502 _aHepatic C virus (HCV) infects approximately 4% of the population worldwide. HCV establishes a chronic infection of the liver that causes cirrhosis and can lead to the development of hepatocellular carcinoma. HCV is transmitted exclusively through direct blood-to-blood contacts between humans. Current drug therapies for HCV infection often fail to resolve viral infection; therefore, new approaches for treating HCV infection are crucial in controlling chronic HCV infection. The viral protease NS3 has been identified as an attractive target for anti-HCV drugs, because its activity is indispensable for processing many of the NS proteins of HCV. Although the NS3 helicase is a potentially attractive target for anti-HCV drugs, no helicase inhibitors have yet entered clinical trials. In this research we are going to isolate the NS3 region of HCV genome, from different HCV positive patients with the help of RT PCR. Then sequence these, and observe similarities in these sequences. The main objective of the present study is to determine variation in the non structural gene NS3 of hepatitis C viruses isolated from patients suffering from hepatitis and reporting to local hospitals. The ultimate objective would be to utilize this data generated in this study attempting to prepare vaccine against this virus.
