desire category, were further proceeded for serum collection to check the concentrations of serum electrolytes and minerals like Na, K, Cl, Cu, Zn and Fe through different methods like Na CHAPTER 6 SUMMARY Summary 46 and K were checked by flame photometer, Fe, Zn and Cu were checked by atomic absorption and Cl were determined by titration at WTO Laboratory and the Laboratory of Environmental Science, Department at University of Veterinary and Animal Sciences Lahore. Analysis of variance ANOVA (1 way factorial) technique were used with Random Complete Block Design (RCBD) to compare serum electrolytes and trace elements concentration in gastroenteritis with the concentration of healthy one. Considering the importance and utilization of dogs in our country and substantial losses occur due to deficiency of vital electrolytes and minerals at different dehydration levels due to diarrhea and vomiting irrespective of the cause, the present project were planned to give proper guidance to dog’s owners for their treatment and quick recovery by knowing about the decreasing serum concentration of essential electrolytes and vital minerals during different dehydration levels have different age groups. From the present study the value of sodium was observed significantly reduced (P<0.05) in all test groups (A, B, C) as compared to control unhydrated group of dogs when serum chemistry was done/ According to result of present study, the concentration of Na in healthy dog was 146.47 ± 3.06 (mmol/l) in infected dogs the concentration was Na= 144.40 ± 3.61 (mmol/l) while normal range of Na= 142-150mmol/l. Chloride value was observed significantly reduced (P<0.05) in all test groups (A, B, C) as compared to control unhydrated group of dogs when serum chemistry was done. This Chloride value was also significantly reduced (P<0.05) in group A as compared to the groups B and C, while chloride value in group B was significantly reduced (P<0.05) as compared to group C. Iron value was observed significantly reduced (P<0.05) in all test groups (A, B, C) as compared to control unhydrated group of dogs when serum chemistry was done. This value was also significantly reduced (P<0.05) in group C as compared to the groups A, but the iron value has no significant effect between group A and group B. Zinc value was observed significantly reduced (P<0.05) in all test groups (A, B, C) as compared to control unhydrated group of dogs when serum chemistry was done. Zinc value was also significantly reduced (P<0.05) in group C as compared to the groups A, but the iron value has no significant effect between group A and group B. Electrolytes such as sodium (Na), chloride (Cl), magnesium (Mg), potassium (K), Cupper (Cu), and iron (Fe) are involved in several physiological processes like conduction of electrical impulse through nervous system and muscle contraction and their imbalance could lead a lowering of animal performance. Coccidiosis cause losses through morbidity and hidden effects on feed intake, efficiency of nutrients utilization and also reduce growth rate in young animal, as a result, it leads to reduction in productivity and performance of the infected animal. The present study was designed to study the prevalence and chemotherapy of coccidiosis in camel. For this, fecal samples of 100 camels of both sex and of various aged were examined. For this purpose, 10-15gm fresh fecal sample was collected into a polythene zipper from the rectum of each camel in a container with ice packs and transported to Medicine lab, UVAS Lahore. The fecal sample was examined by using direct smear method and floatation technique and OPG were determined by McMaster technique. By using these techniques the prevalence of coccidiosis in the camel was found to be 13% i.e. 13 animals of variousage of both male and female had coccidiosis. For chemotherapy, 12 animals were used by dividing into 3 groups containing 4 animals in each group randomly.4 positive animals were given amprolium 50 mg/kg BW for 5 days daily PO. 4 positive animals were given toltrazuril20mg/kg BW PO once and neem seed at the dose rate of 100mg/kg BW to 4 positive animals once PO. The fecal sample were again collected from camels to which drug had used at 14 and 21 days and comparison of drug were done to determine the efficacy among these drugs by OPG counts , Toltrazuril was found to had more efficacy measured by OPG count reduction using ANOVA in SPSS.There is coccidiosis in the camel which caused weakness, off-feed and diarrhea in camels. There are both allopathic and herbals drugs used for the treatment but allopathic drugs were found to be had more efficacy. Among other bacterial etiology of bovine mastitis, the Staphylococcus aureus is overwhelming to control and is well-known to cause subclinical and contagious mastitis. Methicillin resistant Staphylococcus aureus is our prevailing field issue. In view of the economic importance of Staphylococcus aureus mastitis, the current project was designed to study the Clinico-epidemiology of multiple drug resistant Staphylococcus aureus in bovine mastitis. A total number of 900 milk samples (n=450 cattle, n=450 buffalo) were collected from Faisalabad district of Punjab, The collected samples were processed in laboratory of Microbiology and Medicine, Faculty of Veterinary Sciences, University of Veterinary and Animal Sciences, Lahore. Primarily, screening of subclinical mastitis was done by Surf Field Mastitis Test (SFMT). Milk samples were spread out primarily on blood agar to rule out possibility of loss of later growth. Colonies with round and golden color characteristic were put to sub-culturing on Mannitol salt agar as differential and selective medium. The morphological and microscopic clarification was done under microscope using Gram’s staining technique. Various biochemical tests including coagulase and catalase were applied. Prevalence of subclinical mastitis was found 55% (495/900), however significant differences were found among different tehsils of district Faisalabad. The prevalence of subclinical mastitis in cattle from district Faisalabad was found 54% (243/450) that upon comparison between different cities presented significant difference. While buffaloes presented 56% of subclinical mastitis. Comparisons of subclinical mastitis among different tehsils were Summary 86 found significant. Among quarter based prevalence of subclinical mastitis and prevalence of blocked quarters the number of blocked quarters were found 5.58% (201/3600) with highest percentage of blocked quarter noted in case of front right followed by rear right , front left , and rear left with 6.89, 6.56, 4.67, and 4.22%, respectively. However, the quarter based prevalence was found 32% (1088/3399) from bovine. The association of bovine subclinical mastitis with different risk factors presented significant association with few exceptions. Age, parity and body health status of animal were found non- significant with prevalence of mastitis. The breed and open rearing system presented significant relation with chances of mastitis. Chi-square test was used to statistically correlate the risk factors and prevalence of subclinical mastitis. P-value less than 0.05 was considered as significant. Molecular confirmation of S. aureus was done by using coag gene through PCR technique. The S. aureus which were isolated from milk samples were put to antibiotic (oxacillin) sensitivity test for estimation of prevalence of methicillin resistant S. aureus from the bovine mastitis. Molecular identification of mec-A gene in staphylococcus aureus was done through PCR. The PCR confirmed methicillin resistant S. aureus isolates from cattle (n=20) and buffalo (n=20) were tested for their in-vitro drug response. However, Ciprofloxcin, Moxifloxacine, Linezolid, and Trimethoprim + Sulphamethoxazole were found 100% effective against multiple drug resistant Staphylococcus aureus and Levofloxacin showed 90% efficacy in bovine. While Oxytetracycline, Tylosin, Gentamycin, Amikacin, Vancomycin, and Fusidic acid were also found sensitive moderately except cefoxitin which was responsible for 100% resistane in bovine. Gentamycin was found to much more effective in buffaloes rather than cattle. Summary 87 The study provided current status of Staphylococcus aureus infection with higher percentage in bovine mastitis. The prevalence of mecA gene revealed variation in methicillin resistant Staphylococcus aureus. Reproduction is important to get good production and profit in dairy sector. Reproduction in buffalo is compromised due to its small size ovaries, poor ovarian reserves and less pronounced estrus intensity, resulting as low fertility. Synchronization techniques including CIDR based protocols are well established in cows and getting popularity in buffaloes but with low results, comparatively. Therefore, some modifications are required based on physiology of estrus cycle in buffalo. This was hypothesized that additional injection of Dalmavital in CIDR base protocol will enhance the pregnancy rates and embryonic liability by minimizing the oxidative stress. Therefore, present study is conducted to evaluate the effect Dalmavital on estrus response, Estrus intensity, Pregnancy rates and embryonic losses in CIDR synchronized Nili-Ravi buffalo. For this, buffaloes were scanned ultrasonically for the reproductive tract evaluation. Reproductively sound buffaloes were selected and randomly allocated to one of the two treatment group. 86 buffaloes with normal reproductive tract were assigned in two groups 1; CIDR group (n = 43) and 2; CIDR+D (n = 43). AI was performed twice at 48 and 60 hours after CIDR removal. Estrus response (ER) did not differ significantly (P >0.05) in groups, CIDR and CIDR-D but estrus intensity (EI) was statistically significant (P<0.05) in treatment group. Pregnancy rates were also non-significant (P>0.05) in treatment and control group but improved comparatively in CIDR-D group (63% in CIDR-D group and 56% in CIDR group). Embryonic and fetal losses were also non-significant (P>0.05) between the control and treatment group. Results were also compared in cyclic and non-cyclic, Milking and dry, BCS and parity. Results were non-significant in milking and dry, BCS and Parity. Pregnancy rates were found different significantly (P<0.05) in cyclic and non-cyclic animals, when treatment Summary 27 is ignored. From the present study it can be concluded that Dalmavital may have good effect on estrus intensity in CIDR synchronized Nili-Ravi buffalo. Nili-Ravi buffalo is known as black gold of Pakistan. They produce about 2500 liters of milk with 6.5% butter fat. Despite of benefits, this breed is highly influenced with low reproductive activity that include prolonged pubertal period, poor exhibition of estrus, inadequate ovarian activity, long calving interval, high embryonic mortality and low fertility rate with artificial insemination. These factors reduce buffalo’s reproduction which leads to great economic losses. Therefore, there is dire need to address these problems and to orchestrate novel approaches to enhance the reproductive efficiency of buffalo. In cows, during last six to decades, researchers have considerably devised certain strategies to synchronize estrus with the help of prostaglandins, progestagens and estrogens. The advent of these hormones or synchronization protocols helped significantly in improving reproductive management. Moreover, the use of fixed time artificial insemination protocols resulted in acceptable fertility. However, these tools provide inconsistent results to manage reproduction in buffalo. 46. Comparative Efficacy Of Mycotoxin Binders And Effects Of Aflatoxin B1 On Health Status Of Lactating Beetal Goats

The most toxic is the aflatoxin B1 (AFB1) and is considered as Hepatocarcinogenic. In present study aflatoxin B1 contamination status of different concentrate feeds (cotton seed cake, wanda, wheat bran and homemade concentrate mixture) of dairy goats was investigated in district Lahore of Pakistan. Twenty goat farms were randomly selected and 40 feed samples (two from each farm) were collected. The samples were analysed for the estimation of AFB1 using reverse phase High Performance Liquid Chromatography (HPLC) in Quality Operations Laboratory, UVAS, Lahore. AFB1 was detected in 33 feed samples out of 40 thus with a percent contamination rate of 83%. The quantity of aflatoxin B1 ranged from 0-225.736 ppb (μg/kg). The maximum level of AFB1 was detected in cotton seed cake (mean level 137.059 ± 22.293 ppb) and minimum level was detected in wheat bran (mean level 5.676 ± 1.047 ppb). Amongst the positive concentrate feed samples, 28 samples (85% of the positive) were having the concentration of AFB1 higher than the permissible level recommended by European Communities which is 5 ppb for concentrates. Thus the milk from goats consuming AFB1 contaminated concentrate feed can be a potential hazard for public health. This part of the study fulfills the first objective of the research project, which is to determine the aflatoxin B1 contamination status of concentrate feeds of dairy goats in District Lahore. Beetal is the best dairy goats’ breed of Pakistan. In second part of the study, lactating beetal goats were used. In this experiment, effects of dietary AFB1 on health status of lactating beetal goats and its transfer from feed into milk as metabolite aflatoxin M1 (AFM1) were investigated. Thirty two lactating beetal goats of 3-4yr old, 6-8 wks. lactation period and weighing 40.91 ± 0.285 Kg, were selected from Small Ruminant Farm, Pattoki, University of veterinary and Animal 97 Sciences Lahore, and were randomly divided into four groups A, B, C and D, each having 8 animals. Group A was kept as negative control, while groups B, C and D were fed with aflatoxin B1 through naturally contaminated cotton seed cake. The cotton seed cake was having high level of AFB1 which was 150 μg/kg. After 7 days of adaptation period, each animal of groups B, C and D was individually fed with 30μg, 40μg and 50μg of aflatoxin B1 per day respectively through naturally contaminated cotton seed cake for a 10 days period., thus each animal of groups B, C, and D was fed with 200 g, 266.66 g, and 333.33 g of cotton seed cake per day respectively, to provide the required amount of AFB1 to each experimental animal. Milk samples were collected 24hr before the first AFB1 feeding and on 3rd, 7th and 10th day post aflatoxin B1 feeding. The samples were tested for AFM1 through HPLC, somatic cell count (SCC) and total viable count (TVC). Blood samples were analyzed for aspartate aminotransferase (AST), alanine aminotransferase (ALT) and alkaline phosphatase (ALP) levels before and after aflatoxin B1 administration. AFM1 was detected in all milk samples of the Groups B, C and D, with concentration beyond permissible level which is 0.05ppb (European communities).The amount of AFM1 excreted in milk was positively correlated with the amount of aflatoxin B1 ingested. The AFB1 was excreted in the milk as AFM1 at 1.35-1.59%. The mean SCC of groups A, B, C and D was 1.44×106 ± 1.75×105, 4.13×106 ± 4.09×105, 3.63×106 ± 4.60×105, 1.38×106 ± 1.56×105 cells/ml respectively. TVC obtained was 2.4×104 ± 1.9×103, 4.3×104 ± 1.6×103, 4.8×104 ± 3.3×103, 3.6×104 ± 3.3×103 cfu/ml of milk for group A, B, C and D respectively. The mean ALP levels were 62 ± 7, 223 ± 22, 147 ± 14, 175 ± 16 U/L for group A, B, C and D respectively. Mean levels of AST of group A, B, C and D were 79 ± 2.214, 110 ± 5.386, 104 ± 2.015, 126 ± 9.456 respectively. Mean levels of ALT of group A, B, C and D were 14 ± 0.326, 20 ± 1.118, 21 ± 1.106, 28 ± 1.250 U/L respectively. The ingestion of AFB1 even at low dose of 30μg in goats resulted in excretion 98 of AFM1 in milk beyond the permissible level. SCC, TVC, AST and ALT levels increased with ingestion of dietary AFB1. This part of the study fulfills the 2nd objective of the research project which is to investigate the effects of dietary aflatoxin B1 on health status of lactating beetal goats and its transfer from feed into milk as AFB1. In the third part of the study, the efficacy of two different mycotoxin binders Toxfin® and Elitox® was determined, in terms of reduction in milk AFM1 excretion, improvement in milk quality and liver health. Groups A and B were kept negative and positive controls respectively. Experimental animals, cotton seed cake and number of groups remained the same as used in part 2 of the experiment. After initial 10 days of the experiment, each animal of groups B, C and D was daily fed with 266.66 g of cotton seed cake to provide 40 μg of AFB1 per day to each animal, for next 7 days. In the meantime, group C was fed with mycotoxin binder Toxfin® (Kemin industries, Inc. USA), at a dose rate of 3 g per Kg of cotton seed cake, while group D was fed with mycotoxin binder Elitox® (Impextraco Belgium ) at a dose rate of 1 g per Kg of cotton seed cake. Toxfin® and Elitox® both significantly decreased the excretion of AFM1 in goats’ milk, detected through high performance liquid chromatography (HPLC). Toxfin® and Elitox® reduced the excretion of AFM1 in milk by 56 % and 48 % respectively, thus the detoxifying ability of Toxfin® in feed was significantly higher than Elitox®. The SCC and TVC remained statistically unchanged after the administration of mycotoxin binders. The enzyme activities of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were not significantly affected by the mycotoxin binders. The alkaline phosphatase (ALP) level significantly increased in animals receiving either Toxfin® or Elitox®. The mycotoxin binders used in this study were not able to significantly improve milk quality and liver’s health in AFB1 treated goats, during the experimental period. This part of the study fulfills the 3rd objective of the research project, which was to determine the comparative efficacy of two different mycotoxin binders in lactating goats fed with mold contaminated diet. These are acting as vectors for numerous animal and human diseases like Bovine ephemeral fever, Filariasis, Dengue fever, Malaria, Yellow fever, Japanese encephalitis. The vector control strategies are the potent tool to minimize the risk of transmission of these infectious agents. In Pakistan, these vector control strategies mainly rely on chemical insecticides. The irrational use of these chemicals is the leading cause of emerging insecticide resistance in these mosquitoes. It has been also revealed that these conventional insecticides are not eco-friendly, has drastic effects on non-targeted biota and also having the potential of cross resistance among various compounds. To mitigate this issue, special attention is required to use natural products and plant origin as an alternate to these chemical insecticides. As these herbal plants are not eco-friendly but also having least drastic effects on non-targeted population. Cympbogon citratus (lemon grass) could have good adulticidal and larvicidal activity against mosquitoes of genus culex. The mosquitoes of genus Culex were identified on the basis of distinguishing morphological features as described by standard published morphological key. After identification the adults of mosquitoes of genus Culex were further reared in the insectary of Department of Parasitology, UVAS, Lahore to take F1 and larval stages of mosquitoes of Culex spp. Briefly, female mosquitoes were feed on periodic blood for oviposition. These emerging L3 & L4 larvae and 2-3 days old adults were further used in protocol. The fresh green leaves of C. citratus were used in current study. After collection and identification the aqueous and methanol extract were taken by Soxhlet apparatus (Govindarajan et al., 2011). Afterwards different concentrations of these extracts were prepared to calculate the diagnostic dose of these extracts. Then the larvicidal and Summary 46 adulticidal efficacy of these extracts were evaluated by using CDC bottle bioassays as described by (WHO2005) The LC50 and LC90 were calculated by using probit analysis. Staistical package of social sciences (SPSS), version 13.00 were used for this purpose. Duncans’s Post HOC Test were applied to compare the main mortalities among the group of different concentrations of plant extracts and control group. 48. Prevalence, Molecular Diagnosis And Chemotherapy Of Degnala Disease In Large Ruminants Of Punjab.

Degnala disease reduces the production of these animals directly. Along with other side issues related to Degnala disease, this study was done to diagnose the actual cause of Degnala disease by applying different latest scientific techniques. Prevalence along with risk factors was calculated in the rice growing areas of Punjab, Pakistan. Fungal isolation (n=40) was performed from the rice straw feedings of the Degnala disease affected animals through the technique of spot culture on SDA. Then these fungal isolates were identified through comparing their microscopic and macroscopic characters. Then toxigenic potential was checked for all these isolates through the application of TLC and HPLC. After that, from those isolates which were positive for mycotoxin production potential, most cytotoxic isolate was checked with the application of MTT assay. Then the most cytotoxic isolate was inoculated on non-contaminated rice straw and fed to the experimental animals to see a similarity of natural cases of Degnala disease. Finally treatment was conducted to see a proper combination of various drugs against this disease. Toxigenic potential of different candidate fungi, isolated from rice straw feeding of Degnala disease affected bovines was analysed along with Species, age, gender and season wise prevalence. Out of 1536, 104 (6.77%) cases showed positive signs for this disease with a significant association (p<0.05) between rice straw feeding in buffaloes, winter season and bovines having an age of more than one year. Complete Blood Count showed marked increase in erythrocyte sedimentation rate and all white blood cells numbers, except lymphocytes in positive cases. There was a significant increase (p<0.05) in Alanine amino transferase, Aspartate amino transferase and Alkaline phosphatase noticed in Liver Function Test. At the same time, increased value of Creatinine was noticed in Renal Function Test. For isolation and screening of toxigenic fungi, rice straw samples (n=40) being fed to the positive cases were processed further, out of which there were 85 fungal isolates mainly of Aspergillus (57), Penicillium (10), Fusarium (04), Zygomycetes (03), Curvularia (01) and unidentified (10). All isolated fungi were subjected for mycotoxin production and only 11 showed mycotoxin producing capability (including Aspergillus, Penicillium and Fusarium isolates) analysed by Thin Layer Chromatography and quantified through High Performance Liquid Chromatography. It is concluded that all the fungi, contaminating rice straw feeding of Degnala affected animals are not toxigenic. This work will help in establishing major mycotoxin producing fungi leading to the probable cause of Degnala disease in bovine. With the help of MTT assay on vero cell line, most cytotoxic fungus was identified. After an incubation with vero cells, OD values of all the candidate fungi were compared through one way ANOVA. Results of this analysis showed that Fusarium was at the highest ranking and then was the A. flavus with a significant value of 0.006 and 0.039. Finally it was concluded through these systematic steps of converging the diagnosis that, out of all the 85 suspected fungi, Fusarium (isolate number S 8.1) was the most cytotoxic isolate obtained from the rice straw feedings of Degnala affected animals in our study. For molecular diagnosis of the most cytotoxic isolate of Fusarium, PCR was conducted and the results showed that ultimately the final PCR product was successfully amplified against the mentioned primer of ITS conserved region for Fusarium genera and the DNA product was with a length of 570 base pairs. Experimental feeding trials were conducted by inoculating Fusarium (the most cytotoxic isolate) and A. flavus (second most cytotoxic one after Fusarium) separately and in combination compared with the negative control group, all groups were of eight animals each. It was concluded that alone Fusarium was able to produce Degnala disease, while its combination with A. flavus was more lethal. Ultimately the treatment trials proceeded with penta-sulphate, oxytetracycline and antiseptic topical application as therapeutic treatment were shown to be very effective against Degnala cases. While in all the affected animals feeding of affected rice straw was ceased. Only withdrawal of affected rice straw from the feedings of Degnala affected animals was not effective unless proper treatment as mentioned here was not conducted. analysed The expected results of the study shall be helpful to make exact diagnosis and treatment of infected buffaloes and cattle that is further helpful for timely prophylaxis and control of the Degnala disease in the rice growing areas of Pakistan and South Asia.
