Publisher: 2010 Dissertation note: Dengue haemorrhagic fever (DHF) is an acute febrile disease caused by any of the four serotypes of dengue virus (DENV I , DENV 2, DENV 3 and DENV 4), belonging to flaviviruses. It is a public health problem of growing importance in areas where the insect vector, Aedes aegypti mosquitoes are present. A total of I 50 clinically suspected dengue patients were recruited, mean age of the patients was 31 .86+1 .66. The maximum age of the patients was 81 years and minimum age of the patient was 5 years. Out of I 50 patients 24.0% (n=36) were tested negative for any of the four serotype of the Dengue virus. The analysis showed that 45.3% (n=68) patients had DENV2 serotype, 16.7% (n=25) had DENV 3 serotype and 14.0% (n=21) patients were diagnosed to have DENV 4 serotype; none of the 150 patients was reported to have DENV I serotype. The most prevalent serotype was Dengue virus serotype 2 and Dengue virus serotype 4 was observed as lowest, while Dengue virus serotype I was having zero prevalence. Out of 150 patients 58.7% (n=88) were male while 41.3% (n=62) were females. Among the male population 31 were affected by Dengue virus serotype 2, 18 males were Dengue virus serotype 3, and Dengue virus serotype 4 was observed in 18 males while 21 males were reported negative for any of the serotype. In contrast among female population Dengue virus serotype 2 was present in 37 females, Dengue virus serotype 3 was observed in 7 and Dengue virus serotype 4 was fixed in 3 females only; while 15 females were negative for any of the serotype of Dengue virus. The data showed that p-value (p< 0.005) depict the association between gender and serotypes of Dengue fever. Our Research point out the prevalence of three out of four serotypes among the Pakistani population, which is a shocked fact as per pathogenesis theory that second infection by a different serotype may prove fatal for previously recovered person. Therefore a strict mosquito control programme is direly needed to prevent these incoming events to occur, the only hope and solution is in our environment. Moreover masses education regarding dengue fever might prove helpful in preventing zoonoses.

Differntial Diagnosis Of Malaria And Dengue Fever On The Basis Of Clinical Findings And Laboratory Investigations

Publisher: 2010 Dissertation note: I took two hundred (200) patients in total for purpose of my study. I included all cases with pyrexia of unknown origin with chills and rigors with 6-7 days history. These cases were first evaluated for Malaria by making their thin and thick films for malarial parasites.
There were thirty patients out of two hundred who were positive for malarial parasites. There complete blood picture was done that is RBC count, Heamogolobin percentage, platelet count, WBC count and ESR. The cases who were negative from malaria were further evaluated for dengue viral infection by doing capture ELISA 1gM. Before doing ELISA 1gM dengue strip method test was done and the cases who were positive on strip (Paper Chromatography) were included in 1gM ELISA study. The cases that were positive for 1gM ELISA were studied for same blood investigation which was mentioned earlier. It was also found that there had been some incidence of dual dengue infection and malaria and the incidence rate was 2%.
Now after collecting the data it was analyzed by SPSS. It was inferred afterwards from the data that all the patients +ve for dengue 1gM had been facing with low platelet count increased reticulocyte count, increased hemoglobin, decreased WBC and no significant effect on ESR had been seen. About 83% of dengue 1gM patients were having decrease platelet count. This thrombocytopenia varies from person to person and an inverse relationship has been found between dengue 1gM and platelet of the patients.
The intensity of thromobocytopenia was more in old age patients or in patients with poor health status or in those patients in which tire of anti dengue 1gM was very high. This thromobocytopenia can be used as a diagnostic tool in addition to clinical history in patients who live in periphery where the facility of ELISA is not available. The rise in platelet number indicates recovery of the patients and it should be monitored daily till the complete recovery of patients is achieved.
The rise in hemoglobin concentration has also been noticed due to hemo concentration about 76% of patients with anti dengue 1gM positive were having elevated level of hemoglobin that is ranging from 17-19 gram/dl. The increase in RBC count has also been noticed in association with increased hemoglobin concentration a mild fall in WBC count has also been noticed i-e upto 4000 in 76% of the patients. In those patients who were +ve for malarial parasites and negative for dengue 1gM, such changes in blood pictures were not appreciated although the vector of both diseases is same but AD's mosquitoes which is the carrier of dengue virus (an ARBO virus) causes more severe form of disease.

Publisher: 2010 Dissertation note: Dengue is the mosquito born viral illness which causes a broad spectrum of disease ranging from in apparent infection, flu like mild undifferentiated fever and classical DF to the most severe form DHF and DSS in which rate of morbidity and mortality is very high. Dengue infection also causes liver damage and this liver impairment is more marked in case of DHF. My study was also aimed at depicting the fact that dengue infection also causes hepatic impairment which is directly related.to the level of dengue 1gM in the body. The serum liver enzyme level rises as the infection progress.
I conducted my study over 200 patients of different age groups including both sexes. All of the patients were having history of pyrexia of unknown origin for the last 7-lO days not subsiding inspite of taking treatment. Some of them presented with mild to moderate petechiae rashes on their body. I categorized the patients into two categorize according to the WHO criteria that is the patients having high grade fever with rash and thrombocytopenia were designated as dengue hemorrhagic fever while those without rash and hemorrhagic manifestations were said to be dengue fever group. Firstly I screened all patients for anti dengue 1gM by rapid strip testing method which shows dark pink line in addition to control line in positive cases within few minutes. The strip +ve cases were further confirmed by using capture ELISA, The Dengue 1gM Capture ELISA (MACELISA) is the immunoenzymatic system recommended by the Pan American Health Organization and the World Health Organization for the serological diagnosis of dengue virus infection due to its high sensitivity, ease of performance, and use of a single acute- phase serum sample. Then all the ELISA +ve cases were estimated for hepatic dysfunction by performing their liver function test. For performing liver function test the serum was obtained by centrifuge machine and enzyme including AST, ALT and Alkaline phosphatase were done by using Merck biochemistry Analyzer which calculated the enzyme itself and displayed the reading in U/L. The analysis established that DF is more common in 21-30 years of age group, more prevalent in males and the level of serum liver enzyme rises as the serum level of dengue 1gM raises that is with the increase of severity of infection.

Publisher: 2011 Dissertation note: Mycotoxins are secondary metabolite toxins produced by fungi in or on grains, cereals and nuts used as feed in the poultry industry. Mycotoxicity in birds has been well documented and its severity increases in combination with other toxins. The study determined synergistic pathological responses in quail chicks when fed different level of Aflatoxin B1 (AFB1) and Ochratoxin A (OTA). A total of 245 quails chicks were divided into seven groups (G1-G7) having 35 quail chicks in each. OTA mixed feed was fed to quail chicks at a dose rate of 1 and 1.5 ppm in G1 and G2 respectively. G3 were fed 2 ppm OTA + 1 ppm AFB1. AFB1 was offered in the feed at a level of 1 and 1.5 ppm in G4 and G5 respectively. G6 birds were fed 2 ppm AFB1 + 1 ppm OTA while G7 acted as a control. All the birds offered toxin free basal diet for first 7 days. Day 7 was considered zero day of experiment. At this day chicks shifted to different groups of 35 each (G1-G7). Group G7 was control group and offered toxin free diet. Birds were monitored twice daily for clinical signs. Randomly selected six birds from each group were slaughtered at day 14, 21, and 28. Blood samples with and without anticoagulant were collected for hematological and biochemical studies respectively. Morbid tissue of liver, kidney, and intestine were collected for histopathological studies. The OTA groups developed anemia manifested by a significant decrease in the red blood cell count, packed cell volume percentage and hemoglobin concentration, while increase erythrocyte sedimentation rate at the end of the experiment all groups showed significant reduction in red blood cell count. This reduction was found to increase with time proportionally to the level of OTA and AFB1 alone or in combination exposure. Clinical signs in chicks administered AFB1 and OTA included depression, decreased feed intake and decreased body weight. Severity in clinical signs was dose related. Pathological lesions in liver of these chicks were hemorrhages, fatty change, centrilobular necrosis and periportal fibrosis. Microscopically, liver showed vacuolation, fatty change, congestion and individual cell necrosis. Kidney of these chicks included pyknotic changes in the epithelium of proximal and distal convoluted tubules. Severe necrotic changes in the collecting ducts and accumulation of pink homogenous material in the lumen of tubules. Intestine showed hemorrhages, edema, degeneration and infiltration of mononuclear cells were observed. OTA damaged intestinal mucosa more severely than AFB1. Serum biochemical study indicated a significant decrease in total serum proteins and increase in urea and creatinine. It is concluded that AFB1 and OTA are capable of inducing hematological and histopathological alterations in quail chicks at higher dietary concentrations, either individually or in combination.

Mycoplasmas are the smallest known bacteria, 300-800 nm in diameter and are capable of replicating outside the cell. Mycoplasma synoviae is a member of the class Mollicutes, order Mycoplasmatales, family Mycoplasmataceae. Mycoplasma synoviae (MS) is considered economically to be most important pathogen. Mycoplasma synoviae infections occur in poultry worldwide, affecting poultry and causes diseases like respiratory distress, synovitis and arthritis. Mycoplasma is transmitted from infected to healthy birds both by horizontal and vertical routes. Horizontally disease is transmitted via infected and healthy carrier birds, hatchery, housing, equipments, feeding and during transportation.
To have an insight on pathogenesis and reliable diagnostic techniques, the present project was designed to know comparative sensitivity of rapid agglutination test and polymerase chain reaction for MS diagnosis and to study the gross lesion and histopathological changes in chicken joints produced by MS.
The birds showing clinical signs that included respiratory i.e. tracheal rales, conjunctivitis, coughing, sneezing, ocular and nasal discharge and infectious synovitis were selected for sample collection. Initially the collected sera samples were examined by Rapid Serum Agglutination test. RSA and PCR tests were used in order to confirm the pathogenic agent. RSA and PCR positive samples were further processed for histopathological study in order to identify the lesions in tissues produced by causative organism. In field visits it was observed that the suspected birds were with pale comb, mild to severe lameness, dull, depressed, ruffled feather, conjunctivitis, oculo-nasal discharge, tracheal rales and greenish or sulfur faeces. Birds hock joints, toe joints and paws pad were swelled. The infected birds were occasionally found with generalized infection. The infected birds complicated with other diseases of poultry such as Newcastle and infectious bronchitis causes infection airsacculitis.
Rapid serum agglutination test was conducted at 14 broiler breeder farms. The birds at a farm were showing respiratory and infectious synovitis signs and symptoms, suspected to Mycoplasma synoviae. The tests were performed at the spot. A total of 239 sera samples were examined out of which 63 (26.35%) sera samples were positive for MS. The clinical samples were identified and confirmed as Mycoplasma synoviae infection by PCR. The amplified PCR product was given about 211 bp size while PCR buffer was used as negative control. A total of 213 samples were subjected to PCR and 65 (30.52%) revealed PCR positive results for tracheal swabs, 28.16% (20 samples out 71) showed positive results. For tracheal and lung 33.38 % (24 out of 71) and 29.57% (21 out of 71 samples) were positive, respectively. The PCR test successfully amplified the DNA of MS clinical positive samples. Sixty five out of 213 Mycoplasma synoviae isolates were positive in MS specific PCR while the other 148 samples were negative. The sensitivity and specificity of molecular method Polymerase chain reaction was 100 percent.
For histopathological studies the samples of different organs including trachea, lungs, liver, hock joints (articular cartilage, piece of synovial membrane) and foot pad were further processed. The trachea was examined. There was epithelial degeneration, desquamation. congestion, haemorrhages and inflammatory cell infiltration. The lungs were examined and it was revealed that there was marked congestion, haemorrhages, necrosis and mononuclear cells infiltration. Liver showed infiltration of lymphocytes, plasma cells and macrophages. Articular cartilage showing chondrocytes degenration. Synovial membrane was thickened due to infiltration of lymphocytes and plasma cell. Foot pad showed hyperkaratosis and thickning of epidermis, acanthosis, degeneration of cartilage, infiltration of both mononuclear and plasma cell.
It is concluded from findings of present study that PCR is more appropriate technique than RSA for timely diagnosis of Mycoplasma synoviae. However combination of findings of both techniques may be utilized for accurate detection of Mycoplasma synoviae from broiler breeder in district Abbottabad.

Histopathological Investigation Of Pleuropneumonia In Buffaloes Caused By Mycoplasma Bovis

Publisher: 2009-2011 Dissertation note: This study was conducted by keeping in view the worldwide importance of Mycoplasma bovis to cause pneumonia and many other diseases, as it causes great economic losses to bovine industry. In the current project the incidence of Mycoplasma bovis to cause pleuropneumonia was studied, and its respective histopathological changes in lungs of the pneumonic adult buffaloes and buffalo calves were examined. 100 lung samples for this purpose (50 lung samples from adult buffaloes and 50 lung samples from buffalo calves) were collected from the Lahore Bakar Mandi Abbatoir. Samples were collected on the basis of following criteria: Red hepatization, grey hepatization, multifocal abscess, necrotic lung tissue. These samples were then divided into two portions, one half placed in 10% buffered formalin in the bottles and other half kept in sterile polythene bag. The portion of lungs for bacteriological study was kept in ice box. Histopathological procedure was performed in the pathology department of University Of Veterinary And Animal Sciences Lahore. The samples were subjected to histopathological procedures and then slides were observed microscopically for the changes. Microscopically pulmonary odema, consolidation, caseous necrosis, abscess infiltration of mononuclear cells, plasma cells, macrophages, neutrophils infiltration were observed. For culturing of Mycoplasma bovis PPLO broth was prepared and samples were inoculated in the broth medium. At 7th day of inoculation the yellow color of the broth medium appeared which was indicative of positive samples. 30% positive samples in adult buffaloes and 36% in buffalo calves were obtained. These samples were then inoculated on the PPLO agar plates for further precision of results. On agar plates typical colonies of the Mycoplasma were observed under bright field compound microscope and 60% positive samples in adult buffaloes and 66% in buffalo calves were obtained. Next step towards the confirmation of Mycoplasma bovis was specific acridine staining, in which positive of Mycoplasma bovis samples gave dull yellow to colorless appearance of yellow broth medium and gave egg fried colony on agar. 78% adult buffalo and 67% buffalo calves showed positive results. These samples were then subjected to final confirmatory test which was growth inhibition disk test, in which hyper immune sera was raised in rabbits and filter paper disks soaked in this sera were used to check the zone of inhibition on cultured agar plates. 70% positive samples in adult buffaloes and 75% in buffalo calves were obtained which confirmed the presence of Mycoplasma bovis. CFU/ml of the positive samples calculated between 105-108. So the incidence of Mycoplasma bovis to cause pneumonia in adult buffaloes and buffalo calves calculated was (10% and 12%) respectively.

Exposure to Cd is known to cause harmful effects of different levels of the trophic chain, because of bioaccumulation. Toxic effects of Cd are observed in the kidneys, lungs, testes, prostate, and the erythropoietic system in chicken and rat. These effects are associated with teratogenesis and carcinogenesis. Zinc posseses protective and antagonistic action in the uptake and toxic effects of cadmium, probably because of Zn-induced synthesis of metallothionein that detoxifies Cd by firmly binding this metal. The objectives of present study are:
1) To understand the effect of zinc on cadmium toxicity in Japanese quails
2) To study the effect of zinc and cadmium on liver and kidney function along with histopathological changes.
This experimental trial was carried out at Avian Research and Training Centre (ARTC), UVAS Lahore and tests were performed at Pathology Deptt UVAS Lahore. Day old Japanese quails (C. coturnix japonica) (n = 560) were used in this experiment. For this purpose, a total number of 560, day old chicks of Japanese quail were procured from the hatchery of ART Centre. They were assigned to seven dietary treatments. There were four replicates in each treatment group and each replicate was of twenty chicks.
A =Control diet group will receive only basal diet without any supplementation. B= Basal diet + 50 mg/kg Cd, C = Basal diet + 75 mg/kg Cd, D =Basal diet +100 mg/kg Cd E=Basal diet+50 mg/kg Cd+40 mg/kg Zn, F=Basal diet+75 mg/kg Cd + 40mg/kg Zn and G =. Basal diet+100 mg/kg Cd + 40mg/kg Zn. Each group consisted of 80 birds. The body weight of each the birds was carried out weekly. Blood samples were collected on the 21st (3rd week), 28th (4th week), 35th (5th week) and 42nd (6th week) days of treatment from each group to evaluate the liver and kidney functions and vital organs were collected after slaughtering from each group and then histo-pathological analysis was done. The data was analyzed by two way ANOVA.

Etiological Study Of Pancytopenic Children

Publisher: 2011 Dissertation note: Pancytopenia is a hematological condition in which there is a decrease in all three cell lines of peripheral blood i.e. erythrocytes, leucocytes and platelets leading to anemia, leucopenia and thrombocytopenia. Complications of anemia, repeated infections and bleeding tendencies are sometimes horrifying and may result in death of individual.
The present study was designed to analyze the underlying pathology, different clinico-haematological features and importance of bone marrow study in one hundred children presenting with pancytopenia. Present study was carried out in pediatric laboratory of Mayo Hospital, Lahore, Pakistan..
Detailed history was taken in all cases. Complete blood counts were done on an automated blood analyzer (Sysmex Kx-21). For counter check of total leucocyte count, differential leucocyte count and platelets, smears were also prepared and stained by using Giemsa stain. Red cell morphology was done on blood smear for theconfirmation of red cell indices. A total volume of 3 ml venous blood was drawn into a syringe. Out of which 1.0 ml was delivered into EDTA containing vacutainer and remaining 2 ml blood was transferred to a plain glass tube. After clotting and centrifugation serum was obtained for screening of hepatitis B surface antigen and antibodies against hepatitis C virus. Bone marrow aspiration was also performed where indicated.
Megaloblastic anaemia (42%) Aplastic anaemia (26%) and ALL (8%) were found to be the common causes of pancytopenia in our scenario. Less common causes of pancytopenia were infections (8%), mixed deficiency (4%), MDS (4%) and lymphoma (4%). In all above mentioned cases clinical manifestations and peripheral blood counts played an important role in their evaluation.
Two cases of haemophagocytic syndrome (2%), a rare cause of pancytopenia were also diagnosed in this study.
This study also explained the importance of physical examination, peripheral blood findings and bone marrow examinations in the management of pancytopenic patients. Peripheral blood film and bone marrow aspiration should be performed simultaneously in pancytopenia patients when the diagnosis is not confirmed. Bone marrow examination in most cases gives the specific diagnosis. However, in few cases, additional tests may be required.
Serum vitamin B12 and folic acid levels may be needed for confirmation of megaloblastic anemia. Serum iron, TIBC and iron staining on bone marrow smears may be required in iron deficiency anemia. In cases of leukemia flow cytometry study may be more helpful in reaching a final diagnosis. Bone marrow biopsy is mandatory in aplastic anemia.

Pathology Of Experimental Enterotoxemia In Sheep And Goats

Publisher: 2012 Dissertation note: During the present study intestinal scrapings were collected from sheep suspected for enterotoxaemia. Samples were subjected for the isolation by repeated culturing in Reinforced Clostridium medium. Growth on blood agar revealed characteristic colonies of Clostridium perfringens after 18-24 hours. Biochemical and mice inoculation tests were performed. Isolated organism was identified by indirect ELISA. The pure growth was subcultured in RCM in bulk quantity and calculated the dose for experimental infection.
In the second part of the study, sheep and goats were procured, dewormed and kept in the experimental house of UVAS, Lahore. The experimental infection comprised of the whole culture of C. perfringens type D was inoculated intraduodenally via para-mid line between animals of group A and B while in animals of group C and D inoculated only starch solution to achieve the objectives. Accumulative clinical score in sheep was found to be 9 to 16, 13 to 22 and 15 to 23 at 10, 20 and 30 hours PI respectively while in goats the accumulative scores varied from 5 to 15, 9 to 16 and 14 to 21 at above mentioned time intervals. The highest mean score for clinical findings in sheep was anorexia, frothing followed by dehydration while in goats, the highest mean score was recorded for diarrhea dehydration, and anorexia. No significant clinical findings were noted in control groups.
There was a significant increase in blood glucose, urea and serum creatinine in infected group of sheep and mean values reached up to 141 mg/dl, 92 mg/dl and 7.5 mg/dl respectively at 30 hours PI while in goats a similar pattern was observed with the mean values raised to 142 mg/dl, 111 mg/dl and 10.2 mg/dl for blood glucose, urea and serum creatinine respectively. There was no significant change found in RBC and platelet count of both species but there was an early increase in the mean WBC count of sheep 19.7x103/µl at 10 hours but then decreased to 14.7x103/µl at 30 hours PI while in goats it was 23.6x103/µl and then decreased to 15.3x103/µl. The mean PCV % age increased in animals of both infected groups but more in goats and reached to 52% in 30 hours.
During the third part of the present study, the animals were slaughtered and PME performed. The accumulative score for gross lesions were recorded and it was found between 14 to 24 in animals of group A with the highest score for congestion and edema of different organs whereas in goats it was between 12 to 22 with congestion and hemorrhages of intestine having highest scores. The samples were kept in formalin for histopathological examination and accumulative lesions score was noted in different organs. The highest mean score in sheep was recorded in kidneys and lungs and in goat intestine, lung and kidneys were the major organs affected.
A polymerase chain reaction was optimized under our own laboratory condition for the detection of alpha and epsilon toxins of Clostridium perfringens type D from different tissues. The erythrocytes of different species were used to know the sensitivity against culture supernatants of C. perfringens type D. It was observed maximum hemolysis occurred in human erythrocytes (68%) followed by mice (57%) at 37oC. It was also recorded that a significant increase was found at 37oC as compared to25oC except for dog and rabbits where no significant difference was observed.

Changes in hematological, serum biochemical and histopathological parameters are good indicators for diagnosis of theileriosis. For this purpose a total of 100 blood samples from infected sheep as well as 50 blood samples from healthy sheep were collected on the basis of clinical examination and peripheral blood smears from different farms in and around Lahore. The collected samples were processed at Clinical Pathology Laboratory, Department of Pathology, University of Veterinary & Animal Sciences, Lahore. Hematological parameters were measured by automatic hematology analyzer & serum biochemical parameters were measured by using commercial kits by colorimetric methods using spectrophotometer. Histopathological changes were observed by making slides of tissue samples, and observed under microscope. Molecular confirmation of Theleria sp. was done by using Polymerase Chain Reaction. Finally the data obtained for hematological and serum biochemical parameters were analyzed by using Student's t test.
Previous studies on serum biochemical parameters and hematological parameters were mostly done in large ruminants. But there is little investigation available on the biochemical and hematological alterations of theileriosis in small ruminants. In this study blood samples taken from sheep were examined under microscope by making thin blood smears using Giemsa stain. Theileria piroplasms were seen in RBCs at 100x. Blood samples were also processed for PCR test. In all Theileria ovis positive samples 520-bp fragment was generated on gel by using primers TSsr 170F and TSsr 670R for sheep. All samples positive on microscopy were also positive by PCR. This study provides the base line data for molecular diagnosis of Theileria ovis in sheep in Pakistan. Hematological and biochemical parameters were evaluated in the sheep naturally infected with theileriosis while healthy sheep were selected as control. Hematological findings revealed a significant decrease (P<0.05) in RBC count, WBC count, Hb Conc. and PCV in infected animals as compared with healthy animals. Serum biochemical findings also revealed alterations in activities of enzymes and plasma proteins. A significant decrease in total proteins, albumin, glucose and creatinin and triglycerides was observed while significant increase (P<0.05) in ALT, AST, bilirubin was observed in affected animals as compared with healthy animals. A non significant increase in urea and cholesterol concentration was also observed in infected animals as compared to healthy animals. Different tissues were examined to study gross and histopathological changes. Liver was large, pale and friable. Prescapular lymph nodes were swollen and spleenomegaly was also seen. Kidney showed hemorrhagic spots and some were mottled. Histological examination of the lymph nodes revealed edema, widened intercellular spaces, parenchyma degeneration, massive lympholysis, widened sinuses at some areas and macrophages. Splenic nodules were sparse, diminished in size and lymphocyte depletion in follicles was prominent.In liver, mild infiltration of leukocytes around blood vessels, vacuolation in hepatocytes, increase in sinusoidal space and atrophy of hepatic cord was seen and hepatic cord was broken. In kidney, degeneration of tubular epithelium cells was seen. This study concluded that theileriosis in sheep was associated with some alterations in blood parameters and histopathological changes which could be useful in the diagnosis of ovine theileriosis.

However infectious and noninfectious diseases are major threat to the growth of poultry industry. Among the infectious diseases, infectious bursal disease (IBD) is one of the major malady which causes great economic losses to poultry in terms of both morbidity and mortality. IBD virus was first identified in 1962 in gumboro, since then the disease spread to all parts of world. Until 1980s IBD virus was efficiently controlled by use of vaccine however with the emergence of variant strains of virus it was become difficult to control the disease. Studies on molecular structure of IBDV revealed that it has a hyper variable region in VP2 gene. Mutations in this hyper variable region lead to emergence of different strain of virus and hence causing vaccine failure.
In Pakistan there is an outbreak of disease each year despite of vaccination. So there was a need to characterize field isolate and vaccine isolate of IBDV to find any mutation in field isolate of virus. The current study was designed by keeping above factors in mind. Purpose of present study was to compare local field and vaccine isolate of virus. RNA was isolated by TRIzol method from both vaccine isolate and bursa collected from field. RNA was converted to cDNA by using oligoDT. In next step VP2 gene was amplified by using a set of primers P1 and P2.
cDNA was digested by 2 enzymes BstN1 and MboI. Results of RFLP showed that there was a difference in genetic sequence of both strains. Howeverthe pattern of disease produced by both of the strains was same.
Chicks were given experimental infection at age of 2 week and slaughtered after 3 days post infection. Gross and histopathological lesions were observed and compared to each other and also with negative control. Statistical analysis was done by using one way ANOVA followed by Duncan's multiple range test.

A total number of 144 commercial broiler 1-day old chicks were reared in the experimental sheds of the Department of Pathology, University of Veterinary & Animal Sciences, Lahore, The birds were fed with balanced commercial feed and water ad libitum. The birds were divided into 3 groups; A, B and C having fourty eight chicks each. Birds of all groups were sub divided into four groups of each i.e. A1, A2, A3 and A4; B1, B2, B3 & B4 and C1, C2, C3 and C4, respectively. Each of the sub groups containd 12 birds. Sub groups A4, B4 and C4 were control group with no medication. The birds of groups A, B and C were fed with poultry feed containing dry powder of neem leaves @ 2 gm, 4 gm and 6gm per kg of feed respectively. The birds of groups A1, B1 and C1 were treated with the herb from day 0 to 42 of their life. The birds of groups A2, B2 and C2 were given the neem from day 14 to 42 of their life, whereas the birds of groups A3, B3 and C3 were treated with the herb from day 28 to 42 of their life. Difference between weekly weight gain in the birds of groups A1, B1 and C1 was non significant (P>0.05) however the difference between weight gain in the treated and control groups was significant (P<0.05). The birds treated with the herb from day 0 of their life showed more weight gain. There was no difference in the haematological indices between all of the treated groups and the control groups. The neem treated birds showed increased antibody titers against ND and IBD viruses as compared to control groups. The values of ALP and ASTshowed decreasing trend when the level of neem leaf meal was increased in the ration. Serum creatinine and serum uric acid values posed a slight declining trend in the neem fed birds. There was a decrease in serum cholesterol level in the neem treated bird groups, the higher the concentreation of the herb, the lower the cholesterol value. The organ body weight indices showed that there was no significant difference in liver, spleen and thymus weights among treated groups as well between treated and control groups. There was absence of prominent gross pathological lesions in liver, spleen, kidneys and thymus, however some treated groups showed mild hypertrophied liver and kidneys as did the organs of the birds in control groups. No histopathological changes except a few mild changes were observed in liver, spleen, kidneys and thymus in the birds of experimental groups.

Pathological Findings Of Field Isolated Peste Des Petits Ruminants Virus (Pprv) In Experimentally Infected Goats

It is highly contagious viral disease of economic point of view. It produces hurdles in farming of small ruminants in areas where it is present such as Africa and Asia. Those areas where the disease is endemic and huge numbers of small ruminants are reared by needy farmers; it threats their subsistence. Epidemically the disease identified since 1991 in Punjab province of Pakistan.
The aim of the present study was detection of PPR virus through Reverse Transcription-Polymerase Chain Reaction during period of incubation. The course of the disease was monitored through regular clinical examination and hematological profile. The nature of the disease was evaluated through gross and microscopic lesions. The comparative proteomic analysis of field and vaccinal PPR virus was done through SDS-PAGE.
A total of twenty healthy teddy goats were purchased from local market of Lahore and were reared for 21 days. The goats were randomly divided into Group-A (experimental group) and Group-B (control group), with ten goats (n=10) in each group. Experimental infection of field isolated PPR virus was given intratracheally to the goats of group-A. The goats of group-B served as un-infected control group. For early detection of PPR virus through RT-PCR, ocular and nasal secretions were collected on day 1, 3, 5 and 10 from experimentally infected goats and on day 5 and 10 from goats of control group. Clinical examination of all goats of both groups was performed on daily basis. For hematological analysis, 2.5 mL blood was drawn from jugular vein on day 0, 1, 3, 5, 7, 9, 12, 15, 18 and 21st day post infection from all goats of both groups. Gross and microscopic lesions were recorded after slaughtering one goat from each group at day 7, 14 and slaughtering all the remaining goats of both groups on day 21 post infection. Two goats of the experimental group were died naturally, one goat at day 8 and one goat at day 13 post infection. Proteomic analysis of PPR virus harvested from experimentally infected goats was done through SDS-PAGE and was compared with PPR virus vaccinal strain Nigeria 75/1.
The results showed that PPR virus was detected by RT-PCR in PPRV infected goats at day three post infection and before the occurrence of acute clinical signs. The PPR virus was not detected in uninfected goats throughout the studied period which showed that there was no natural circulation of virus in the area of experimental sheds. The clinical examination showed significant increase in rectal temperature, pulse and respiration rates in PPRV infected goats as compared to uninfected goats. From the comparison of hematological parameters in PPR infected and uninfected goats, it was observed that PPRV is linked with some obvious alterations in its hematological profile. PPR in experimentally infected goats led to obvious decrease in the number of leucocytes, erythrocytes, platelets, hemoglobin estimation and packed cell volume but marked increase in mean corpuscular hemoglobin, mean corpuscular hemoglobin concentration and mean corpuscular volume as compared to uninfected goats. The gross lesions were specific for PPR which were prominently observed in the digestive and respiratory systems and lymphoid organs. The microscopic lesions revealed that there was congestion in trachea, sloughing in mucosa of rumen, mitotic activity in cardiac myocytes and hemorrhagic & effaced mediastinal lymph nodes of infected goats. From the comparative proteomic analysis through SDS-PAGE, differences in bands of proteins were observed.

assessment, about 250 million cattle are endangered by this disease and millions of high milk
yielding cattle are at risk of exposure to disease. It acts as a bigger restraint on livestock
improvement and production in many developing countries (Nagore et al. 2004). Theileria
annulata is the main specie that causes high morbidity and mortality. It causes heavy economic
and production losses in cattle in tropical and sub-tropical regions. The recorded mortality rates
in cattle reached to 70% (Moor house et al. 2001).
Theileria species are intracellular obligate hemoprotozoan parasites. All Theileria species
are dangerous and cause disease but two of them are important for livestock. Theileria parva and
T. annulata produces diseases named as East Coast fever and tropical theileriosis in cattle
respectively. Genus Theileria has many other species like T. buffeli, T. taurotragi, T. velifera, T.
sergenti and T. mutans. These species cause infections in wild and domesticated ruminants.
Theileria species present in large and small animals show signs like fever, anorexia,
swelling of the superficial lymph nodes, dyspnoea, lethargy, progressive anemia, constipation,
diarrhea, lacrimation and nervous symptoms (Saeed et al. 2010; Irvin and Mawmachi 1983). A
pronounced rise in body temperature, reaching 40-41.5 °C is pursued by lacrimation, depression,
swelling of the superficial lymph node and nasal discharge. The characteristic sign of tropical
theileriosis is anemia and finally haemoglobinuria occur with heavy weight losses. The clinical
course of the disease alter from per acute to acute or sub-acute to chronic (Oliveira- Sequeira et
al. 2005). The disease is lymphoproliferative in its early phases resulting enlargement of lymph
nodes, later on enters lymph destructive phase which is associated with a pronounced
leukopenia. In the piroplasms phase in erythrocytes, the parasite becomes infective for the tick
(El-Deeb and Younis 2009).
Trans placental Bovine Tropical Theileriosis causing a deadly disease in a 3 day old
neonate cross bred calf and cerebral form of the disease (turning sickness) in a cow were
incriminated to T.annulata infection. It mainly depends upon the harmful effects of the T.
annulata on lymphoid tissues and susceptibility of the host (Sudan et al. 2012).
Theileriosis is prevalent in various regions of the world including Pakistan. It is transmitted by
Hyalomma species ticks. These ticks spread T. annulata which causes tropical theileriosis
(Durrani et al. 2009). The developmental stages of Theileria inside the Hyalomma ticks varies in
different shapes and forms (Hamed et al. 2011). Therefore to increase the milk and meat
production of cattle we can prevent the spread of the disease by controlling ticks
(Hekmatimoghaddam et al. 2012). The sufficient amount of Hyalomma ticks are found in warm,
commonly hard marshland and in central and Southern Europe, south west Asia and Southern
Africa having very long dry season. A toxin is produced in the adult ticks. This toxin produce
clinical signs of mucus membrane hyperemic and moist profuse eczema (Adam et al. 2000).
The sporozoites of Theileria enter into cattle host during tick feeding and they
immediately infect mononuclear leukocytes, these sporozoites develop into macroschizonts and
induce proliferation of the host cells. Macroschizonts constantly mature into microschizonts and
finally into merozoites, which are discharged from leukocytes. These merozoites attack
erythrocytes and mature into piroplasms, become available to ticks. Infective sporozoites,
injected during tick feeding, rapidly enter target cells, escape from the surrounding host-cell
membrane and differentiate to schizonts that interact with different host-cell components
(Dobbelaereand Rottenberg 2003). This interaction includes host cell signaling pathways that
regulate proliferation and cell survival (Chaussepied and Langsley 2011) and thus cause
blastogenesis and clonal expansion of predominantly T and B cells (Fawcett et al. 1982; Baldwin
et al. 1988; Spooner et al. 1989). Merozoites released from these schizonts subsequently infect
red blood cells and become trophozoites. Lymphocytic stage of Theileria (schizonts) is the cause
of many of the severe disease manifestations like lymphadenopathy, pyrexia, thrombocytopenia,
and panleukopenia (Homer et al. 2000). Marked anemia, anisocytosis, pikilocytosis, and
leucopenia were commonly observed in bovine theileriosis (Ceci et al. 1997). Cattle may survive
the disease, but recovery and convalescence may be protracted and incomplete, this leads to
permanent debilitation, loss of productivity and prolonged carrier state. (Shahnawaz et al. 2011).
Cattle with subclinical infection in endemic regions become carrier of piroplasms and act as a
source of infection for the vectors (Brown 1997; Brown 1990; Uilenberg 1995).
The diagnosis of theileriosis in acute cases is majorly done on clinical signs and Giemsa
stained blood smears of cattle but the detection of agent is not reliable and is almost impossible
in carrier stage. Advances in molecular biological techniques have resulted in the improved
detection, identification, and genetic characterization of many hemoparasites. Species specific
polymerase chain reaction (PCR) has been developed for the detection and identification of
various Theileria species and has been shown to have higher sensitivity and specificity compared
with serological assays and examination of Giemsa-stained blood smears (Bhoora et al. 2009).
Primers were derived from the gene encoding the 30-kDa major merozoite surface antigen for T.
annulata (Aktas et al. 2006).
Most of the previous studies on haematological parameters in T. annulata infection were
carried out on experimentally infected cattle (Sandhu et al. 1998; Singh et al. 2001). The present
investigation was conducted to study haematological parameters in cattle naturally infected with
T. annulata. Hematology has been broadly used in attempts to give information about disease
condition, performance problems and health in cattle (Rezaei and Naghadeh. 2006).
Hematological and sero-biochemical alterations are the indicators of severity of disease and are
considered to be good tools for the diagnosis, prognosis for effective therapy (Col and Uslu
2007; Nazifi et al. 2010b).
Lahore is one of the larger district in the Punjab province of Pakistan. Different cattle
breeds are reared by the people of the area for meat and milk production. The exact current
situation about the prevalence and pathogenesis of Theileriosis in the selected area is unknown.
The present study was conducted to screen cattle by finding schizonts or piroplasms in Giemsa
stained thin blood smears at slaughter house of district Lahore (Aktas et al. 2006) and later to
confirm through Polymerase Chain Reaction (PCR) (Chaisi et al. 2013) in order to implement
efforts and regulation to eradicate the spread of disease in the selected area. Data generated from
this study provided the latest status of Theileriosis, sex wise prevalence and its pathogenesis in
cattle population of Lahore. The study has also provided the necessary information to formulate
strategies for control of disease in the area. An investigation was also undertaken to ascertain the
changes in haematology as a result of Theileria annulata infection. These studies will help better
understanding of the pathogenesis and supportive therapy of this disease.

Publisher: 2015 Dissertation note: Among the non-contagious diseases Bovine ephemeral fever is important disease of cattle the course of the disease is usually three days due to which it is called “three days sickness”. This is transfer to other cattle through insects Culicoides (biting midges a group that include many kinds of flies) and mosquitoes. Bovine ephemeral fever virus (BEFV) has been collected from Culicoides coarctatus, Culicoides brevitarsis, and Anopheles bancroftii (Walker et al. 2012). Cattle and buffaloes are the main species affected from Bovine ephemeral fever (BEF) which gives huge economic losses to the dairy sector. The etiologic agent, Bovine ephemeral fever virus belong to Rhabdoviridae family, enveloped (negative sense) ssRNA virus. It generally recur in Australia, Asia, and Africa also in the Middle East (Walker 2005).
The theme of the present study was detection of BEF virus through Reverse Transcriptase-Polymerase Chain Reaction from the cattle suspected for bovine ephemeral fever virus on the basis of clinical signs. Hematological profile and serum calcium level were checked in the confirmed positive samples for BEFV.
A total of 50 blood samples were collected from the suspected animals in aseptic condition using a sterilized disposable syringe and were preserved in vacutainers (Anticoagulant added n = 50, without anticoagulant n = 50). The 10 blood samples were collected from healthy animals in vacutainers (Anticoagulant added n = 10, without anticoagulant n = 10).
Buffy coat were separated from blood samples and from this the RT-PCR was performed and successfully diagnosed the BEFV infected cattle. Hematology and serum calcium were performed for both confirm positive and healthy animals.
The result showed that BEF virus was diagnosed with the help of RT-PCR in samples suspected for BEFV infection, and there was no virus detected in samples taken from healthy animals. Comparison of hematology between BEFV infected cattle and healthy animals were performed there was no changes in the RBC, Hemoglobin, Hematocrit, MCV, MCH, MCHC, and MID (it include monocyte, eosinophils, and basophils) except Neutrophils, which number was increases and lymphocytes which was decreased in BEFV infection, while in healthy animals there was no change in the whole hematology. Serum calcium was also determined there was decrease in serum calcium level of BEFV infected cattle, while in the healthy animal samples there was no change in the serum calcium level.

They are of major economic importance in commercial chicken flocks, causing huge losses. As both in humans and animals it is well documented that preceding respiratory infection of virus predispose individual to bacterial infection. Moreover, mix infection in poultry which occur when different organisms simultaneously invade birds is a major threat to poultry industry causes highly epidemic debilitating disease with high mortality which eventually leads to economic catastrophe. In recent past prevalence studies of field, E. coli had been reported with high prevalence and exaggerated disease along with other respiratory pathogens, additionally IBV had also isolated from same flocks in same season. Although a plethora of pioneering work had been done on IBV and E. coli in the previous decades but still a window in time exist in revealing there co-infection. Looking to field scenario in our country, the present study was designed to study an ideal challenge model for IBV and E. coli, by reproducing the natural infection.
80 SPF day old broiler birds were arranged into four groups, (A, B, C and D). Each group was comprised of 20 birds. Group D served as uninoculated control while, Group A and B were challenged with IBV on 23rd day of trails, and Group B and C were inoculated with E. coli infection on day 26th. Birds, (n=3) from each group were slaughtered on various days post infection, gross and histopathological lesions were observed and serum samples for HI were taken, throughout experiment. Variable clinical signs were recorded in various groups. In IBV infected group, respiratory distress i.e., tracheal rales, coughing, sneezing and gasping were noted during early stages, later up to 10 days post infection watery diarrhea with ruffled feathers were observed. In mix infected group clinical signs manifested rapidly and were persistent with high severity. Gross lesions in mixed infection were more profound,
including; airsacullitis, tracheitis with catarrhal exudation throughout respiratory tract; severe sepicemic lesions i.e. perihepaitis, pericarditis, pneumonia and polyserositis with swollen and pale kidneys distended by urates. 5 birds died in mix infected group revealed ascites with asphyxiation of trachea with caseous exudate. While in IBV infected group lesions were mild and confined to trachea, airsac and kidneys. Mortality was high in mix infected followed by IBV in which two birds died. While in E. coli and control group mortality were not noted. Histopathological lesions in mix infected group were aggravated markedly tracheal epithelium degeneration, deciliation and sloughing; congestion, interstitial nephritis, leukocytes infiltration, tubular degeneration and necrosis while were observed. In lungs, pneumonia of peribronchiolar area and interstitium with lymphocyte and macrophages infiltration, additionally degeneration and vacuolization of hepatocytes with focal necrotic areas were also noted. In IBV and E. coli group microscopic lesions were of mild degree. GMT of both IBV and mix infected birds were high but were not significant different (P>0.05). Among the groups, statistically significant increase in FCR of birds in mix infected group was observed followed by E. coli, with IBV infected came third in the row. On the bases of these findings we might conclude that mix infection of IBV and E. coli causes severe lesions with high morbidity and mortality.

Studies On The Effect Of Garvit-Pro® (A Commercial Product) On Broiler Chickens Vaccinated Against Infectious Bursal Disease

The disease affects the primary lymphoid organs (bursa of Fabricius) of the poultry and cause mild to severe immune suppression in affected birds. In accordance of the mentioned fact, present experimental project was designed to evaluate the effects of immune boosters after IBD vaccination and compare the effects of GarVit-Pro with only vaccinated birds. Humoral immune response against IBD virus was measure through Enzyme Linked Immunosorbent Assay (ELISA). There were total 90 bird, divided into 3 groups with 30 birds in each group. The blood sample were collected from different experimental groups of broiler chickens on 07, 14, 21, 28 & 35 days of age. Among the various experimental groups of broiler chickens the significantly highest antibody titer was recorded in group B fed with GarVit-Pro (Garlic Supplement) as compared to the other vaccinated and unvaccinated groups. The histopathological scoring of Bursa of Fabricius and thymus in different experimental groups was observed at day 07, 14, 21, 28 and 35. The bursal lesion scores of Group B (GarVit-Pro Treated) was lower than the other two groups. The GarVit-Pro helped to enhance the antibody titer against IBD virus after vaccination. These findings suggest and advocates that GarVit-Pro (garlic supplement) can effectively stimulate and enhance the immunity in broiler chickens. GarVit-Pro can be potential ameliorator against different vaccines and their unwanted/suppressive effects in broiler chicks. It was proved that GarVit-Pro is able to implement immune response and have a patent immunomodulatory effect in chickens

Pathological Investigation And Molecular Detection Of Avian Pathogenic E.Coli Serogroups In Broiler Birds

The present study was attempted to scan the rfb gene clusters in APEC predominant serotypes O1, O2 and O78 strains and to develop Multiplex PCR method for serotyping of the O-antigens. The Multiplex PCR method was used for the identification of serotypes of APEC. The second part of the study was to study the pathological lesions caused by most prevalent serogroup in experimentally infected broiler chicks.
A total of 100 tissue samples (lungs and livers) were collected from colibacillosis suspected broiler birds. Streaking was done from these samples on three different media and it was found that 80% isolates were positive on MacConkey media, 60% were positive on EMB media and 40% were found pathogenic for E.coli on Congo red media.
The colonies which were of pink color on congo red media were considered as pathogenic. DNA was extracted from these colonies by boiling method by picking single colony from each petri plate. Extracted DNA was further used for PCR to confirm the three serogroups i.e O1, O2 and O78.
The PCR results showed that 8% isolated samples were found as pathogenic as O2 strain was found dominant among all. Only two genomic DNA samples were found of O1 serogroup After confirmation of serogroups inoculum of Avian pathogenic E.coli O2 strain was prepared to experimentally infect the broiler birds. Birds were infected at the age of day 7 via intratracheal route.
Following the experimental infection of birds, they were monitored for any pathological lesions which were not present significantly while some birds were off feed, reluctant to move, head down posture and were keeping themselves in isolation.
Postmortem of dead birds was performed and pathological lesions were noted. Livers were found to be congested, enlarged and white fibrinous layer over liver was present. Lungs were also affected with the disease and white layer was present on lungs too. Lungs were consolidated and congested.
Histopathology of lungs and livers was performed. It was noted that there was mononuclear cells infiltration and thin fibrinous layer over liver. Thickening of the liver capsule was noted due to infiltration of mononuclear cells and there was marked congestion in hepatic portal areas and the central vein. There was atrophy of adjoining hepatic cords due to greatly distended and congested sinusoids. Besides these changes, hepatocytes in various stages of degeneration along with hemorrhages, areas of congestion and fatty changes in a few places could be seen.
There was infiltration of heterophils, severe congestion, lymphocytes and macrophages in the wall of the bronchus as well as in the peribronchial alveoli. There was marked presence of granuloma in lungs. Some birds displayed thickening of the pleura and consolidated areas covered with yellowish fibrin in lungs.
The experimental infection of avian pathogenic E.coli confirmed the hypothesis that it causes pronounced histopathological lesions in broiler birds.

Publisher: 2016 Dissertation note: Canine Parvo virus infection is causing fatal disease to dogs resulting in severe morbidity and mortality. It is becoming disastrous for canids in Pakistan. It is important to study the pathogenesis of this deadly virus. Parvovirus infection has become more pathogenic with passage of time and its pathogenicity may be enhanced due to possible mutation in field virus. The possible mutations may be the cause of vaccine failure and rapid outbreaks.
In present study, sum of 50 fecal samples from different areas were collected and properly labelled. Both serological and molecular confirmation was done.
Haemagglutination test was used as a screening test. HA test was performed to check agglutinating activity of samples as described by (Desario et al. 2005). Haemagllutination was done and only 20 samples were found positive. Positive samples were selected for polymerase chain reaction. DNA of selected samples was extracted through kit method according to protocol provided by the manufacturer. Nano drop was done to check quantity and purity of viral DNA. PCR was performed. Already reported primers were used for detection of CPV (Buonavoglia et al. 2001). The PCR product was confirmed by running the PCR product on 1.2% agarose gel. Electrophoresis was performed at 110 Volts for 30 minutes. The amplified PCR product was further purified using GeneJet Gel Extraction Kit using instructions provided by the manufacturer. Purified samples were sent to commercial lab for Sanger sequencing. The obtained sequences were examined using BLAST (Basic Local Alignment Search Tool). Sequences were compared with other sequences already obtainable from the GenBank database (http://www.ncbi.nlm.nih.gov) and were carefully aligned by BioEdit software (version 220.127.116.11) using the ClustalW method. The MEGA software program, version 6.0 was used to construct a phylogenetic tree using the neighborhood joining method.
Complete blood count was performed on whole blood samples. The samples were divided into two groups (infected & non-infected) based on the results of PCR. Paired T-Test was applied on the results obtained and results showed that all parameters statistically showed significant difference between two groups except WBC’s but still the number of WBC’s decreases in infected animal like other parameters. Serum chemistry analysis was performed on serum samples. These samples were also divided into two groups (infected & non-infected) based on the results of PCR. Paired T-Test was applied on the results obtained and results revealed that all parameters statistically showed significant difference between two groups except ALT and creatinine levels.
Results showed that new CPV-2a was found to be the major circulating strain of Pakistan as out of 7 sequenced samples, 6 were identified as CPV-2a and only 1 of them was CPV-2. PCR was found to be the most sensitive assay for the detection of CPV, as compared to HA and ICT- CPV antigen detection kit. The strain in vaccine was identified as CPV-2, this finding was considered important because it indicates one of the reason for vaccine failure. The phylogenetic analysis of strains showed that all the samples collected from different regions of Pakistan falls in one clade and branching pattern is shared with the neighboring countries especially China. This information reveals that our virus is genomically similar to our neighboring countries India and China. Upon hematology and serum chemistry analysis, it was revealed that the virus does effect the blood parameters and all the red blood cell indices were lowered except platelet count.
The present study enabled us to study pathogenesis, to determine currently prevailing strain of CPV in Pakistan, identify the cause of disease occurrence even after vaccine, to study pathogenesis of CPV, to characterize and construct phylogenetic tree of currently prevailing canine parvovirus field isolates.

The liver has an excellent ability to recover from injury and regenerate new tissues. Liver is a highly vascularized organ and chances of bleeding are very high in hepatic surgeries. Liver is directly supplied by hepatic artery and drained by portal vein. We often come across certain problems in liver which require surgery. When an incision is given in liver there is a high risk of bleeding. In severe cases animal can lead to death due to hypovolemic shock. In this study we used latest and most advanced hemostatic agents by using Surgicel Snow and Surgicel Fibrillar to control bleeding during hepatotomy in dogs. These hemostats are affordable, safe, reliable and convenient.
Considering the significance of hemostasis in surgical interventions, this project was designed to compare the advance hemostatic agents e.g. Surgicel Snow, Surgicel Fibrillar and sterile Surgical Gauzes. This study helped us to identify the best method that can be used for future surgical interventions in dogs, with the aim to minimize surgical bleeding in liver surgeries to accentuate early recovery.
On the basis of findings of this study: it was concluded Surgicel Snow is best hemostat than Surgical Fibrillar. It is the best hemostat with minimum blood loss and least effects on liver function.

Effect Of Unilateral And Bilateral Thyroidectomy On Hormonal And Biochemical Parameters In Dogs

Publisher: 2016 Dissertation note: Thyroid gland is one of the most important endocrine gland of animal body. T3 and T4 are two major hormones secreted by thyroid gland. These hormones have a major role in metabolism of cells and organs. In normal thyroid both these hormones have anabolic effect. Thyroid tumors account for 1.2-3.8 % of all types of tumors in dogs. Unilateral, bilateral or subtotal thyroidectomies are practiced to remove benign or malignant thyroid carcinomas.
The present study was conducted on 12 mongrel dogs. These dogs were rendered to 3 groups with 4 dogs in each group. One group acted as control, in which no surgery was performed, second group underwent unilateral removal of thyroid gland while the third group underwent bilateral removal of thyroid gland. Free thyroxine, thyroid stimulating hormone level, calcium, W.B.Cs, R.B.Cs and haemoglobin concentration were monitored for 21 days post-operatively.
The data was expressed as mean and standard deviation. Results of the study were analysed by 2 samples T-test, one way ANOVA using multiple comparison test on SPSS version 16.0.
Based on these results it was concluded that bilateral thyroidectomy was effective in relieving hyperthyroidism but there is a need to immediately initiate levothyroxine therapy. Free T4 level drops near to zero by day 21 and TSH level rises by many folds. On the other hand, in unilateral procedure Free T4 level remain in normal range and 75% of dogs require no levothyroxine therapy. Level of calcium in unilateral and bilateral thyroid removal remains within normal range provided parathyroid glands are preserved properly. Haemoglobin concentration in both the procedures is reduced significantly postoperatively. There is a need to monitor blood parameters postoperatively.

Use Of Xylazine And Detomidine Alone And With Ketamine Anesthesia For Caponization In Pigeons

In group A, after surgical preparation, birds were given xylazine injection intramuscularly at dose rate of 16mg per kg body weight. In group B, birds were given detomidine injection intramuscularly at dose rate of 1.4mg per kg body weight. In group C, birds were given ketamine-xylazine injection intramuscularly at the dose rate of 30mg and 8mg per kg body weight respectively. In group D, birds were given ketamine-detomidine injection with the dose rate of 30mg and 0.7mg per kg body weight respectively. While regularly checking reflexes after short intervals of time. After achieving of the surgical stage of anesthesia, the surgical operation was beginning. The data was analyzed by One Way ANOVA followed by multiple comparison tests using Statistical Package for Social Science (SPSS) version 20.0. The purpose of designed research was to compare the efficacy of anaesthesia alone and with the combination of the other anaesthesia during the surgery of the pigeons. Different parameters were used to check the efficiency of the anaesthesia. All birds were thoroughly examined to rule out any other anomaly before study. The surgery was performed at Surgery Section, Department of CMS, UVAS, Lahore for 3 months’ duration (including adaptation and postoperative period).
The verdict based explanations for better and reliable anaesthetic combination used in pigeons and to evaluate safe anaesthetic combination and to find ideal pre anaesthetic with minimum effects. We concluded that detomidine alone can be used just for non-surgical
procedures because it produces the light sedation but superficial analgesia with smooth induction and recovery but xylazine can also be used if detomidine is not available. Xylazine produces superficial analgesia and light sedation but for the short time comparatively. For major and minor surgeries both combinations of ketamine i.e. (K-Xp) and (K-Dp) are more reliable and safe. Both cocktails produce the deep analgesia and anaesthesia as well, with rapid and smooth induction and recovery.

Comparative Efficacy Of Ozone And Gentamicin Sulphate On Uterine Infections In Crossbred Dairy Cows

It is mostly agreed that uterine diseases in the cow after parturition have a negative effect on overall reproductive performance. Ozone which is assumed as a very potent oxidant, is one among these alternative techniques. The advantage of using ozone rather than antibiotics is in lowering the incidence of bacterial resistance in consumers of foodstuffs of animal origin, and other advantage is that ozone has no withdrawal period for milk, meat and other products. Ozone breaks through the microorganism (bacteria and germs) cell membrane, and also destroys viruses by diffusing through the protein coat in the nucleic acid core, resulting in damage of the viral nucleic acid.
The study was conducted on 50 animals which were randomly divided in two groups. Both groups were having 25, 25 animals. Group A received Ozone while group B received gentamicin sulphate intra uterine. Uterine lavage was taken twice, once before applying treatment and second after 8 hours of applying treatment. Samples were cultured for bacteriology to detect E.coli, F. necrophorum, A. pyogenes and St. pyogenes. Number of positive cases in Group A was 12, 9, 10 and 7 for E. coli, F. necrophorum, A. pyogenes and St. pyogenes respectively. Number of positive cases in Group B was 10, 8, 11 and 8 for E. coli, F. necrophorum, A. pyogenes and St. pyogenes respectively. After applying certain biochemical tests for each bacteria, bacteria was confirmed.
Difference of the colony forming units of before and after applying treatments for each bacterium in both groups was calculated. This difference was compared with difference of the colony forming units for same bacteria of other group by using Independent 2 samples T-test with the help of Statistical Packages for Social Sciences (SPSS Inc.) for the windows Version 13.3 (Chicago IL, USA).
Results were interpreted on the basis of level of significance. Differences among the groups were considered significant at P < 0.05. E. coli, F.necrophorum and St. pyogenes were highly significant as the P value for group differences was less than 0.05. Group differences among S.pyogenes showed no significance as the P>0.05.
The results showed that ozone is better in efficacy as compared to gentamicin sulphate on uterine infections in cows.

Study On The Pathogenesis Of Field Isolate Of Salmonella Pullorum In Experimentally Infected Broiler Chicken

to 1.5 million people. As use of modern techniques and medicine has reduced a risk lot but it is
still facing threat from many diseases. One of these is Salmonellosis which is and hatchery borne
infection and it also has a zoonotic importance. This organism is gram negative bacterium, non
acid fast, uniform staining and non spore forming bacillus.
Pullorum disease produced by it is also known as bacillary white diarrhea due to whitish
droppings produced by birds. Day old chicks are affected because it can be transmitted vertically.
Infected fomites and utensils also play a role in the transmission of infection. Growth of S.
pullorum rapidly occurs upon beef agar on nutrient media and growth is best at 37 °C. Selective
media such as selenite, tetrathionate broths and differential media such as MacConkey, bismuth
sulfide and brilliant green agar are best for the proper growth of organism.
The organism was isolated from the 30 different infected samples of broiler birds taken
from different farms and was grown upon the selective media. The colonies were picked from
the cultures and were subjected to PCR for the further process. After confirmation with PCR,
inoculums was prepared with a bacterial load of 2 x 107 (CFU) in 0.5 ml of normal saline at the
ph of 7.2.
Chicks were randomly divided in to three groups as group 1, 2 and 3, each group
containing 30 chicks. Group 1 was infected with field strain of S.pullorum, Group 2 with
vaccinal strain and Group 3 was set as control group and no infection was given to it. Samples
were collected from birds at 10th, 15th, 20th and 25th days of age. Observations were made upon,
clinical signs, histopathological lesions, mortality and morbidity rate, FCR and gross
Clinical signs included were anorexia, depression, huddling, labored breathing, loss of
feed and water intake, ruffled feathers, reduced mean body weights, low FCR and pasty vent.
Gross and histopathological lesions were much observed and were prominent in the birds
affected with field strain of S. pullorum. One Way-ANOVA was used for statistical analysis in
between the groups. Gross pathological lesions included were liver congestion 55%, cheesy
material in caeca 70% in the birds which were affected with field strain of S. pullorum.
Histopathological findings in the birds affected with field strain were monocytes in spleen,
leukocytes in intestine and heterophils in liver. Low FCR was recorded in infected birds of
Group 1 and was 1.63 at 25th day. Mortality rate in the birds affected with field strain was
maximum 43.33% and morbidity rate was 73.33% at the 10th day of age. Birds of group 2 were
less affected and only showed medium level of lesions. No specific lesions were seen in group 3.
The total prevalence of of S.pullorum after conformation with PCR was 20% out of total 30
infected samples. The organs for histopathology taken were liver, spleen, caeca and intestine and
were preserved in 10% formalin prior to the infection.
The overall pathogenesis of experimental bacteria after given through oral route was that
it first localizes in digestive tract from where it enters into the blood stream and invades in the
different organs and tissues at different time intervals producing lesions and immunological

Pathological Studies On Contagiouscaprine Pleuropneumonia In Small Ruminants Of District Dera Ismail Khan, Khyber Pakhtunkhwa

Publisher: 2017 Dissertation note: These experiments presented the pathological studies on Contagious caprine pleuropneumonia in Distract Dera Ismail Khan at Govt. slaughter house through gross pathology, hematology, competitive ELISA, histopathology and molecular identification (PCR). For this purpose the blood and tissue samples were collected from contagious caprine pleuropneumonia suspect at govt slaughter house Dera Ismail Khan. On the basis of clinical examination, the animals were showing the elevated temperature, nasal discharge, painful cough, dyspnea and weakness.
On postmortem examination the lungs showed congestion and marbled appearance (Figure no 4.1). There was straw color pleural fluid in the chest cavity (Figure no 4.2). Total of 50 samples 25 blood and tissue samples of sheep and 25 blood and tissue samples of goat were collected for the different parameters of the study. The hematology was performed at SEENA Lab in Distract Dera Ismail Khan. The hematology data was further compared and analyzed by using statistical analysis (independent t-Test). RBCs values of positive and negative cases of CCPP in goats were 9.50±0.27 and 3.15±0.39respectively. There is significant difference (p<0.05) between positive and negative cases of CCPP in goats regarding RBCs values. WBCs values of positive and negative cases of CCPP in goats were 8.31±0.38 and 17.19±1.22respectively. There is significant difference (p<0.05) between positive and negative cases of CCPP in goats regarding WBCs values. PCV values of positive and negative cases of CCPP in goats were 4.03±.63 and 17.11±1.5respectively. It is characterized by intravascular hemolysis, haemoglobinemia, haemoglobinuria ultimately leading to anemia. The exact pathogenesis is yet unknown as there are many diversified etiological factors have been associated with this disease. All the relevant information is relatively scanty. Consequently present study has been aimed to study all possible risk factors associated with this disease in tehsil Bhalwal of district Sargodha where a large number of increasing cases were reported by the local governmental body. Etiological, hematological and biochemical risk factors were quantified to facilitate control measures and upcoming research priorities.
This study was conducted from the period of about 4 months from November 2016 to February 2017. Cross-sectional epidemiological observations were documented on hemoglobinuric and healthy buffaloes for hematological and biochemical study related to parturient haemoglobinuria. The sample size was determined to three hundred and eighty four animals.Present study was observed during the period of four months (November 2016 to February 2017). Out of 384, forty animals (n=40) were confirmed with post parturient hemoglobinuria. The point prevalence observed during the period of four months was 10.4%.
Buffaloes showing signs of hemoglobinuria along with parturition history, pale mucous membranes, mild tachycardia and dyspnoea was assumed as affected with post-parturient haemoglobinuria while animals suffering from other problems like babesiosis causing red urine were omitted from the study after verification of diagnosis through giemsa staining. The blood samples were processed for haematological analysis for the final confirmation of positive
haemoglobinuric buffaloes. Blood sample collected and placed in EDTA vacutainerswas processed for hematology to study hemoglobin (Hgb) values, total erythrocytes count (TEC), erythrocytes sedimentation rate (ESR) and hematocrit (Hct), total leukocyte count (TLC), mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH) in addition to mean corpuscular hemoglobin concentration (MCHC) by using haematological analyzer. Haematological analysis of all the samples was made from Department of Pathology, UVAS, Lahore.Serum samples of all buffaloes were analyzed for biochemical analysis asalkaline phosphatase (ALP), serum urea, glucose,bilirubin, creatinine, calcium, phosphorus, copper, and molybdenum. Moreover, urinalysis was done for gross and biochemical analysis.
Results of the study revealed significant difference among complete blood count (CBC) includingHgb, TEC, Hct and TLC, ESR, MCV and MCH. However, there was no significant variation among MCHC values in affected buffaloes. Serum biochemistry also revealed significant difference of various parameters including ALP, creatinine,BUN, total bilirubin, phosphorus, copper and molybdenum. However, no significant difference was detected among the healthy and affected groups regarding blood glucose and serum calcium levels. There was significant elevation in pulse and respiration rates in buffaloes affected with hemoglobinuria.
The results regarding mineral analysis of the soil shows significant difference in phosphorus and copper. Moreover, mineral levels of soil and serum of animals showed significant relation of phosphorus levels, followed by the levels of molybdenum. Calcium and copper levels also showed moderate relationship.
Observations regarding parity/lactation number reveal the highest incidence rate of 35% among buffaloes at 3rd lactation, followed by buffaloes at 4th, 2nd, 5th, 1st and 6th lactation, respectively. Milk production showed direct relationship with buffaloes affected with post parturient hemoglobinuria.
From the present study, it is concluded thathemoglobinuria was observed in buffaloes of tehsil Bhalwal may be due to variation of soil composition particularly the deficiency of Phosphorus which may lead to the lysis of erythrocytes and hemoglobinuria through various pathways. However, efficient replenishment of minerals content in fodder producing soil is necessary to overcome the disease in buffaloes affecting from parturient hemoglobinuria in the aforementioned area.

industrialization. Lead poisoning has been recognized as a major public health risk,
particularly in developing countries. It is nephrotoxic, hepatotoxic, neurotoxic, carcinogenic
and mutagenic for animals and human. Sewerage water, fertilizers, leaded-gasoline and lead
based batteries are the sources of lead contamination in soil and forage. The lead particles are
taken up by animals from contaminated forages and excreted in animal products like milk and
The presence of Pb in drinking water, waste water, plant products and animal
products has been studied which is a serious risk for animal and public health. The
legislations for the disposal of household wastes and industrial effluents are very poor in
Pakistan. The calculation of safe Pb levels in different products is still to be needed.
Pathological effects of higher Pb levels have not been studied in Pakistan. The present study
was aimed to unveil the toxic effects at constant dose of Pb over a period of three months in a
local sheep breed of Pakistan. The status of Pb toxicity was also investigated in a polluted
area around sewage drain and mutton slaughter house at District Jhang, Pakistan.
The Pb concentration in soil, forage and irrigating water was found to be below the
permissible limits and was safe for agriculture but long-term ingestion of low Pb
concentration may have cumulative effect. The serum Pb concentration was found to be
above the recommended safe limits for producing Pb toxicity in animals. The different tissues
like kidney, liver and skeletal muscles also contained higher Pb level from the permissible
limits and found to be unsafe for public use. Kidney showed the highest Pb concentration and
the muscle contained the least Pb level in the present study.
The erythrocyte count, hemoglobin concentration and packed cell volume showed
inverse correlation with Pb concentration and mean values were below the normal range in
Pb treated sheep but anemia was not developed. The erythrocyte sedimentation rate was also
influenced by given dose of lead acetate during third month of treatment. The white blood
cells also revealed no effect on given dose of lead acetate in Lohi sheep in this study.
The biochemical parameters of field and treatment group showed higher concentration
as compared to control group of Lohi sheep but their means were falling within the normal
range of reference values. The disturbed biochemical parameters in apparently healthy sheep
with higher serum Pb concentration were indicative for liver and kidney damage.
Lohi sheep exhibited less effect on given dose of lead acetate during first two months
but more pronounced changes of chronic Pb toxicity were observed during last month of trial.
The histological changes were not observed on early period in lead acetated treated sheep.
The characteristic histological changes were observed on last slaughtering at day 90 in kidney
and liver including degeneration and focal areas of necrosis, dilatation of blood vessels with
accumulation of red blood cells and fibrosis in some areas. The nuclear changes were more
typical with intranuclear inclusion bodies in renal tubular epithelial cells but less
distinguishable in hepatocytes.
It was concluded that soil, forage and water contained low Pb levels in the study area.
The ingestion of low Pb level for longer period had cumulative effect in animals. The animals
might be resistant to low Pb level but their products are a severe risk for public health. So the
necessary measures should be adopted to minimize the heavy metal contamination in animal

It is an economically important viral disease of sheep and goats causing varying degree of morbidity and mortality in susceptible animals which may be as high as 100 and 90 per cent, respectively. PPR is responsible for serious socioeconomic problems. There is no data available regarding pathogenesis and field virus characterization to compare it with vaccinal strain for any difference. Nigella sativa(Black Seed) has antiviral activity against many viruses. Therefore present studywas undertaken to investigate the antiviral effect of Black Seed in vivo and in vitro against PPR virus. Further more time course detection of virus is still needed to be studied.
Nigella sativa (Black seed) has antiviral activity against PPR virus.
Pathogenesis can better be studied through histopathology, necropcy findings and morphometric changes.
A total of 250 clinically positive samples suspected for PPR virus were included in the study. Samples were consisted of nasal, ocular and anal swabs; whole blood in EDTA were collected from suspected animals. In case of mortality morbid material included lungs, liver, spleen and mysenteric lymph nodes were included in the study. Samples were subjected to immune capture Elisa for detection of viral antigen in suspected samples. Samples which found positive foe IC – Elisa were then subjected to RT-PCR for confirmation of virus. After confirmation of virus through IC – Elisa and RT-PCR the positive samples were subjected to virus isolation on vero cell. After isolation of virus, the TCID 50 of the virus was calculated for preparation of inoculum for further use. In this experiment mesenteric lymph nodes and spleen found to be major organ for isolation of PPRV.RT-PCR found to be most reliable and confirmatory diagnostic test for PPRV. Field Virus adaptation on vero cells found to be difficult to optimize.
In this experiment antiviral activity of black seed was checked on vero cells infected with PPRV. Three extracts of N. Sativa were prepared to check the in vitro antiviral activity of black seed. In this study poly saccharides extracted from black seed found to be more effective against PPRV. Adaptation of field virus was done on Vero cell line. Antiviral activity of Black Seed extract was determined in vitro on Vero cell on bases of CPE (Cytopathic effect). The ethanolic and aqueous extract were found to be more toxic to consistency of monolayer of vero cells. The TCID50 of virus was calculated after treating cells with different extracts. In this study poly saccharides extract exhibit lower TCID50‘s as compared to ethanolic and aqueous extract which showed higher TCID50’s.So less cytopethic effect was observed in vero cells treated with black seed extracts. Antiviral activity was determined on base of CPE.
Pathogenesis of virus in natural host was studied through time course detection of virus in body secretions, blood, organs. Histopathological changes were studied.20 goats were procured from market divided into four groups (n=5) A,B,C and D. In animals of group A prophylactic effect of N.Sativa was studied. In group B complete pathogenesis of PPR virus was studied without any prophylactic or therapeutic measure. In group C therapeutic effect of N. Sativa was studied after onset of clinical picture of disease. At the end of this experiment, clinical picture, gross pathology, histopathology, and morphometric changes revealed that N. Sativa has noticeable prophylactic effect on PPR infected goats. It can be used as a therapeutic agent in PPR infected goats but it can’t control pathological effect of virus after onset of infection.
Data collected were statistically analyzed by using Microsoft Excel (Microsoft Excel, 2007) and SPSS (for Windows, Version 16.0). The data were put the descriptive analysis and Chi square test was employed to test the significance and test of hypotheses
It was concluded that Black Seed therapy possessed marvelous prophylective effect against PPR virus and RT-PCR was the most efficient methodology to confirm the virus.

Comparative Efficacy Of Advanced Surgical Hemostats In Rabbit Spleen

abdomen. Splenic accidents such as spleen rupture and splenic hemangiosarcoma, are most
common cause of intra-abdominal hemorrhage in dogs. When an incision is given on spleen there
is always a high risk of bleeding. In severe cases animal often leads to death due to massive blood
loss and hypovolemic shock. The objective of study was to check the efficacy of latest and
advanced surgical hemostats i.e., Surgicel Original and Surgicel Snow to control bleeding when
spleen is severed either accidentally or surgically. These hemostats are affordable, safe, reliable
and convenient. For this, Rabbits were arranged in three groups. Group A consisted of 4 Rabbits
on which Surgical Gauz was used. In Group B Surgicel Original was used. On rabbits of Group C
Surgicel Snow was used. Hemostasis time of all the groups was monitored and number of
leucocytes, erythrocytes, platelets AST, ALT and ALP were evaluated. The results of present study
clearly indicated that Surgicel Snow is better choice to control hemorrhage than Surgicel Original.
Therefore, when ligation and other conventional methods of controlling blood stand impractical,
Surgicel should be used to control arterial, venous and capillaries bleeding during surgery.
On the basis of findings of this study, it has been concluded that Surgicel Snow is better hemostat
than Surgical Original. Surgicel Snow yields ideal stoppage of hemorrhage and does not impair
vital function of spleen.

Comparative Pathological Association Of Slc11a1 (Nramp1) Gene With Susceptibility And Resistance To Brucellosis In Local Pakistani Buffaloes

For water buffaloes Brucellosis is a pricey disease (Bubalus bubalis). In Pakistan 5 known breeds of buffalos are; Ravi, Nili, Kundi Nili-Ravi and Aza Kheli. The study was conducted on 200 animals of two breeds including Nilli-Ravi and Kundi around farms of Punjab. 200 serum samples from two breeds were screened for brucellosis. Dormant infections and lengthy incubation of the pathogens bounded the effectiveness of programs based upon the elimination of infected animals. As comparative genomics is playing a pivotal role in the genetic dissection of complex traits such as infectious diseases resistance using this information we can oppressed for disease resistance with genetic choice as a method to the regulator of brucellosis in our local breeds such as Nili-Ravi buffalo. Blood sample (5mL) was collected in EDTA vacutainers from each animal. Serum from blood samples were collected in 3 mL eppendorf tubes with the help of sterile pipettes. Serum samples were screened out for brucellosis by Rose Bengal Plate Tetst (RBPT). Rose Bengal Antigen was purchased from Veterinary Research Institute (VRI), Lahore. The antigen was stored at 40C in refrigeration during the study according to manufactures recommendation. DNA quantification was performed on “Thermo Scientific NanoDrop spectrophotometer ND-2000” and by running extracted DNA on 0.8% agarose gel. Nanodrop spectrophotometer provides direct and easy measurements within a 5 second only by using just a pipette and wipe. PCR for amplification was done with a total volume of 20μl by adding primer pair and extracted DNA to the PCR master mix in following concentrations. 2μl of forward and reverse primer was taken respectively. 4 μl of PCR grade water was added and DNA was taken in 2 μl
Quantity. The total volume of master mix obtained was 10 μl. The thermocycler (Bio-Rad, CFX ™, Singapore) was set with the conditions for the total of 35 cycles of PCR. Initial denaturation of DNA samples were performed at 940 C for 5minutes. Then in 2nd cycle denaturation was performed at 94 0C for 30 seconds. Annealing step was performed at 63 0C for 30 second. Then, initial extension of DNA samples was conducted at 720 C for 30 second. Finally, extension was performed at 720 C for 5 minutes to get results. The PCR product was confirmed by running the PCR product on 1.5 % Agarose gel electrophoresis was performed at 110 Volts for 30 minutes. In Nilli-Ravi, among 12 positive samples by PCR, 08 were found resistance to brucellosis through sequencing study of Nramp 1 BB, only one sample was found to be susceptible to brucellosis through genotypic study of Nramp1 AA and 3 samples were found to be neutral may be susceptible or resistance to brucellosis. In Kundi, among 38 positive samples by PCR, 03 were found resistance to brucellosis through sequencing study of Nramp 1 BB, and 6 sample were found to be susceptible to brucellosis through genotypic study of Nramp1 AA and 29 samples were found to be neutral may be susceptible or resistance to brucellosis. This research clearly demonstrates that Kundi is more susceptible to brucellosis then Nilli-Ravi. Conclusion: We concluded that Nilli-Ravi is more resistant to brucellosis as compare to Kundi breed Natural resistance against brucellosis in cattle is linked to the Nramp 1 gene. Polymorphisms at the bovine Nramp 1 3/ untranslated region (3 UTR) are associated with natural resistance against brucellosis.
