1. Effects Of Aflatoxin In Poultry

Material type: book Book; Format: print ; Literary form: not fiction Publisher: 1988Dissertation note: A total number of 300 samples of finished commercial poultry feeds, obtained from poultry feed mills (75 samples), wholesale poultry feed dealers (70 samples) and poultry farms (155 samples) were examined for aflatoxin contamination.It was found that 126 (42%) samples, including 18 (24%) from mills, 19 (27.1%) from dealers and 89 (57.42%) from farms were samples was carried out both by the aqueous acetone method, and the chloroform extraction method. The extracts were qualitatively examined by quick screening method, minicolumn chromatography method and thin layer chromatography method. All of three methods gave comparable results. The quantity of aflatoxin in contaminated samples ranged from 20 microgram/kg to 2000 microgram/kg. The levels of aflatoxin in majorityofthe contaminated samples (90.5%) ranged from 20 microgram/kg to 150 microgram/kg feed and only 9.5% of samples contained higher levels of aflatoxin. Most of the samples containing higher levels of aflatoxin came from commercial poultry farms. These farms had complaints of high mortalities and poor performance in broilers and low production and low mortalities in breeder flocks. The experimental production of aflatoxin was carried out on long grain rice using a toxigenic strain of Aspergillus parasiticus (FRR-2752). The rice cultures were incubated at 28 degree centigrade in an atmosphere of high humidity. In the present study a maximum yield of 803 microgram aflatoxing rice was obtained. The determination of LD50 of aflatoxin was carried out in 210 one day old Hy-Bred broiler chicks with an average weight of 38 gram each. The chicks were divided into 21 groups labeled from 1 to 21 with 10 birds in each. A single dose of aflatoxin, ranging from 32.82 mg/kg body weight to 0.33 mg/kg body weight, was inoculated into the crops of chicks in groups 1 to 19, the group 20 acted as solvent control and the group 21 as aflatoxin free control. The birds were observed for 7 days post inoculation. The physical state and mortalities were recorded. The birds which had received higher levels of aflatoxin died within few hours of inoculation showing symptoms and lesions of per acute aflatoxicosis. The LD50 was calculated by Abbot's probit method and was found to be 9.278 mg/kg body weight. The pathological effect of a single dose of aflatoxin B1 on the immunocompetent organs was studied in a group of broiler chickens. Day-old 60 Hy-Bred broiler chicks were raised on aflatoxin free feed for 3 weeks and then divided into six groups labeled 1 to 6 with 10 birds in each. A single dose of pure aflatoxin B1 at dose rates of 8, 16, 26, 50 and 100 microgram/birds was given to the birds in groups of 1 to 5 respectively, the sixth group acted as toxin free control. The birds were maintained on aflatoxin free feed and water ad lib and observed for 3 weeks post inoculation. The bursa of Fabricius, thymus and spleen of each bird was removed and histologically examined, No appreciable histological changes were seen in the organs of birds which had received 8, 16 and 26 microgram AFB1/bird while reductions in size accompanied with other degenerative changes were seen in the thymus glands, bursa and spleens of birds, which had been injected 50 and 100 microgram AFB1/birds respectively. The normal tissues of these organs were replaced by the inflammatory and fibrous tissues. No changes, gross pathological or histological, were seen in the thymus glands, bursa and spleen of control group birds. The Immunomodulatory effects of a single dose of 100 microgram aflatoxin B1/bird on the development of immunity against Newcastle disease virus vaccine was studied in broiler and layer chickens. A group of 120 one day-old Hy-Bred broiler chicks were divided ito two groups with 60 birds in each and raised for 3 weeks on an aflatoxin free feed.Three randomly selected chicks were bled on the first 7th, 14th and 21st days of age for the determination of maternal haemagglutinin inhibiting (HI) antibody titers. At 3 weeks of age the broilers of each batch were further subdivided into groups labeled as T, T1,T2, K1 and K2 with 10 birds in each. A batch of layer chicks was raised for 8 weeks on aflatoxin free feed and then divided into groups T, T1, T2, K1 and K2 with 10 birds in each. Three randomly selected chicks were bled on day one and thereafter weekly for determination of maternal H1 antibody titers till the 7th week of age. The birds in groups T received vaccine and aflatoxin simultaneously, the birds in groups T1 received vaccine 72 hours before toxin and the birds in groups T2 received toxin followed 72 hours later by vaccine. The birds in group K1 acted as toxin free vaccinated control and the bird in groups k2 acted as toxin free unvaccinated control. The birds were maintained on aflatoxin free feed for further 4 weeks. Three randomly selected birds of each group were bled weekly for the determination of serum H1 antibody titers. At the end of 3rd week post inoculation one batch of broilers and the layer groups were challenged with a virulent strain of Newcastle Disease Virus while the other batch of broilers was given a booster dose of vaccine. All of the birds in unvaccinated aflatoxin free control groups (k2) died within 72 hours of challenge, while the rest o the birds survived. The survivors were bled and sacrificed one week after challenge/booster vaccination. The Sera of birds were examined for H1 antibody titers. The results showed that the administration of aflatoxin alongwith, immediately before or after vaccination depressed the development of H1 antibodies significantly. Immunomodulation caused by continued feeding of aflatoxin on the development of immunity against Pasteurella multocida vaccine were studied in layer and broiler chickens. Seventy two one-day old hyline layer chicks were raised for 6 weeks on aflatoxin feed and then divided into groups T, T1, T2, T3, K1 and K2 with 12 birds in the Seventy two Hy-Bred broiler chicks, one-day old were divided into 6 groups T, T1, T2, T3, K1 and K2 with 12 birds in each. A toxic feed containing 2.1 microgram of aflatoxin/gram was prepared. The Birds in groups T were given toxic feed for 42 days (21 days before and 21 days after vaccination). The birds in groups T1 were fed toxic meal for 21 days before vaccination and those in groups T2 were fed toxic meal for 21 days after vaccination. The birds in groups T3 received a single dose of 9.278 mg Aflatoxin/kg body weight on the day of vaccination. The birds in the groups K2 as toxin free vaccine free control. All of the birds, except those in the groups K2, were vaccinated with Pasteurella multocida vaccine at the end of 3rd week of age in the broilers and 9th week of age in the layers and challeneged with a virulent Pasteurella multocida organisms 3 weeks post vaccination. After challenge the birds were shifted to aflatoxin free feed till the termination of the experiment. In layers one bird each of group T and T3 died. In unvaccinated toxin free control group of layers as well as broilers (K2) 11 out of 12 birds died. Six broilers of group T and 5 broilers of group T3 died after challenge. The experiment was terminated on the 7th post challenge. The survivors were bled and sacrificed. Serum was collected from 4 randomly selected birds on the day of vaccination and thereafter weekly till the termination of the experiment. The antibody titer were determined by IHA and ELISA tests. Continued feeding of aflatoxin depressed the development of humoral immunity against Pasteurella multocida vaccine, the depression being more pronounced in broilers. The pathological effects of aflatoxin were studied in 3 weeks old broiler chicks. A batch of 36 one day-old broiler chicks was raised on aflatoxin free feed for 3 weeks. On 21 days of age the chicks were divided into 3 groups T, T1 and C with 12 birds in each. A single dose of 9.278 mg/kg body weight was injected into the crop of birds in group T. The birds in group T1 were maintained on a feed containing 9.278 mg aflatoxin/bird (2.1 ug/g feed) for 4 weeks. The third group (k) acted as control. The experiment was terminated on the 49th day of age by sacrificing the survivors. The liver of birds in group T1 were enlarged, and the heart were atrophied. Regenerative changes, some bile duct hyperplasia and fatty changes were seen in liver of birds in group T. in Birds of group T2 the liver was enlarged and showed nodular hyperplasia. Histologically the liver tissue showed acute necrosis, pericellular fibrosis, nuclear dissolution, bile duct proliferation and lymphoid hyperplasia. Degenerative changes were also seen in the heart and kidneys and other organs. No gross or histological changes were present in the organs of the control group (K) birds. The effects of aflatoxin on the live weight, dressed weight, the weight of liver, heart and gizzard, some seral enzymes and bilirubin were studied in a group of 165 broiler chicks. Day old Hy-Bred broiler chicks were raised on aflatoxin free feed for 3 weeks and then divided into 3 groups T, T1 and K with 55 birds in each. The birds in group T received a single dose of 9.278 aflatoxin/kg body weight while the birds in the group T1 received 9.278 mg aflatoxin/bird which was mixed in feed and offered ad. Lib. To these birds over the next 4 weeks. The birds in group K acted as the toxin free control. The experiment was terminated on the 28th day post inoculation by sacrificing the survivors. Five chicks from each group were bled through cardiac puncture and sacrificed, daily from day one (21st day age) to the 7th day post inoculation (28th day age) and thereafter weekly till the end of the experiment. Serum of these birds was examined for SGOT, SGPT, LDH, SAP and Bilirubin. Continued feeding of aflatoxin or administration of a single dose of aflatoxin significantly depressed the live weight, dressed weight and weight of heart, while it significantly increased the weight of liver and gizzard. Administration of a single dose of aflatoxin produced dramatic increase in the volume activity of SGOT, SGPT, LDH,SAP bilirubin within 24 hours of toxin administration, the values remained higher during the first week and thereafter slowly came down. In birds fed on contaminated meals the enzymic activity and bilirubin gradually increased during the first week and remained high till the termination of the experiment. In the birds of control group the activity of these parameters remained on baseline levels. No Carcinogenicity was seen in any of the internal organs of layer chickens which had been raised for 1 year on feed containing 2 ug aflatoxing. No aflatoxin or aflatoxin residue could be detected in the liver, kidneys and breast muscles of broilers, which had been fed contaminated meals for various lengths of time, and were shifted to aflatoxin free feed 7 days before slaughter. Aflatoxin was recovered from the liver and kidneys of layers which were feeding a contaminated meals at the time of sacrifice, the rate of recovery being 1 microgram/100 grams liver tissue and less than 1 microgram/100 gram kidney tissue. The serious food shortage and increasing competition between human beings and poultry regarding the consumption of cereal grains demanded that cereals should be now spared for human consumption and exploration of non conventional feed sources should be carried out in order to keep the business of poultry industry running well. Rice polishing is a by-product of rice milling so it is abundantly available in Pakistan because rice is the third largest crop produced in the country. Rice polishing compares well with the cereal grains in chemical composition. So it contains more fat and protein as compared to maize but high in fiber contents. Due to high fat contents rancidity of Rice polishing can pose some problems if it is fed after prolonged storage, specially in hot weather. Extrusion cooking is a new technique which stabilizes the high fat diets and make them fit for use in the animal rations. For this reason full fat rice polishing was subjected to Extrusion Cooking which was done at National Feed Mill. Present Experiment was conducted to replace maize with extruded rice polishing and to see the effects on the performance of broiler chicks. Six starter and six finisher rations were prepared and designated as A, B, C, D, E and F . All rations were isocaloric and isonitrogenous. Starter ration and finisher ration A. were kept as a control containing 35 % Maize and 0% extruded Rice polishing, which was replaced by extruded rice polishing in ration B, C, D, E and F. on protein equivalent basis at 15, 20, 25, 30 and 35% level In this way a complete 100% replacement of maize with extruded Rice polishing was made One hundred and eighty day old chicks were randomly divided into eighteen experimental units of 10 chicks each. Each experimental unit was placed individually. The starter rations were fed up to 28th day of age while the Finisher rations from 29th to 42nd day of age. Results of the present study indicated that the weight gain up to 25% extruded rice polishing level was promisingly good. Maximum weight gain was recorded in Ration B (15% extruded rice polishing). Second best results were noted in the control ration but the performance of the chicks fed on 20% extruded rice polishing was almost equal to that of control ration. It was noted that the weight gain started to decrease gradually as the level was raised up to 35% of the total with each increasing increment. Best feed utilization was observed in the chicks fed on ration B, whereas poor utilization of feed was noted in higher levels of extruded rice polishing in the diet having more feed consumption compared to weight gain. Better performance up to 25% level indicated that Extrusion improved the nutritional quality of rice polishing by stabilizing it and inactivating Lipases. The extrusion also made starch more available for the birds and reduced the anti-nutritional elements of rice polishing. This is evident from the fact that the level of rice polishing effected the health of the chicks. It was therefore concluded from the present study that extrusion is an excellent technique for converting low quality feed stuffs into feeds with higher nutritional quality. Specially extrusion of low priced feed ingredients such as rice polishing and it replacement with a costly feed ingredient such as wheat or maize holds good economic prospects for feed millers. According to an assessment, about 250 million cattle are endangered by this disease and millions of high milk yielding cattle are at risk of exposure to disease. It acts as a bigger restraint on livestock improvement and production in many developing countries (Nagore et al. 2004). Theileria annulata is the main specie that causes high morbidity and mortality. It causes heavy economic and production losses in cattle in tropical and sub-tropical regions. The recorded mortality rates in cattle reached to 70% (Moor house et al. 2001). Theileria species are intracellular obligate hemoprotozoan parasites. All Theileria species are dangerous and cause disease but two of them are important for livestock. Theileria parva and T. annulata produces diseases named as East Coast fever and tropical theileriosis in cattle respectively. Genus Theileria has many other species like T. buffeli, T. taurotragi, T. velifera, T. sergenti and T. mutans. These species cause infections in wild and domesticated ruminants. Theileria species present in large and small animals show signs like fever, anorexia, swelling of the superficial lymph nodes, dyspnoea, lethargy, progressive anemia, constipation, diarrhea, lacrimation and nervous symptoms (Saeed et al. 2010; Irvin and Mawmachi 1983). A pronounced rise in body temperature, reaching 40-41.5 °C is pursued by lacrimation, depression, swelling of the superficial lymph node and nasal discharge. The characteristic sign of tropical theileriosis is anemia and finally haemoglobinuria occur with heavy weight losses. The clinical course of the disease alter from per acute to acute or sub-acute to chronic (Oliveira- Sequeira et al. 2005). The disease is lymphoproliferative in its early phases resulting enlargement of lymph nodes, later on enters lymph destructive phase which is associated with a pronounced Introduction 2 leukopenia. In the piroplasms phase in erythrocytes, the parasite becomes infective for the tick (El-Deeb and Younis 2009). Trans placental Bovine Tropical Theileriosis causing a deadly disease in a 3 day old neonate cross bred calf and cerebral form of the disease (turning sickness) in a cow were incriminated to T.annulata infection. It mainly depends upon the harmful effects of the T. annulata on lymphoid tissues and susceptibility of the host (Sudan et al. 2012). Theileriosis is prevalent in various regions of the world including Pakistan. It is transmitted by Hyalomma species ticks. These ticks spread T. annulata which causes tropical theileriosis (Durrani et al. 2009). The developmental stages of Theileria inside the Hyalomma ticks varies in different shapes and forms (Hamed et al. 2011). Therefore to increase the milk and meat production of cattle we can prevent the spread of the disease by controlling ticks (Hekmatimoghaddam et al. 2012). The sufficient amount of Hyalomma ticks are found in warm, commonly hard marshland and in central and Southern Europe, south west Asia and Southern Africa having very long dry season. A toxin is produced in the adult ticks. This toxin produce clinical signs of mucus membrane hyperemic and moist profuse eczema (Adam et al. 2000). The sporozoites of Theileria enter into cattle host during tick feeding and they immediately infect mononuclear leukocytes, these sporozoites develop into macroschizonts and induce proliferation of the host cells. Macroschizonts constantly mature into microschizonts and finally into merozoites, which are discharged from leukocytes. These merozoites attack erythrocytes and mature into piroplasms, become available to ticks. Infective sporozoites, injected during tick feeding, rapidly enter target cells, escape from the surrounding host-cell membrane and differentiate to schizonts that interact with different host-cell components (Dobbelaereand Rottenberg 2003). This interaction includes host cell signaling pathways that Introduction 3 regulate proliferation and cell survival (Chaussepied and Langsley 2011) and thus cause blastogenesis and clonal expansion of predominantly T and B cells (Fawcett et al. 1982; Baldwin et al. 1988; Spooner et al. 1989). Merozoites released from these schizonts subsequently infect red blood cells and become trophozoites. Lymphocytic stage of Theileria (schizonts) is the cause of many of the severe disease manifestations like lymphadenopathy, pyrexia, thrombocytopenia, and panleukopenia (Homer et al. 2000). Marked anemia, anisocytosis, pikilocytosis, and leucopenia were commonly observed in bovine theileriosis (Ceci et al. 1997). Cattle may survive the disease, but recovery and convalescence may be protracted and incomplete, this leads to permanent debilitation, loss of productivity and prolonged carrier state. (Shahnawaz et al. 2011). Cattle with subclinical infection in endemic regions become carrier of piroplasms and act as a source of infection for the vectors (Brown 1997; Brown 1990; Uilenberg 1995). The diagnosis of theileriosis in acute cases is majorly done on clinical signs and Giemsa stained blood smears of cattle but the detection of agent is not reliable and is almost impossible in carrier stage. Advances in molecular biological techniques have resulted in the improved detection, identification, and genetic characterization of many hemoparasites. Species specific polymerase chain reaction (PCR) has been developed for the detection and identification of various Theileria species and has been shown to have higher sensitivity and specificity compared with serological assays and examination of Giemsa-stained blood smears (Bhoora et al. 2009). Primers were derived from the gene encoding the 30-kDa major merozoite surface antigen for T. annulata (Aktas et al. 2006). Most of the previous studies on haematological parameters in T. annulata infection were carried out on experimentally infected cattle (Sandhu et al. 1998; Singh et al. 2001). The present investigation was conducted to study haematological parameters in cattle naturally infected with Introduction 4 T. annulata. Hematology has been broadly used in attempts to give information about disease condition, performance problems and health in cattle (Rezaei and Naghadeh. 2006). Hematological and sero-biochemical alterations are the indicators of severity of disease and are considered to be good tools for the diagnosis, prognosis for effective therapy (Col and Uslu 2007; Nazifi et al. 2010b). Lahore is one of the larger district in the Punjab province of Pakistan. Different cattle breeds are reared by the people of the area for meat and milk production. The exact current situation about the prevalence and pathogenesis of Theileriosis in the selected area is unknown. The present study was conducted to screen cattle by finding schizonts or piroplasms in Giemsa stained thin blood smears at slaughter house of district Lahore (Aktas et al. 2006) and later to confirm through Polymerase Chain Reaction (PCR) (Chaisi et al. 2013) in order to implement efforts and regulation to eradicate the spread of disease in the selected area. Data generated from this study provided the latest status of Theileriosis, sex wise prevalence and its pathogenesis in cattle population of Lahore. The study has also provided the necessary information to formulate strategies for control of disease in the area. An investigation was also undertaken to ascertain the changes in haematology as a result of Theileria annulata infection. 5. Toxicopathological Alterations And Tissue Residue Of Colistinsulphate (Polymixin E) In Broilers

It is contributing 6.1% and 10.8% share in total contribution of agriculture and livestock the national GDP, respectively. Total poultry meat production in the last year was 987 thousand tones (Economic survey of Pakistan 2013-2014). Because of high density and lack of biosecurity measures, outbreaks of infectious disease are more common and often occurs. Different antibiotics are used rational and irrational to treat and control these outbreaks. Colistin is among the most widely used antibiotic in intensive poultry farming, it is used to combat gram negative bacterial infections particularly for collibacillosis and gastrointestinal tract infections (Hussain and Khalil 2013). Colistin belongs to the polymixins group of antibiotics. It is also known as Polymixin E. Polymixins are structurally related substances consisting of a cyclic peptide with a hydrophobic tail, mainly produced from gram positive nitrogen fixing bacteria Bacillus polymyxa. Polymixins are classified into six subtypes namely polymixin A to F and among these polymixin B and E are commonly used for the treatment of gram negative infections in animal and human beings. Colistin is mixture of two subtype’s colistin A (polymixin E1) and colistin B (polymixin E2). Commercially, it is available in two forms colistin sulfate and colistimethate sodium but colistin sulfate is more stable cationic water soluble salt (Landman et al. 2000) and is easily available in markets. colistin sulphate has lipophilic and lipophobic(hydrophilic) properties, that makes it easier to interact on bacterial cell membrane by displacing counter ions in lipopolysaccharides, influencing Mg efflux that’s why it is effective against gram negative bacteria (Poole and Sheffield 2013). Polymixins also presents antipyretic activity by binding endotoxins, especially lipid A, (the active compound of lipopolysaccharide). Owing to this property it is extensively used to treat gram negative infections in livestock and poultry industries (Hanasawa et al. 1990). The use of colistin was restricted in the past due to its toxic properties mainly in nephrotoxicity. There are reports available in the literature indicating that the parental use of colistin leads to the alterations of biochemical parameters in chicken i.e. increase in glutamic oxaloacetic transaminase, catalase activity, creatinine while, adversely affect total plasma proteins (Ibrahin et al. 2011). In ostriches it leads to the development of neurotoxic signs along with histologically odema formation in pericardium, intestinal serosa and heart was observed (Landman et al. 2000). Toxic effects of colistin were also reported in humans including nephrotoxicity, increased serum creatinine during the treatment (Falagas et al. 2005). As bioavailability for oral route is minimal and it is used parentally to treat systemic infections. Parenteral route may cause toxopathological effects in birds. Other effects includes change in aspartate aminotransferase and alanine aminotransferase which indicate liver damage. Red blood cells, white blood cells, packed cell volume and hemoglobin estimation were also decreased (Saleemi et al. 2013). The incidence of nephrotoxicity is severe but less common in oral administration as polymixins absorption through gastrointestinal tract is very slow and limited, and cannot be detected in plasma concentrations at ordinary doses (Falagas and Kasiakou 2006). Instead of all these toxic effects, now a days it is frequently used for the treatment against gram negative bacterial infections (Vaara 2010). The emergence of bacterial resistance has become an important public health hazard throughout the world). Due to irrational use of antimicrobial drugs against different diseases (Naqvi et al.2013. Colistin is one of the most widely used antibiotic in poultry industry against diseases like collibacillosis, salmonellosis and clostridial infections through drinking water, feed additives and parenteral route in developing countries like Pakistan (Tanweer et al.2013). Parenteral use of colistin sulphate may leads to the deposition of residues in different tissues. The presence of its residues in edible parts may lead to the toxicity as well as development of antibacterial resistance in human. Keeping in view the potential toxic effects and other hazards relating to the extensive use of colistin in poultry birds, present study has been designed to investigate dose dependent patho-morphological alterations and tissue residue in broiler chickens. Fleas are one of the most important ectoparasites with more than 2,000 species worldwide affecting mammals, birds, and reptiles (Hsu, 2003). In some locations, fleas represent over 50% of all the dermatological cases presented to small animal clinics. Most are limited to hosts with nests as this can provide conditions for the completion of their life cycle (Linardi & de Avelar, 2014). While fleas on pets are generally considered a nuisance that may cause some dermatologic problems, they are also responsible for the transmission of several important diseases in humans and animals (Dryden & Rust, 1994). They have been involved in transmission of cat scratch disease (Bartonella henselae) (Chomel et al., 2006; Comer et al., 2001), Rickettsia typhi (Murine thyphus), Rickettsia felis (Finkelstein et al., 2002; Rolain et al., 2005), and also serve as the intermediate host for the tapeworm Dipylidium caninum (Rust & Dryden, 1997) and several trypanosomatids (Coutinho & Linardi, 2007). The term ‘‘cat flea,’’ which is the approved common name for Ctenocephalides felis felis (C. f felis), can occasionally cause confusion. When it appears in print, it refers to the specific flea genus and species and not to fleas recovered from cats. There are four recognized subspecies of C. felis throughout the world: Ctenocephalides felis damarensis and C. felis strongylus occur primarily in East Africa, C felis orientis occurs in India and Australia, and the widespread C. f felis occurs in all continents except Antarctica and is the only subspecies that occurs in North America (Rust & Dryden, 1997). The cat flea, C. felis, is a clinically important parasite of domestic pets, being responsible for the production of allergic dermatitis, serving as the vector of Introduction 2 various bacterial pathogens, and being the intermediate host for filarid and cestode parasites. Flea allergy dermatitis is the most common dermatologic disease of dogs and a major cause of feline miliary dermatitis (Dryden & Rust, 1994; Rust & Dryden, 1997). Clinical features vary from asymptomatic to severe hypersensitivity reactions with restlessness, alopecia from scratching and biting resulting in a pruritic papular dermatitis. Vacuuming of carpets, furniture cushions, rugs, or other substrata, with a vacuum machine containing a ‘‘beater bar,’’ will remove many of the flea eggs and larvae. In addition, cocooned pupae at the upper levels of the carpet can also be affected. The vibration also stimulates adult fleas to emerge from their cocoons so that they can be collected in the vacuum machine. Therefore frequent vacuuming, during a flea infestation, can reduce the overall flea burden in the home. It should be ensured that vacuum bags are disposed of properly, to prevent recolonization of the home with flea stages previously removed by vacuuming. Because outdoor development of immature flea life stages is limited to shaded areas, altering outdoor environments to eliminate such habitats can effectively reduce flea populations. Because urban wildlife, such as opossums, raccoons, and foxes, are good hosts for cat fleas, pet owners should avoid encouraging visitations by wildlife, which will affect flea and tick control (see later discussion). Treatment of indoor and outdoor environments with insecticides requires knowledge of what to use and where to use it. For this reason, it is suggested that pet owners consult with a licensed pest control specialist for such applications (Angelbeck-Schulze et al., 2014; Perrins & Hendricks, 2007). In line with increasing urbanization over the last few decades, flea species that infest pets have become household pests. Thus, and for reasons of animal and human welfare, the control of fleas is of great importance worldwide. Despite the increase in the number of products available and Introduction 3 their use, flea infestation of cats and dogs is still widespread in Europe and on other continents, whereas resistance of these insects against many chemicals has been detected (El-Gazzar et al., 1986). Cat fleas are the most important ectoparasite of cats and dogs worldwide. During the past ten years, topical and oral applications of insecticides such as fipronil, imidacloprid, lufenuron and, most recently, selamectin have revolutionized cat-flea control. Recent studies show that these therapies eliminate the need to treat indoor and outdoor environments, and their use markedly reduces the severity and prevalence of flea allergic dermatitis. Surveys have yet to reveal the development of insecticide resistance to these chemical compounds. Extending the longevity of these effective host-targeted therapies should be a major goal of the veterinary community (Rust, 2005). 7. Seroprevalence Of Brucellosis In Ruminants At Sale And Purchase Markets And Abattoirs In Three Selected Districts Of Punjab, Pakistan

There is a lot of research work on livestock farms but the sale and purchase markets and abattoirs are neglected. Pakistan is an agricultural country and livestock contribute a major share to agriculture in Pakistan GDP and its economy. Sheep, goats, cattle and buffaloes contribute in the economy of Pakistani people by milk and meat production on majority basis. Livestock contribution to the agriculture value added is 56.3%, while it contributes 11.9% to the national GDP during 2014-15. More than 8.0 million rural families of the country are involved in raising livestock. But productivity of livestock is questioned by a lot of factors. Infectious zoonotic diseases are among these. Brucellosis is one of the major and unreported problem amongst livestock markets and abattoirs of our country. It results in losses to the economics of the farmers or owners in terms of poor health, abortions, long calving interval, production of weak offsprings and deprived quality of milk and meat. This disease is under investigated and hence people are unaware of this deceptive problem. Serological surveillance is a good and cheaper tool to diagnose this problem. Determination of the seroprevalence is important to know the load of disease and pockets of infections in the area of Punjab, Pakistan. There is also a need to know which diagnostic antigen is more sensitive and specific from disease eradication and control point of view. A total of 300 animals serum samples were collected from three districts of the Punjab province and tested by conventional screening test i.e. Rose Bengal Plate Agglutination Test (RBPT) by using two antigens of different countries origin i.e. VRI (Lahore, Pakistan), IDEXX (Pourquier, France) and ID.vet, France and then tested with indirect Enzyme-linked Immunosorbent Assay (iELISA). The data thus obtained regarding seroprevalence was analyzed using Chi-square SPSS version -19 (Nahar et al. 2009). Of the total 300 animals (77 male and 223 female), 1.3% male and 35.66% female were seropositive for anti-Brucella antibodies. Different risk factors were also included in study like sex, area, age, gender, abortion, reproductive problems and also history of orchitis in male. All of these risk factors were statistically analyzed to reveal the truth about ruminants brucellosis. Among the various risk factors studied, the risk factors including history of abortion (16.44%), orchitis (1.3%), retained placenta (26.47%), species cattle (35.55%) and in Toba Tek Singh (47.36%) were statistically significant which could be potential source of threat for humans and other animals. The testing was performed using conventional methods as well as using iELISA. It is concluded that iELISA is more sensitive and specific than conventional screening tests but molecular diagnosis is highly recommended for future studies. The current study will help to minimize and eradicate the low prevalence of ruminants brucellosis by creating awareness between the farmers and through vaccination and herd immunization. By implementing the policy of testing and culling of positive reactors will be pivotal to achieve the objectives. For confirmatory diagnosis all the sera were further tested with iELISA. For this purpose an exclusive commercially available iELISA Kit (ID.vet, France) was used which would be able to differentiate anti-Brucella antibodies of B. abortus and B. melitensis and this iELISA kit would be able to be used for multispecies including cattle, buffaloes, sheep and goats. 8. The Effect Of Freeze Dried Stabilizers On The Infectivity Of Newcastle Disease Virus

Poultry industry of Pakistan is playing an important role in livestock. One of the major problems due to viral disease is Newcastle disease. Live vaccines are mostly available in lyophilized form to control the Newcastle disease and can be produced on large scale at low cost, in which different types of stabilizers are used but problem is that during the freeze drying process, storage and transportation, the infectivity titre of the virus is lost, result reduced in efficacy of the vaccine. The freeze dried stabilizers protects the virus from drying during lyophilization. They protect the viruses from heat shock due to rise of temperature during vaccine transportation. The freeze dried stabilizers in six combinations (Lactose-Skimmed Milk-Egg Albumin LSE, Sucrose – Egg Yolk SE, Sucrose-Skimmed Milk- Egg Albumin SSE, Skimmed Milk SKM, Wey-bridge Medium WBM and Sucrose-Skimmed Milk SSK) were used in ND Lasota vaccine. Total 50 vials of each stabilizer were freeze dried and stored at 4-8 0C in dark. Three vials from each set were proceeded for EID50 determination at day zero then every fortnight. The freeze dried stabilizers LSE and SSE maintained the NDV stability with low reduction in mean infectivity titre log 100.40 EID50 up-to 90 days. The mean infectivity titre of NDV vaccine for both stabilizers was 106.93 then followed by SKM (10%Skimmed milk), SSK (5% Sucrose, 10% Skimmed milk), WBM (2.5% Lactalbumin Hydrolysate, 5% Sucrose, 1% Monosodium glutamate) and SE (5% sucrose, 30% Egg yolk). The freeze dried stabilizers SKM, WB and SSK maintained the NDV stability with low reduction in mean infectivity titre log 100.34 EID50 up-to 60 days. The lowest stability was achieved by SE. 9. Treatment Of Long Oblique Tibial Fracture With Intramedullary Pin Alone And Full Cerclage Wires In Cats

Leg fractures in cats are quite common problem. Fractures treated with biomechanically sound fixation and proper attention to soft tissues will be most likely to heal with a functional outcome. However, osteomyelitis, bone sequestration, and joint ankylosis still continue to be major factors in inhibiting healing and bone functionality. This research project was designed with the aim to test the effectiveness and accuracy of three sets of bone splintage devices in healing long oblique mid-shaft tibial fractures in cats, with minimal side effects and stress to the patient. For the study total twelve adult cats as and when presented at Pet Centre, University of Veterinary & Animal Sciences Lahore, and Saleem Veterinary Clinic and petshop Lahore, having long oblique fracture of tibia irrespective of breed, sex and considering almost similar age group, both stray and domesticated were selected. These cats were divided into three groups i.e groups A, B and C comprising of four cats in each group, and designated as 1, 2 3 and 4 respectively. They were admitted as clinical trial patients and were kept in separate cat cages at Saleem Veterinary Clinic until the completion of the study. Specific tagged collars were applied on all the cats. They were kept under comfortable environment and on a standard commercially available cat feed diet with ad labitum access to water. All the cats of three groups, having long oblique tibial fracture were treated with 3 separate techniques for comparison. In group A, fracture was reduced and immobilized with intramedullary pinning. In group B, fracture was treated with full cerclage wires alone while cats of group C were treated with intramedullary pin and cerclage wires. Thus comparative fracture healing and efficacy of individual techniques in their respective cases were evaluated. Subsequent comparative parameters were studied which included physical examination of wound, lameness grading, radiological scoring for callus formation, fracture line union, fracture alignment as well as callus remodelling. The collected data regarding study parameters were illustrated by descriptive statistics and non-parametric analysis of variance (Kruskal Walis Test) using IBM SPSS Statistics version 20. Conclusion: Therefore, as advocated by the results of this study, intramedullary pinning (preferably threaded) with at least 2 - 3 full cerclage wiring was a better choice for the correction of a long oblique mid-shaft tibial fracture in cats. The combination of both these techniques together give a strong backbone for bone healing to take place with minimum amount of stress to the bone fragments and also to counteract different forces acting on the fracture site.Both these techniques were easy, required little soft tissue manipulation, were cost-effective, required less equipment and could easily be applied by an average practitioner.
