One way of enhancing protein supply is to expand poultry production along with increasing production of other micro livestock such as Japanese quail (Coturnix coturnix japonica) having low maintenance cost, short generation intervals, early sexual maturity and better resistance to diseases and its meat being rich in high quality protein having high biological value with low caloric content. Profit can be optimized by minimizing feed cost that accounts for 60-70 % of the total production cost and any improvements in the performance of birds by manipulation of feeding strategies inevitably have a profound effect on profitability. Any effort to improve commercial poultry production and enhance its efficiency needs to emphasize on better utilization of existing resources. Among different feeding management schemes and strategies phase feeding may be employed with the logic seems to feed birds for shorter periods of time to exactly meet but not exceed the amino acids requirements hence improvement in carcass characteristics and reduction of dietary cost. Commercial availability of very vital limiting amino acids (lysine) has set a new tendency of formulation of poultry feeds having low protein level with addition of amino acids. Lysine, being utmost essential amino acid is used as a reference for other essential amino acids. Feed restriction program may be another managemental tool that may elicit compensatory growth, improved feed efficiency, carcass quality and birds are not exposed to sub optimal level of nutrients but the efficiency of utilization of these nutrients may be improved. On the other hand breed, strain, management and sex differences for carcass traits have also been reported. Very little research focus on the subject has necessitated conducting the ABSTRACT vii present study undertaken in Japanese quails on the similar pattern as adopted in broiler industry to make quail production more cost-effective and commercially viable at Avian Research and Training (ART) Centre, University of Veterinary and Animal Sciences, Lahore, Pakistan. A series of experiments at Avian Research and Training (ART) Centre, University of Veterinary and Animal Sciences, Lahore, Pakistan was run to assess the effect of different management interventions on growth performance, carcass characteristics and blood biochemical profile in Japanese quail. The first experiment was aimed to examine the growth performance and economic efficiency involving 1440 day-old Japanese quail (Coturnix coturnix japonica) chicks. Three dietary lysine levels (1.3, 1.4-1.2 & 1.5-1.3-1.1 %) in 3 different phases were allocated to four different close-bred stocks (Imported, Local-1, Local-2 and Local-3) of Japanese quails to assess their comparative growth performance by replicating each treatment for three times. The experimental day-old quail chicks were randomly divided into 36 experimental units of 40 chicks each. Quails under 1st treatment were fed a diet with 1.3 percent lysine throughout the grow-out period of 28 days, while, those under 2nd treatment were allotted diet with 1.4 percent lysine up to14 days of age and then subsequently reduced to 1.2 percent lysine up to 28 days. The 3rd treatment was split into 03 different phases. The first phase was up to 9th, 2nd up to 19th and 3rd up to 28th day by allotting diet containing 1.5, 1.3 and 1.1 % lysine, respectively. Weekly data on growth performance were recorded and analyzed through ANOVA technique in CRD under factorial arrangement and the comparison of means was worked out using DMR test by the help of SAS 9.1. Maximum (P≤0.05) feed intake; body weight gain and improved FCR were observed in three phase dietary lysine regimen leading to maximum profit margins. viii In the 2nd experiment same experimental design and phase feeding was practiced to observe organ development. Sexing with in treatment was done at the age of three weeks and quails were maintained separately for one week. At 4 week of age, 3 birds/ replicate from either sex were slaughtered through Halal Muslim method for studying carcass characteristics. Two birds per replicate from either sex were used for serum analysis of glucose, cholesterol, urea, albumen and total protein using standard procedures. The analysis showed three phase dietary lysine regimen than other dietary lysine regimens improved (P≤0.05) slaughter characteristics i.e. post slaughter weight (g), dressing percentage with and without giblets, breast yield (g), thigh yield (g), giblet weight (g), liver weight (g), keel length (cm), shank length (cm), weight of visceral organs including intestinal weight (g) and intestinal length (cm). However, heart weight (g), gizzard (empty) weight (g), serum glucose, cholesterol, urea, albumin and total protein were not significantly affected by dietary lysine regimen. While, different close bred stocks did not show any significant differences. Third experiment was executed to examine the growth performance and economic efficiency of Japanese quail (Coturnix coturnix japonica) subjected to different feed restriction regimes at ART Centre, UVAS, Lahore. For this purpose a total of 3200 quail chicks from four different close-bred stocks were allocated to four different feed restriction regimes comprising four close-bred stocks (Imported, Local-1, Local -2 and Local-3) at the age of 10 days. The experimental quails in group 1 were fed ad-libitum (20.30% CP, 1.3% Lysine, as recommended by NRC) throughout the experimental period to serve as control while groups 2, 3 and 4 were provided with 1 hour feed- 3-hour off, 2-hour feed- 2hour off and 3-hour feed-1hour off feeding regimes, respectively. The analysis of data showed that the maximum feed intake was observed in ad-libitum fed group whereas the highest body weight gain was observed in ad-libitum and 3 hour ix fed quails. The best FCR leading to maximum profit margin was observed in 3 hour-fed group. Different close-bred stocks could not express any significant difference in growth parameters. In the 4th experiment same dietary plan of time restriction as in 3rd experiment was adopted to observe organ development. At the termination of the experiment (at the age of 38 days), 6 birds (3 male and 3 female) from each replicate were randomly picked up and slaughtered (by Halal method) to study different slaughter parameters. Significantly higher (P≤0.05) carcass weight, mean dressing % with and without giblet, mean thigh weight was observed in ad-libitum and 3 hours fed quails while significantly lower mean dressing %, liver weight, gizzard weight, giblet weight, breast weight and mean intestinal length and weight in one hour fed quail. Blood profile showed significantly higher (P≤0.05) serum glucose, urea, albumin and total protein level in ad-libitum and 3-hours fed quails while significantly higher (P≤0.05) serum cholesterol level was observed in one hour fed quails. Heart weights (g), keel length (cm), shank length (cm) were not affected significantly among different treatments and close-bred stocks. Conclusion Based upon the findings of the present study it may be stated that 1. Maximum (P≤0.05) feed intake; body weight gain and improved FCR were observed in three phase dietary lysine regimen leading to maximum profit margins. 2. Significant improvement in carcass characteristics was recorded in three phase dietary lysine regimen. 3. The best FCR leading to maximum profit margin was observed in 3 hour-fed group in Japanese quails when subjected to different feed restriction regimens. x 4. Three hour fed quails showed superior carcass characteristics at par with ad-libitum fed groups especially in terms of carcass weight, dressing percentage and thigh weight. 5. Significantly higher (P≤0.05) serum glucose, urea, albumin and total protein level were recorded in ad-libitum and 3-hours fed quails while significantly higher (P≤0.05) serum cholesterol level was observed in one hour fed quails. Suggestions and Recommendations Four lysine dietary regimens having 1 week each may successfully be employed in Japanese quails in order to get maximum profit. It may further be recommended that Japanese quails may be subjected to feed restriction of 1-hour after 2nd week. The present series of experimentation is a step towards optimizing the nutritional and managemental strategies in Japanese quails, however, a lot more is still needed to be worked out in this direction. Histatin is only antimicrobial peptides present in primates like monkey chimpanzees and human. It’s also contain anti-fungal as well as anti-bacterial effect against staphylococcus aureus. In this study showed the effect of human salivary Histatin against staphylococcus aureus. Salivary Histatin may show antibacterial activity against Staphylococcus aureus with its efficiency having link to age, gender and diabetic status. Total sixty-four saliva sample will be collected on the basis of gender, age and presence of diabetic status. The antibacterial activity of saliva was observed against Staphylococcus aureus by disc-diffusion method. DNA was extracted and HTN1 gene was amplified using specific primer. This study was helpful in demonstrating antimicrobial ability of Histatin proteins present in human saliva. It also provide insight with regards to age, sex and/or immunocompromising ailment (in this case, diabetes) having an effect on the ability of these proteins, thus, opening new doors when it comes to combating fungal infections in both human and animal subjects. The HTN1 gene sequenced and BLAST results proof that variation in Histatin anti-bacterial property in diabetic patients and was not due to mutation in the nucleotide sequences of the decreases in salivary Histatin was due to other reason not due to mutation in these individuals. The age bases study HTN1 gene BLAST results was found 99% similarity with the other age groups. Summary 40 The statistical analysis of healthy people with age and zone of inhibition was found ANOVA P<0.000. The increases of age will decreases the salivary Histatin anti-bacterial properties. The optimum antibacterial activity was measured 2cm in diameter. The present results indicated that healthy human saliva possess antibacterial ability against Staphylococcus aureus. Mutation in NS4B encoding gene may be targeted to develop an effective vaccine against HCV before virus invasion in host immune system. Serum was collected from 20 patients who are infected with HCV. RNA was isolated from these samples to reverse-transcribe it into cDNA. This cDNA were PCR-amplified and amplicons were detected by UV light after their separation through agrose gel electrophoresis followed by seguencing of NS4B encoding gene. Variations in HCV NS4B gene was analyzed by using Mega 6 software and sequence 2 blast analysis NCBI tool. Protein modeling was performed with phyre 2, DIANNA tool and I-Tasser online server and quality of model and antigenicity was checked by Vaxajen V 2.0 and Epijen softwares. Conservative analysis and epitope mapping were performed by using Immune Epitope Database (IEDB). A systematic approach was employed for the prediction of potential epitopes in NS4B protein. Vaxijen V 2.0 was used to determine overall antigenicity of NS4B using cut off value of 0.4. Values above this threshold level show probable antigens in the protein. The topology of NS4B was determined by TMHMM server with the help of membrane topology data. Regions outside membrane and transmembrane were eradicated for epitopes prediction. T cell epitopes propred was used with proteosome cleavage site filter of 4% threshold. From the following data analysis 12 to 15 possible T-cell epitopes can be predicted; only these T cell epitopes were included in conservation analysis. The present study provides information about mutational changes in HCV protein NS4B and thus preliminary data for the development of vaccine against HCV. This can also lead to future prospects for diagnosis, treatment and prognosis of HCV. This study was designed to develop a suitable semen extender for cryopreservation of Nili Ravi buffalo semen that can improve conception rate in buffaloes. Experiment-I, an attempt was made to develop semen extender with optimal osmotic pressure for buffalo semen using tris citric acid (TCAE), skim milk (SME) and coconut water (CWE) extenders (each extender have 260, 270, 280, 290 and 300 mOsm/kg osmotic pressure levels). In Experiment-II, best extender (TCAE: 300 mOsm/kg) of experiment-I was tried to improve post thaw spermatozoa characteristics by supplementing antioxidants [0.0, 1.75, 2.0 and 2.25 mM butylated hydroxy toluene (BHT) and 0.0, 2.0, 5.0 and 8.0 mM L-cysteine]. Post thaw spermatozoa motility, viability, plasma membrane integrity (PMI), DNA damage rate and lipid peroxidation were assessed in first two experiments. In Experiment-III, pregnancy rate assessment of extended semen was carried out by using Trial extender (best of experiment II) or Control extender of Semen Production Unit (SPU), Qadirabad, Pakistan (50 inseminations of each extender). Higher spermatozoa motility at ≥ 270 mOsm/kg was noted in TCAE than both SME and CWE could be due to less intracellular ice formation in zwitterions extender. Higher spermatozoa viability in TCAE and CWE compared to SME may be attributed to extender effectiveness. Higher acrosomal integrity rate at 300 mOsm/kg in TCAE and SME may be because of less intracellular ice formation in isotonic extenders. At 290 mOsm/kg, higher spermatozoa PMI in SME and lesser DNA damage in three extenders might be due to lesser intracellular ice formation at cryopreservation. Decreased spermatozoa DNA damage in SME might be due to the presence of natural antioxidants i.e., casein. Higher lipid peroxidation in CWE than TCAE and SME may be due to presence of natural antioxidants (in SME) and higher cell dehydration potential of TCAE. Higher spermatozoa motility recorded at 2.0 mM BHT compared to other BHT groups including DMSO might be due to fact that BHT protects spermatozoa mitochondria by reducing oxidative stress. Lower spermatozoa viability, PMI rates and higher DNA damage at 2.25 mM of BHT may be due to BHT toxic effects. Lower lipid peroxidation in BHT treated groups compared to DMSO and BHT control groups might be related to BHT strong antioxidant properties. L-cysteine caused higher spermatozoa DNA damage at highest level (i.e., 8 mM) that could also be due to antioxidant’s toxic effect. Pregnancy rate 18 % higher was noted in Trial than Control semen extender; however no significant difference have been noted that might be due to less no of inseminations. In conclusion, TCA extender (300 mOsm/kg) having BHT (2.0 mM) improved post thaw semen quality and yielded numerically better pregnancy rates. Results of study indicated that osmotic stress damaged the spermatozoa internal structures more severely than injury to plasma membrane. Omega-3 fortified eggs might be helpful in decreasing oxidative stress in healthy human subjects. Efficacy study of omega-3 fortified eggs was conducted against oxidative stress through biochemical analysis of biomarkers of oxidative stress e.g., catalase and superoxide dismutase.Eighteen volunteers were selected and divided in three groups (control, omega-3 supplements + vitamin E and omega-3 fortified eggs). At 0, 30th and 60th day, blood sample was collected and further examined. The data will be analyzed through Analysis of Variance with repeated measures (rANOVA) technique. The significance level will be defined as P ≤0.05 and means will be compared through Duncan’s Multiple Range test for significance difference. Omega-3 bio fortified eggs that would be useful for decreasing the oxidative stress in healthy human subjects.P value is less than 0.05 and results indicate that omega 3 fortified eggs may helpful in decreasing oxidative stress in humans. Also results showed that omega 3 fortified eggs are helpful in improving lipid profile by increasing HDL levels and decreasing LDL levels, decrease in triglyceride and cholesterol also a plus point of this product as it is economic, easy to access and nutritious. Widespread household food insecurity (58.1% households are food insecure) and poor knowledge about nutrient requirements in pregnant and lactating women are the major cause of nutritional deficiencies. This study is planned to assess nutrients’ intake among pregnant and lactating women and to compare with standard requirements. A cross sectional survey design was used to assess the nutrients’ intake of 110 pregnant and 110 lactating women from 6 Union Councils (two urban, two per-urban and two rural Union Councils) of Lahore and Rajanpur districts. Nutrients’ intake was assessed by using a pre-tested structured questionnaire to interview the pregnant and lactating women at one point in time. Weight, height and mid upper arm circumference was measured through anthropometric measures, 24 hour dietary recall, and food frequency was used to assess nutrients’ intake. The data was analyzed bychi-square and cross tabs using SPSS version 20.0 and was presented in percentage, frequencies and charts to assess the nutritional deficiencies.Significance level was defined as p≤ 0.05. The results shows that majority of the women were deficient in energy and nutrient's intake through daily diet. In both districts pregnant and lactating women were not consuming their required amount of various nutrient's according to their maternal status. Results show significant difference in Lahore and Rajanpur districts most of the nutrient's deficiency was present in Rajanpur as compare to Lahore. Severely low dietary intake of various nutrients was calculated and analyzed. According to the results dietary intake of various nutrients was severely low in selected women with following percentage protein was 39.1%, folate 40%, B12 20.5%, B6 74.5%, vitamin C 30.5%, vitamin A 14.1%, vitamin D 69.1%, calcium 26.8%, iron 96.8% and zinc 70.9%. These results shows very adverse situation of important nutrient's intake of pregnant and lactating women through diet. Heavy metals are presentin sweets like candies, chocolates and gums, which are favorite food for children and pregnant women. Lead, Nickel and Chromium are major toxic heavy metals. Main source of lead exposure in children are food, air, water and soil.Accumulation of Lead in different parts of the body has adverse effects and causes many diseases.When chromium is ingested in excess amounts, it induces many toxicity symptoms in human body. At higher levels, nickel accumulates in the lungs and may cause bronchial haemorrhage. Other symptoms include nausea, weakness, dizziness, etc. In Pakistan food safety is on the verge chaos specially there is no sufficient data available on heavy metals in confectionary products. Therefore present study was designed to aware the consumer specially children about hazards of heavy metal in candies and chocolates. Both Local and imported Candies and Chocolates samples (n=120) were collected from local shops anddepartmental stores of Lahore. Samples were analyzed by Atomic absorption spectrometer for heavy metals examination in Environmental Science DepartmentLaboratory of University of Veterinary and Animal Sciences. Concentration of lead in imported candies (n=30) and local chocolates (n=30) found within acceptable value 0.5 mg/kg of Punjab Food Rules 2011(PFR) while imported chocolates (n=30) and local candies (n=30) were not found within acceptable value 0.5 mg/kg of PFR 2011. The variation of nickel was not found within permissible value 0.025 mg/kg of PFA in all (n=120) imported chocolates, local chocolates, imported candies and local candies. By which, lipid metabolism also effects and causes atherosclerosis in vessels which trigger the blockage of blood flow and induces the imbalance for the supply of oxygen to the heart. So this atherosclerosis directly involves in addition of risk factor for coronary heart disease. Preferable study was made to highlight effect of CETP gene at molecular level by comparing control group with the selected patients having coronary heart disease. This study was appreciably made possible by targeting two reported polymorphisms, one in the intron 1 region Taq IB (rs708272) and on exon 14 region I405V (rs5882) of this CETP gene. The study was relatively speculated by the extraction of genomic DNA from all selected blood samples. By selecting two primers, certain segments were amplified for both rs708272 and rs5882 polymorphisms. Analysis of allelic frequencies distribution was calculated by Hardy Weinberg Equilibrium which showed no significance among control and CHD group and there was no association was analyzed in our population by using Fisher’s Exact Test. This is because of small number of samples studied in our population. But maximize concentrations of lipid parameters such as TC, LDL and TG with minimum variation in HDL-C concentration in CHD group as compared to control group that showed the effect of these polymorphisms on the activity of CETP gene with coronary heart disease. It has rapidly increasing demand and competition in national and international markets. Milk consumers in Pakistan often face low-quality, lack of hygiene and absence of cold chains as primary contributors to this low quality. Milk and dairy products also become contaminated during manufacturing and packaging processes. The consumption of low quality milk may cause milk borne diseases. Not only the bacteria but the presence of their enzymes also can cause deterioration of the milk quality. The heat stable enzymes can cause the spoilage of commercial UHT products without presence of any viable count. Aflatoxin (M1) is metabolite of AFB1 in milk. It causes chronic diseases and immunosuppression in children. It is found carcinogenic and cytotoxic in nature. In this project microbial load, free amino acid estimation to predict any protease activity and Aflatoxin M1 were studied in both UHT and Raw milk samples (n=15) procured from local markets of Lahore. Three UHT brands A, B and C were purchased. The UHT milk was studied for microbial growth and protease activity at purchase and at expiry of the products. The microbial load was evaluated by testing of milk for Total viable count, Coliforms, Yeast and Molds, Anaerobic Clostridia and Bacillus cereus in both raw and UHT milk. Protease activity was estimated by assessing the free amino acid by using ninhydrin assay while the Aflatoxin M1 was detected through High performance liquid chromatography. SUMMARY 80 CONCLUSION: It was found that the locally available raw milk quality was poor for consumption and dairy processing for safe and stable milk products. UHT milk quality was found better with low microbial load. Protease activity with reference to free amino acid was detected in raw milk which is indication of the poor milk storage conditions, cold chain maintenance and unavailability of fresh milk. Protease activity was also found in UHT milk and an increase in free amino acid which could be due to heat stable proteases active during shelf life of the milk brands. Aflatoxin M1 was found in majority of raw milk sample which showed the poor animal feed storage and monitoring system. Aflatoxin M1 was also found in some samples of UHT brands with high concentration which depicted that AFM1 was heat stable and it retained in the commercial UHT products as well. It can also be called biomass, bioprotein or microbial protein. Besides high protein content (about 60-82% of dry cell weight), SCP also contains fats, carbohydrates, nucleic acids, vitamins and minerals. Fermentation media containing grinded banana peel as substrate was used to check the production of single cell protein for the selected Arachniotus sp. Different parameters were optimized for higher production of SCP e.g: Incubation period, pH, volume of inoculum, carbohydrate source, concentration of corn steep liquor and ionic salts concentration. The biomass yield was estimated for total protein content by Lowrymethod. Biomass produced from fermentation was used for biological evaluation in feed trials of broiler chicks. It is found that Arachniotus sp gave maximum single cell protein 7.49 g/L using 10 g banana peels at 72 hours incubation period. And protein concentration increased 7.58 g/L by optimizing volume of inoculum 2ml. It is observed in present study carbohydrate source also increases the protein concentration 8.41 g/L when carbohydrate source was optimized (glucose 3%). Later on it was found that nitrogen source also enhance the protein production upto 12.61 g/L by using 2% corn steep liquor. Results also revealed that ionic salt concentration also play important role in the production of biomass protein, addition of 0.075% CaCl2.H2O produced 14.45 g/L single cell protein using above mentioned optimized conditions. 0.050 % K2HPO4 produced 15.06 g/L. Addition of 0.050% MgSO4.7H2O produced maximum protein 15.86 g/L. Biological evaluation in broiler chicks of this biomass protein shown there is no deleterious effects on weight gain, feed conversion ratio, protein efficiency ratio and net protein utilization. Maximum weight gain observed 215.6 grams in the group (C) in which 50% sunflower meal was replaced with biomass protein. Feed conversion ratio in group (C) was 2.64 in which 50% sunflower meal was replaced by biomass protein and in group (B) was 2.51 in which 25% sunflower meal was replaced. And in control group (A) feed conversion ratio was 2.41. Protein efficiency ratio was observed with non-significant value. And same results were shown by Chaves et al (1988) who reported non-significant differences among the standard and test diet when Chaetominumcellulolyticum biomass was fed to chicks. Net protein utilization observed in present study gave significant P value among the groups. So it is concluded that single cell protein produced by this method is cheap and can be used in the food industry as food supplements and can also be included in poultry feed. The study findings suggested that microbial biomass produced by Arachniotus sp using banana peels as substrate can be replaced upto 50% of the protein supply by sunflower meal without any deleterious effects on growing broiler chicks. Keeping in view the importance of foal diarrhea and its impact on the equine industry the current study was designed to determine serum concentration of electrolytes (Ca, K, Na) and trace elements (Li, Cu, Fe) along with the associated risk factors in diarrheic foals. The present study was designed to examine diarrhea in foals at various equine stud farms, private and public veterinary hospitals. For this purpose a total of (n=100 foals) suffering from clinical diarrhea irrespective of the cause of diarrhea selected randomly after clinical examination and (n=10 foals) negative for diarrhea as control were included. Data of each diarrheic foal was collected separately in a data capture form. Blood and serum sample were collected for hematological and serum biochemical studies, respectively. Concentrations of serum electrolytes (Ca, Na, and K) were analyzed by Flame Photometry and trace elements (Cu, Li, and Fe) were estimated through Atomic Absorption spectrophotometer. PCV was determined by micro hematocrit method. Risk factors including species and age were analyzed for association. Foals which were suffered from diarrhea showed increase in Pack cell volume. Serum concentration of Sodium, potassium, calcium, iron, copper and lithium were measured in diarrheic foals by atomic absorption and flame photometer method and were compared with healthy group of foals.Foals having diarrhea showed an acid baseimbalance as a result decrease in serum electrolytes and trace elements. The blood acid-base balance is precisely controlled because even a minor deviation from the normal range can severely affect many organs. The body uses different mechanisms to control the blood's acid-base balance.Sodium and potassium and calcium are important electrolytes imbalance in diarrhea. Potassium level was increased in foals having diarrhea. Calcium concentration in ppm significantly decreased from its normal value. In some diarrheic foals increased in level of calcium was also observed because these foals have blood in their feces that result in the calcium to come in blood as a clotting factor. Trace elements such as Cu, Fe and Li are needed in a very little concentration for many physiological functions. The serum iron was measured in foals having diarrhea.The iron concentration was less than normal value.The deficiency of iron lead to anemia. Clinical signs of anemia because of irondeficiency are rarely seen in foals. Therefore, mineral imbalances influence iron metabolism and may cause iron deficiency in foals. The serum copper concentration was significantly decreased in the diarrheic foals. Copper concentration is also related to the packed cell volume as in anemia copper deficiency is a significant feature. Copper is important element and its deficiency lead to decrease in serum concentration of iron. This decreased level might be because most of the circulating copper in plasma is attached to the serum glycoprotein, ceruloplasmin. Ceruloplasmin has ferroxidase activity and may be required to deliver iron into the circulation Therefore it was observed that low iron concentrations might have resulted from copper deficiency.The concentration of Lithium was also decreased in diarrheic foals when compared to healthy foals. Brucella abortus is the principal cause of brucellosis in cattle and is shed from the infected animal at or around the time of calving or abortion. The present study was conducted on 450 animals on three different strains/breeds of cattle i.e. Exotic (150), Cross-bred (150) and local cattle (150) from 10 different privately owned livestock farms of varying holdings of district Lahore. An epidemiological questionnaire focusing on herd traits as well as husbandry and sanitary practices that could be associated with the risk of Brucellosis infection was completed. Serum samples were collected and analyzed using Rose Bengal Plate Test (RBPT). The serum samples positive for Brucellosis through RBPT further subjected to Serum Agglutination Test (SAT). To check the effect of Brucellosis on hemogram, blood samples from 18 cattle (n=6 indigenous; n=6 cross-bred; n=6 exotic) positive for Brucellosis and 18 animals (n=6 indigenous; n=6 cross-bred; n=6 exotic) negative for brucellosis were collected and processed for TLC, DLC, RBC, Hb, MCV, MCHC MCH and platelets using automated haematology analysed at UDL, UVAS, Lahore. Similarly, to see the effect of Brucellosis on Serum biochemistry, serum samples from 18 cattle (n=6 indigenous; n=6 cross-bred; n=6 exotic) positive for Brucellosis and 18 animals (n=6 indigenous; n=6 cross-bred; n=6 exotic) negative for brucellosis collected and analysed for glucose, total protein, albumin, Creatinine, Alanine Aminotransferase (ALT), Aspartate Aminotranferase (AST) and Sorbitol Dehydrogenase (SD) using commercially available kits. Summary 62 RBPT revealed overall prevalence 17.7% higher than SAT 10.6%. Prevalence of brucellosis is higher in Cross-Bred (22.7%) followed by local cattle (18.9%) and exotic (12%). Hemato-boichemical results showed that increase in TLC, MCV While slight changes in Hb, MCHC, RBC and values of MCV stays within normal range. Human Brucellosis caused by B. abortus, B. abortus are small, non-motile, aerobic, facultative intracellular, Gram-negative cocobacilli. The symptoms of human brucellosis are undulant fever, headache, weakness, body pain, and sometimes endocarditis, orchitis, or arthritis may develop. Prolonged use of combination of antibiotics and human vaccine is main control measure strategies for human brucellosis. This disease is under investigated and hence people are ignorant of this insidious problem. Serological surveillance is a good and cheap tool to diagnose this problem. There is also need to know that which diagnostic antigen (Pakistani or French) is more sensitive and specific from disease eradication and control point of view. Unfortunately annual incidence of brucellosis in whole Pakistan is unknown but they are expected above 2 per 100,000 people. The hypothesis of present study was that brucellosis is considered to be endemic in Pakistan but there is no data available about seroprevalence of brucellosis in pregnant, aborted, rural and urban women with different risk factors in Pakistan. In present study sero-screening of total 199 serum samples of pregnant and women presenting with abortions from district Jhang, district Chinoit and district Faisalabad. Test was performed on 199 samples which showed 40 (20%) positives by RBPT (Pakistan) and 42 (21%) positives by RBPT (France) (Table 4.4). While the indirect modified ELISA showing 5 (2.5%) positive samples. All sera tested positives (5) with modified i-ELIA were also tested positives with another specific and highly expensive ELISA (Verion Serion) at OIE reference Summary 41 Laboratory for brucellosis, Germany. Prevalence data was analyzed by chi square test using SPSS version 20 Software (Apache License, USA) to find out correlation between risk factors and brucellosis prevalence. Risk factors such as consumption of raw milk, contact with animals, and symptoms were significant. In our opinion, indirect modified ELISA is more sensitive than RBPT (Pakistan) and RBPT (French). RBPT, (Pakistan) can be used for primary screening of brucellosis cases because of cross reactivity present in RBPT antigen and confirmation must be made with a more specific and sensitive serological test, such as B. abortus-specific indirect modified ELISA. The results of present study showed that RBPT, Pakistan antigen showed almost same sensitivity and specificity as that of RBPT (Pourquier, France) antigen. Indirect modified ELISA is more specific as compared to RBPT (Pakistan) and RBPT (French). It is highly suggestive to combine serodiagnostics test with molecular detection including PCR and Real time PCR to increase the detection rate of brucellosis. Human ELISA kit is extremely expensive that cannot be afforded in developing countries like Pakistan to screen the human. PCR technique is highly reliable and less time consuming. It is highly suggestive to conduct the study on human brucellosis on other districts of south Punjab as it is believed that brucellosis is endemic due to lack of awareness and vaccination in these areas. It results in losses to the economics of the farmers/owners in terms of poor health, abortions, long calving interval, production of weak offspring’s and poor quality of milk and meat. This disease is under-investigated and hence people are ignorant of this insidious problem. Serological surveillance is a good and cheaper tool to diagnose this problem. Determination of the seroprevalence is important to know the load of disease and pockets of infections in the areas of central Punjab, Pakistan. There is also a need to know which diagnostic antigen is more sensitive and specific from disease eradication and control point of view. A total of 200 camel serum samples were collected from three districts of the Punjab province and tested by conventional screening test i.e., Rose Bengal Plate Agglutination Test (RBPT) by using two antigens of different countries origin i.e. RBPT (IDEXX, Pourquier, France) and RBPT (VRI, Pakistan) and then tested with confirmatory competitive Enzymelinked Immunosorbent assay (cELISA). The data thus obtained regarding seroprevalence was analyzed by using Chi-square and logistic regression IBM SPSS Statistics 20 (Apache software license,USA). Summary 44 Of the total 200 camels (50 male and 150 female), 5% (10 of 200), 4% (8 of 200) were seropositive for anti-Brucella antibodies. Different risk factors were also included in study like origin (nomadic and organized), area, age, gender, season, type of herd, abortion history and orchitis in male. All of these risk factors were statistically analyzed to reveal the truth about camel brucellosis. Of the various risk factors studied, the risk factors including camels (cows) positive history of abortion (45.5%), orchitis (camel bulls) (33.3%), rearing with other ruminants (9.4%), winter season (11.5%), nomadic production system (2.67-6.67%), and area Jhang (6- 12%) and Chiniot (2-8%) were statistically significant which could be potential source of threat for humans and other animals. The testing was performed using conventional methods as well as using cELISA. It is concluded that cELISA is more specific than conventional screening tests but molecular diagnosis is highly suggestive for future studies. ELISA performed in Pakistan (cELISA, Svanovir) and the ELISA (iELISA, ID VET Kit, France) performed in OIE reference lab for brucellosis in Germany showed perfect agreement between them (both tested four camel sera positive). The current study will help to minimize and eradicate the low prevalence of camel brucellosis by creating awareness amongst the farmers and through vaccination and herd immunization of all camel calves at age of 4-8 months. Rb patients and asymptomatic carriers undergo a series of clinical tests for diagnosis and tumor treatment. These clinical examinations prove to be expensive and time consuming. On the other hand if the proband’s RB1 gene mutation status is determined by genetic testing, it can prove as more significant and cost effective diagnostic methods. Secondly, only those asymptomatic or at risk carriers with the mutation, require clinical surveillance while those proven to be unaffected do not require additional clinical examinations. Furthermore early diagnosis of Rb by molecular testing can enable and enhance clinical management, earlier treatment, follow-up care, carrier screening, genetic counseling, prenatal diagnosis and reproductive planning in predisposed families. Irrespective of the importance of molecular testing of Rb patients, in Pakistan only a few clinical reports on Rb are available so, there was a dire need to find RB1 mutations in Pakistani Rb patients and to set a molecular based diagnosis for poor affected families. Keeping in view the importance of molecular diagnosis, in this study a reliable genetic test has been developed to detect the RB1 germline mutations in Pakistani Rb patients. During this study, 70 Rb patients including 38 unilateral and 32 bilateral cases were enrolled, from different regions of Pakistan. By using direct sequencing method, seven novel and twelve reported RBI mutations were found. The novel mutations included three frameshift mutations (c.1116_1119delCACT in exon 11, c.1436_1437delAC in exon 16 and c.2060_2061insTCATT in exon 20) and four substitutions (c.148G>T in exon 2, c.610G>T in exon 2, g.94G>C in exon 7, c.947A>T in exon 10 and g.1991G>C in promoter region) while twelve reported mutations in 146 22 patients included, 9 substitutions (c.160G>T in exon 2, c.289G>T in exon 3, c.751C>T in exon 8, c.920C>T in exon 9, c.967G>T in exon 10, c.1072C>T in exon 11, c.1654C>T in exon 17, c.2063T>C in exon 20 and c.2359C>T in exon 23), one frameshift mutation (c.772_776del in exon 8) and two splice site mutations (c.380+1G>T and c.1215+1G>A in intron 3 and 12 respectively). Mutation detection rate was found to be 77.8% in (7/9) bilateral familial, 50% in (2/4) unilateral familial, 56.5% in (13/23) bilateral sporadic and 14.7% in (5/34) unilateral sporadic patients while overall rate of mutations in bilateral and unilateral patients was detected as 62.5% (20/32) and 18.4% (7/38) respectively. Beside mutations one novel c.940-64C>T (intron 9) and nine reported intronic variants c.380+45 C>T (intron 3), c.501-77G>A (intron 4), c.1128-72T>G (intron 11), c.1695+99A>T (intron 17), c.1695-1696delAA (intron 17), c.1815- 104A>G (intron 18), c.1961-10T>C (intron 19), c.2663+33T>C (intron 25) and c.2664-10T>A (intron 25) were also found. Carrier screening facility was also provided to six asymptomatic siblings (as possible carriers) of familial proband but none of them was found to be diseased. Hopefully, in future the findings and developed protocol of this study will help to reveal the molecular basis of Rb in Pakistani Rb patients which additionally help to secure vision and life of Rb patients. Further, in Pakistan there is dire need to develop “National Rb Registry Centre”, to register all new Rb cases for finding incidence rate and prevalence of Rb in Pakistan. Beside this other related issues like financial constraints, health education, planning and awareness about Rb, occupational training for health providers, capacity building for neonatal ophthalmologic screening and cosmetic rehabilitation for surviving Rb patients are important and should consider. Out of all 30 rabbits 24 rabbits were experimental on which saliva was applied, three were negative control to check natural wound healing, and three were positive control on which wound healing medicine was applied. The 24 experimental rabbits were further divided into four groups with each group consisting of 6 rabbits to check the effect of age on wound healing. The age groups of human samples were divided as 15-25, 25-35, 35-45 and 45-55 (Verma et al. 2013). Saliva of human individuals belonging from these four age groups was applied on the wounds of experimental group. Furthermore, all age groups contained saliva from both gender i.e. each age group consisted of 3 male and 3 female saliva samples. Furthermore, DNA was extracted from blood samples of the same individuals from whom saliva samples were procured. HTN1 gene which is responsible for the production of salivary histatin protein was amplified using specific primers and PCR optimization. CHAPTER 6 SUMMARY 33 The results of this study demonstrated the wound healing properties of histatin proteins present in saliva and thus, providing a basis of using the natural ability of human saliva to act as a major component in the future of medicine for wound healing and preventing wound infections in both human and animals. Ginkgo biloba leaf extract (GBE) and Panax ginseng root extract (PGE) are ancient Chinese herbal drugs that have prominent position in the list of the best-selling natural remedies and are increasingly being used for the treatment of diabetes. The anti-diabetic effect of GBE is attributed to flavonoides while that of PGE is attributed to ginsenosides. In this study, GBE and PGE in combination showed significantly higher anti-diabetic effects than individual extracts in diabetic rats. Adult Wistar rats were allowed to feed on a high fat diet (HFD: 12.7% maize starch, 6.5% dextrose, 3.9% sunflower oil, 31.3% beef tallow and 28.6% casein by weight) for two weeks. The rats were divided into seven groups (08 rats in each group): Non-diabetic control group, Diabetic group, Diabetic + 100 mg/kg G. biloba leaf extract treated group (GBE), Diabetic + 300 mg/kg, P. ginseng root extract treated group (PGE), mixed 1 group : Diabetic + combination of both GBE and PGE at dose of 200 mg/kg/day (50mg/kg/day of GBE and 150mg/kg/day of PGE), mixed 2 group : Diabetic + combination of both GBE and PGE at dose of 400mg/kg/day (100mg/kg/day of GBE and 300mg/kg/day of PGE), mixed 3 group : Diabetic + combination of both GBE and PGE at dose of 600mg/kg/day (150mg/kg/day of GBE and 450mg/kg/day of PGE). At the end of the 14th day, the rats were kept in fasting condition overnight and then a single intra-peritoneal injection of alloxan monohydrate (Sigma, USA) dissolved in 0.5 ml of saline solution at a dose of 120-130 mg/Kg body weight was injected in all rats except for the non-diabetic group which were injected with an equal volume of normal Summary 79 saline. Body weight (BW) and blood glucose were measured at week 1 and week 14. At the end of the experimental period, blood samples in fasting/ basal state were collected from heart puncture for the biochemical parameters. Liver, muscles and adipose tissue were also collected for mRNA expression of genes involved in carbohydrate and fat metabolism. Results were expressed as Means ± S.E.M. Statistical analyses was performed using Statview software (SAS Institute Inc., SAS Campus Drive, Cary, NC, USA). Two-ways repeated measure ANOVA followed by PLSD Fisher's test was performed for BW and blood glucose to assess the effects of time and herbal drugs. For the rest of the parameters, one-way ANOVA followed by PLSD Fisher's test was performed to assess the effect of herbal drugs. Differences were considered significant at P < 0.05. A significant (P < 0.0001) reduction in the BW of the diabetic group was recorded compared to non-diabetic rats and a significant reduction in BW was observed after treatment in all the five treated groups compared to diabetic group. Glycemia was significantly higher in the diabetic rats (P < 0.0001) compared to non-diabetic rats and a significant reduction in the blood glucose level was recorded in all the five treated groups (P < 0.0001) group in comparison to the diabetic group. A significant reduction for fasting serum glucose (FSG) (P < 0.0001) was recorded for all the five treated groups compared to the non-treated diabetic rats. We linked the reduction in hyperglycemia to the mRNA expression of genes involved in glucose metabolism. In particular, we studied the gene expressions of GLUT-4, insulin receptor (IR), insulin receptor substrate-1 (IRS-1) and phosphoenolpyrovate carboxykinase (PEPCK) in liver, muscle and adipose tissue. A significant up-regulation for the mRNA expression of GLUT-4 was observed only in muscle in all the five treated groups, i.e. GBE (P < 0.001), PGE (P < 0.001), mixed 1 (P < 0.0001), mixed 2 (P < 0.0001) and mixed 3 (P < 0.0001). A significant up-regulation was recorded for PEPCK in GBE (P < 0.05), mixed 1 (P < 0.05), mixed 2 (P < 0.05) and mixed 3 (P < 0.05) groups in liver. A significant increase of blood cholesterol was found in rats in the diabetic state (P < 0.0001) and a significant reduction was found only in the mixed 3 (P < 0.001) treated group. A significant decrease was found for VLDL-C in mixed 1 (P < 0.05), mixed 2 (P < 0.0001) and mixed 3 (P < 0.0001) groups. A significant decreased was observed for LDL-C in mixed 1, mixed 2 and mixed 3 (P < 0.0001) groups which previously found to be enhanced in diabetic condition. In case of HDL-c a significant decreased was found for GBE (P < 0.001), PGE (P < 0.05), mixed 1 (P < 0.001), mixed 2 (P < 0.0001) and mixed 3 (P < 0.0001) which was previously found to be increased in the diabetic group (P < 0.0001). Conversely, a significant increase was seen for TG (P < 0.0001) in the diabetic state and a significant reduction was found in all the five treated groups (P < 0.0001). We further studied genes involved in lipid metabolism. In conclusion, we found that both GBE and PGE have strong individual anti-hyperglycemic, anti-hyper-triglyceridemic and anti-oxidative effects in an alloxan monohydrate induced rat model of diabetes. Both also showed strong influence on the activation on the expression of genes involved in the metabolic pathways of glucose and lipid which previously became dysfunctional in diabetic rats. When both these natural remedies were given in combination, synergistic effects were recorded in a dose dependent manner. Further work is needed to evaluate the way by which human beings suffering from diabetes are safely treated with these herbal remedies. Buffalo’s milk contributes approximately 68% to the total milk production in the country. The research in dairy cattle indicated that milk production is associated directly to the type of carbohydrates fed in the diet. Carbohydrate fraction contributes approximately 70% of dry matter in the diets for lactating dairy cows. NFC is a major source of energy for lactating dairy animals. According to NRC a typical dairy ration should contain NFC between 35 to 40 % of the total dietary DM to support optimal milk production. Studies in cattle have shown positive correlation of NFC with milk production.High NFC diets have shown improved production performance in cattleTherefore aim of current experiment was to evaluate the optimal level of straw based non fiber carbohydrate TMR on production performance of lactating Nili-Ravi buffalo. The experiment was carried out at Buffalo Research Institute (BRI), Pattokito evaluate the effect of two levels of NFC on; dry matter intake, body weight, body score, fecal score, milk production, changes in milk composition, milk production efficiency. In this experiment twelve lactating Nili Ravi buffaloes of same parity and comparable milk production were selected. Animals were housed on concrete floor and were fed individually. The shed was well ventilated. The duration of experiment was 50 days. Animals were given adaptability period of 10 days and remaining 40 days for data recording and sample collection. Animals were randomly assigned into two dietary treatments: Treatment 1: Straw based low NFC (32%) TMR as a control and Treatment 2: Straw based high NFC (40%) TMR with n=6 animals per treatment. Diets formulated were iso-nitrogenous. At the start of experiment animals were weighed and then weekly during experimental period. Hand milking was carried out twice daily in the morning and evening at 7.00 and 19.00, respectively. Milk let down was done by calf suckling. During the entire experimental period, ad libitum availability of fresh and clean water to individual animal was ensured. The animals were fed on straw based non-fiber carbohydrate TMR once daily in the morning after milking. TMR offered and refusal was recorded daily. The effect of treatments on milk yield, milk composition, dry matter intake and other variables was analyzed by two independent sample t-tests using Procttest procedure of SAS ( release 9.1; SAS Institute, Inc, Cary, NC). Conclusion In present research animals were given two levels of straw based non-fiber carbohydrate TMR and it was observed that by giving straw based low NFC (32%) TMR the production performance of Nili-Ravi buffaloes were better than NFC (40%). It is also important for amtDNA in Nuclear Mitochondrial sequences (NUMTs) in vertebrates species so that it rarely occurring NUMTs. The control region in equine consist of two highly variable regions (HVR1 and HVR2), 4 conserved blocks (CSB), and variable repeats of 8 bp motifs (Cothran, et al. 2013). The displacement loop hyper-variable region of mtDNA (D-loop) of Pak- local donkeys are very similar to genome of other species; however there are considerable differences in the mtDNAevolutionary rate for different taxonomic groups. The phylogenetic tree based on consensus sequences of 12 Asian Donkeys breeds available on NCBI and sequences of local Pakistani donkey breeds showed their genetic relationship among each other. The clade was consisting of Pakistani local donkey breeds i.e. 3-SF, 7-SF,10-SF,11-SF,12-SF,13-SF, K-3,K-6,K-11,K-13,K-16,K18,K-19 and K-20 showing their high relatedness.K-8 and 8-SF have more mutation rate in the sequence and have more diversity from other individuals. The second major branch was furher divided in two sub branches i.e. Donkey family representing Equusburchellichapmani (JX312729), Equusburchelliquagga (JX312733), Equusgreyvi(NC020432) and Equus zebra (JX312718) clustered together. The second sub branch was consisting of other donkey breeds i.e. Equushemionusonager (JX312730), Equushemionus(NC016061) and Equushemionuskulan (NC018782) grouped together while two Pakistani local donkey breeds i.e. SF-8 and K-8 were clusterd with Equusasinussomalicus (AP012271) and Equusasinus (X97337). The d-loop sequence of Human (Homo sapiens) was taken as out group and it clearly differed and separated from rest of phylogenetic tree i.e. camel, bovine, ovine, caprine and other mammals. The phylogenetic tree constructed under this study not only confirmed the status of Pak- local donkey breeds but also confirmed the genetic relationship among other mammalian species, thus reconfirming the already established biological classification. Epidemic and endemic nosocomial infections caused by K. pneumonia species are leading causes of morbidity and mortality throughout the world. Irrational use of antibiotics is creating antibiotic resistance problems in Pakistan. Excessive use of β-lactam antibiotics is responsible for production of ESBL enzymes by Gram negative bacteria especially K. pneumoniae. A total of 150 samples including urine, pus and sputum were processed for the isolation and evaluation of the ESBL enzyme producing K. pneumoniae as well as their antimicrobial sensitivity pattern. Samples were cultured on cystine lactose electrolyte deficient (CLED), blood, chocolate and McConky agar. Isolates were identified by culture characters, staining reaction followed by biochemical testing through API-20E index system. ESBL production potential was assessed by double disc diffusion method. Out of total 50 urine samples, K. pneumoniae was isolated from 20 samples. Among these 20 isolates, 6 were confirmed as ESBL producers. However, within 50 sputum and pus samples, 13 and 15 isolations were possible out of which 5 and 6 were positive for ESBL production, respectively. Frequency distribution for isolation of K. pneumoniae from urine, pus and sputum samples did not significantly vary from each other. Similarly, ESBL producing potential of all K. pneumoniae isolates also did not significantly vary as p values are higher than 0.05. A total of 17 ESBL producing K.pneumoniae isolates were tested for their antibiotic susceptibility pattern against 12 commonly used antibiotics i.e. Amoxacillin, Ampicillin, Aztreonam, Ceftazidime, Cefatoxime, Ceftriaxone, Cefixime, Imipenem, Meropenem, Ciprofloxacin, Gentamycin and Amikacin. All of the tested K. pneumoniae isolates showed 100 % sensitivity against amikacin, imipenem and meropenem while found to be completely resistant against rest of the antibiotics. Molecular confirmation of ESBL production was done through PCR. DNA samples were extracted from ESBL producing isolates. Amplification of the plasmid DNA region (TEM gene) from the DNA samples was done through PCR. The resulting PCR product was run on 1% agarose gel through horizontal gel electrophoresis. Three samples from urine (S1, S4 and S5) and two samples from sputum (S7 and S8) produced required bands while none of the other samples produced any band. High proportion of ESBL producing K. pneumoniae isolates in present study is an alarming scenario. These organisms are resistant to conventional antibiotics and might spread in the environments thus pose serious threat to the health of human and animal population. Only clinical specimen submitted to the diagnostic lab were tested in present study. Further extensive studies are needed to bridge the gaps in knowledge of antibiotic resistance pattern of K. pneumoniae. Blood samples were collected from 100 domestic cats at Pet Center, UVAS, Lahore,from their ear tips and cephalic/saphenous vein. The blood will be immediately transferred to EDTA coated vacutainers. Stained thin blood smears were observed for intra-erythrocytic bodies and 45 samples were selected after screening. Blood in EDTA were tested for PCR (already optimized) in the Molecular Parasitology laboratory at UVAS, Lahore, to screen for B. felis. ExtractedDNA of confirmed B. felis samples were further processed forLoop-Mediated Isothermal Amplification (LAMP). LAMP primers weredesigned recognizing four sections of the B. felis gene. LAMP reactions of 25µL were standardized at 60°C temperature for 1 hour time using DNA extracted from blood samples of cats found positive on PCR. Briefly, the concentration of FIP and BIP were varied from 0.8µM to 2.4µM, Mg2+from 2mM to 4mM, betaine from 0.2 to 0.8M and dNTPs from 1mM to 4mM.The LAMP reaction was optimized at the final concentration of 0.2µM F3 and B3, 2.0µM of each of the FIP & BIP, 2mM for each dNTPs, 0.8M betaine, 1X reaction buffer, 1µl bst polymerase and 2µl DNA templates at 60°C. Nevertheless, serotypes prevalent in Asia includes A, O and Asia-1. Because of evolution in genomic sequence of FMDV, it is becoming difficult to control the problem through conventional methods. Changes in the genome can be detected using software through sequence analysis. In the software, evolutionary models are used to measure the evolutionary change for the identification of new sub lineages. In current study genomes sequence data (1998 - 2011) of bovine FMD serotypes (A, O and Asia 1) was collected through NCBI in FASTA format. This data was converted into PHYLIP format. On Dell workstation, with Microsoft Windows 8.1 operating system, BioEdit, TipDate V.1.2 was deployed. Sequence data was aligned through CLUSTAL W algorithm of Multiple Sequence Alignment using BioEdit. Using TipDate, genome sequence data was analyzed using three evolutionary models (F84, HKY and REV) and phylogenetic trees were produced showing evolutionary rate and likelihood ratio of FMDV serotypes O, A and Asia-1.. Results of the current study showed higher values of evolutionary rate in bovine FMD virus which was estimated 7.49 x 10-4 with likelihood value -1429.507680 in serotype A, 2.418 x 10-3 with likelihood -3707.168484 in serotype O and2.16 x 10-3 with likelihood value -3723.344884 in serotype Asia-1, respectively. Markove Reversible Model showed higher rates of evolution in all three serotype with best likelihood values. Phylogenetic results showed higher rate of evolution or substitution in viruses. Furthermore serotypes A, O and Asia-1are mutating with passage of time and new variants are being observed. It was also observed that this evolutionary process is continued in these three serotypes during 1998-2011. This study confirmed the evolutionary changes in FMDV serotypes prevalent in Pakistan during the period 1998 – 2011. This study showed that isolate are evolving with increasing rate. High rate of mutation in Asia-1 was observed than serotype A and serotype O. F84 and HKY85 models produced close results but these models are not identical works on equal and unequal base frequencies. Keeping in view the importance of babesiosis under local conditions, the present study was designed to evaluate the status babesiosis in Balochistan. For this purpose field and experimental studies were carried in two districts Quetta and Sibi of Balochistan Province to find out the status of babesiosis in Bhag Nari, Holstein Friesian and Crossbred cattle. During field study epidemiological status of babesiosis was highlighted by selecting 600 cattle randomly from each district. The animals were distributed into 2 major groups i.e. Young animals less than 12 months and adult over 12 months of age. These groups were further sub-divided into Young animals (less than 6 months, up to 9 months and up to 12 months) while Adults animals (up to 2 years, 3 years and over 3 years). The vector of babesia was also kept under keen observation for the prevalence/infestation rate, identification and economic losses caused during the course of study. Blood samples were collected from each animal and processed for blood smears examination and PCR for further confirmation of babesia infection. The blood samples were also processed for hematological study to evaluate the effect of babesiosis on different blood parameters. For experimental study 148 animals were selected through clinical signs of babesiosis, blood smear examination and PCR. Out of theses 40 animals were maintained for therapeutic trail to find out the cheapest and easily available drug against bovine babesiosis. For this purpose Neem leaves were used in decoction form while Imidocarb dipopionate was kept as standard control. The Summary 177 results of epidemiological study revealed higher prevalence of babesiosis (20.5%) in district Quetta while 15.16% was recorded in District Sibi. Similarly higher prevalence was recorded in Holstein Friesian than in Crossbred and Bhag Nari cattle respectively in both districts Quetta and Sibi. Furthermore higher prevalence of babesiosis was recorded in adult groups of Holstein Friesian than in Crossbred and Bhag Nari cattle. Similarly season wise higher prevalence of babesia infection was noticed in summer followed by spring, autumn and winter respectively while higher prevalence was noted in female group of animals than male animals. Blood smears examination and PCR confirmed two babesia species i.e. babesia bigemina and babesia bovis. Similarly Boophilus tick species were identified as the vector of babesia parasites. During present study mixed hemoprotozaon infection of babesia mixed with theileria was recorded in both districts. The results of conventional method and modern diagnostic technique (PCR) revealed that PCR identified higher babesia infection during the entire 4 seasons as well as in all age groups whereas blood smears examination was capable to diagnose babesiosis in adult groups during the months of summer and spring season. Breed wise prevalence was also higher in samples treated with PCR than blood smears examination and even samples that were declared negative by blood smears examination were also found positive. The results of complete blood cell count from blood samples of infected experimental animal showed regenerative, macrocytic hypochromic anemia. Blood smear examination showed presence of many babesia with reticulocytes. Abnormalities in erythrocyte structure were seen. The result of blood parameters of total erythrocyte count, total leukocyte count, packed cell volume and hemoglobin showed significant decrease in all three affected Bhag Nari, Holstein Friesian and Cross bred cattle. The values of MCV and MCH were increased and MCHC was slightly less than normal value. No efficacy of neem decoction was noted against bovine babesiosis. The method involved isolation and identification of MRSA from surgical wounds of hospitalized patients & associated health care workers in a tertiary care hospital in Lahore and healthy volunteers from the community. Plant essentials oils & extracts were evaluated for their antibacterial activity against selected MRSA isolates. Oils were recovered by steam distillation using an all-glass distillation assembly. Then in vitro sensitivity and MICs of plant essential oils were determined using vancomycin and linezolid as commercial standards. The essential oils were screened further for the active constituents by column chromatography using various solvents and identification of compounds were performed by GC/MS analysis and the fractions which showed prompt results were evaluated for antimicrobial activity against the MRSA isolates in quest to find new therapeutic options. Finally effective essential oils and their active fractions were studied for their toxicity using in vitro Genotoxic assays such as Ames and Comet assays. To further ensure their beneficial effects antimutagenic effect of the essential oils were also studied. Prevalence of S. aureus among patients was 52.9%, in HCWs 86.5% and in community 74% with an overall percentage of 72.6%. Among S. aureus those declared as MRSA were 91.8% from patients, 50.6% from HCWs and 59.5% from community with an overall percentage of 62.2% MRSA. Among the isolated MRSA overall 90.6% were Coagulase positive and 75.2% were biofilm positive. SUMMARY 211 The pattern of MRSA resistance against current antibiotics have shown an overall increase in the resistance with maximum shown for lincomycin followed by tetracycline, ampicillin, fusidic acid, amoxicillin and piperacillin with tazobactam. The most effective options among current regime were tigecyclin, amikacin and meropenem showing an overall least resistance. Resistance against linezolid was observed with an overall percentage of 25.6 % and vancomycin 33.3% by disc diffusion method. The MRSA isolates resistant to one or more groups of antibiotics were declared as MDRs. Among patients and health-care workers all were declared as MDRs where as in community 93.1% isolates were MDRs. Upon Protein profiling using whole cell proteins 44 bands of the polypeptides were produced with molecular size 10-200kDa from the three sampling groups and were categorized into 5 clusters showing an overall significance correlation with each other explaining an interesting fact that all these strains were interlinked establishing the fact of flow of hospital acquired MRSA in the community and vice versa. This analysis also gave an insight in explaining the fact of horizontal transmission of infection within the hospital. Keeping in view the raise in resistance among current available antibiotics indigenous medicinal plants essential oils were screened for active constituents exhibiting anti-bacterial effects against MRSA isolates. Maximum yield was obtained from Carum copticum followed by Cuminum cyminum and minimum yield was obtained in case of Zingiber officinale. Upon qualitative analysis of all five essential oils Carum copticum essential oil showed zones of inhibition greater than the standards vancomycin and linezolid followed Cuminum cyminum and Zingiber officinale in all three SUMMARY 212 sampling groups. Anethum sowa and Myristica fragrans essential oils showed no activity against MRSA. Minimum inhibitory concentration of the three essential oils determined by micro broth dilution method indicated that Carum copticum showed least value in all three types of MRSA isolates followed by Zingiber officinale and Cuminum cyminum. Effective essential oils were further fractioned using silica gel gravity columns. All the fractions obtained were screened for the anti-bacterial activity against all three types of MRSA isolates. Only fraction F1 of Carum copticum showed activity greater than pure essential oil and the two commercial standards of vancomycin and linezolid. For the identification of active constituents GC/MS analysis was performed on all three essential oils and their respective fractions. In case of fraction F1 the most dominant constituents were Carvacrol, p-Cymene, Ʈ-Terpinene and Apiol. In other two plants none of the fractions were effective. Therefore it was concluded to use pure essential oils in case of Zingiber officinale and Cuminum cyminum rather than their individual fractions and incase of Carum copticum Fraction F1 has shown superior activity. Finally these essential oils were tested for possible mutagenic effect using bacterial reversion mutation assay and Comet assay. No mutagenic effects were observed at MIC and above doses. These effective essential oils were also evaluated for possible antimutagenic effect. Both Carum copticum and Zingiber officinale essential oils showed strong antimutagenic effects and weak antimutagenic effect by Cuminum cyminum. Upon analysis of nuclear damage none of the plants essential oils and fraction F1 of Carum copticum showed genotoxic effects and indicated to be safe. Thus from the study it was concluded that Carum copticum essential oil and its fraction F1 were the most effective to be further investigated as an alternative treatment for MRSA infections. Newcastle disease virus causes respiratory diseases in humans, birds and other mammals, representing one of the foremost threats to public health. In this study, the antiviral activity of Momordica charantia L. and Ribavirin against Newcastle disease virus was evaluated in-ovo. For each extract of the plant M. charantia and ribavirin 40 embryonated eggs were assigned to 8 groups containing 5 eggs in each group (six groups for antiviral, six groups for embryonic toxicity, and two groups were kept positive and negative control respectively) and marked them with lead pencil. The aqueous and ethanolic extracts of Momordica charantia L. was prepared by using soxhlet extraction technique. From the extract, six different dilutions i.e. 160mg/ml, 80mg/ml, 40mg/ml, 20mg/ml, 10mg/ml and 5mg/ml of the aqueous and ethanolic extracts were prepared in normal saline whereas; six different dilutions i.e. 15μg/ml, 20μg/ml, 25μg/ml, 30μg/ml, 35μg/ml and 40μg/ml of ribavirin were made in normal saline. With ND virus the different concentrations of the extracts of plant were mixed and 0.2 ml of this suspension was injected to 9th to 10th day embryonated eggs along with positive and negative controls having only virus and normal saline correspondingly. Ribavirin, standard drug, was inoculated by following the mentioned manner. These inoculated embryonated chicken eggs were incubated at 370C and were checked after 12 – 72 hours. After 72 hours of post inoculation, all the eggs were chilled at 40C in fridge for overnight stretch of time and the allantoic fluid was collected. Summary 64 The embryo survival percentage, positive or negative spot haemagglutination activity and determination of virus titre by haemagglutination test confirmed the antiviral activity. The embryonic toxicity effects of Momordica charnatia aqueous and ethanolic extracts and ribavirin was assessed by merely inoculating the extracts of respective concentrations as used for antiviral activity in embryonated chicken eggs and incubating for 72 hours. This study was designed to evaluate the effect of ZONP supplementation on histomorphometric parameters of small intestine and cecal tonsil in broiler. The study included 100 day-old broiler chicks which were randomly allocated into four groups (n= 25), each group having five replicates (n= 5). Group A was served as control group and was fed only basal diet (BD), Group B was supplemented with BD plus 80 mg/kg of ZnO, Group C was supplemented with BD plus 40mg/kg of ZONP and Group D was supplemented with BD plus 80mg/kg of ZONP for 35 days. Two birds from each replicate were randomly selected and slaughtered on day 35, birds were excised and samples from small intestine were taken from duodenum, jejunum, ileum and cecal tonsil from ceaca. Paraffin embedding technique was used to process the samples which were stained by Haematoxyline & Eosin technique and combined Alcian blue- PAS. Histomorphometry was done by using software (ProgRes®2.1.1 Capture Prog Camera Control Software). The data was analyzed by using one way-ANOVA. Differences between the groups were compared by Duncan’s Multiple Range Test and were considered significant at P < 0.05. The intestinal parameters, villus height, width, surface area, crypt depth, thickness of lamina propria, muscularis mucosa and muscularis externa were measured. Acidic, mixed and total goblet cells and intraepithelial lymphocytes were also counted in intestinal samples. In cecal tonsils length, width, area and total lymphatic nodules were measured. Summary 38 The results showed that in duodenum and ileum the villus height, width and surface area was significantly higher (P < 0.05) in group C, while that of jejunum was greatest (P <0.05) in group D, the crypt depth of duodenum was deeper in group B. Total goblet cells count was significantly higher (P< 0.05) in group C and D in all three sections of small intestine also the acidic goblet cells of jejunum and ileum, while the mixed goblet cells count was highest (P< 0.05) in group C and D of duodenum and ileum. No significant difference were observed in IEL count between different groups. The length, width, area and total number of lymphatic nodule of cecal tonsils was higher (P < 0.05) in group C and D. In conclusion supplementing ZONP at the dose of 40mg/kg in diet of broilers improved gut morphology and at the dose of 40 and 80 mg/kg improved the mucosal immune cells count. It is defined as a contagious systemic bacterial disease primarily of ruminants. The disease is manifested by late term abortions, weak calves, still births, infertility and also associated placentitis, epididymitis and orchitis, with excretion of the organisms in uterine discharges and milk. With the intensification of the import of animals and the establishment of big farms in the last few years, the incidence of brucellosis increased sharply in many countries, both in man and animals. In this study 360 serum samples were examined from four groups of animals in district Mirpur Azad Kashmir. Blood samples of 3ml from buffaloes, cattle, sheep and goat (n=30) each were taken from three sub-divisions of Mirpur separately. The serum samples were screened by RBPT which is a screening test for brucellosis, and it was observed that 8.6% animals were seropositive by RBPT. The serum samples of cattle were 17.8%, buffalo were 8.9%, goat were 2.2%, and sheep were 5.6% positive respectively. The serum samples positive by RBPT and some randomly taken samples were further confirmed by the use of most specific and sensitive serological test known as i-ELISA. 6.87% animals were confirmed as seropositive by i-ELISA including cattle (17.5%), buffalo (10%), sheep (0%) and goat (0%). All RBPT positive samples were further subjected to RT-PCR. Among these 31 samples 24 were positive for Brucella genus and only 7 samples were negative. Samples were further tested for confirmation of Brucella species. All 24 samples were having Brucella abortus. The data were analyzed by using the Statistical package for the social sciences (SPSS) program version 22. Data were analyzed using Chi-square test and Z-test statistics. Fungal laccases have boundless biotechnological functions across the globe like the decolouration and detoxification of industrial effluent, bleaching of pulp, phenolic elimination from wines, in preparation of biosensors in detergents blocking dye transfer- functions. Laccase showed vast variety of substrates due to this ability they can enhance different types of industrial mechanism such as methylation, demethylation, polymerization, mineralization of pollutants like hydrocarbons. White rot fungus is efficient for the production of laccase using agro-waste as substrate. In this research white rot fungus was isolated from stock cultures of Department of Microbiology, University of Veterinary and Animal Sciences, Lahore, Pakistan and the organism was maintained on Tien& Kirk media slants and petri plates. Solid state fermentation technique was used using basal fermentation medium and agro waste rice bran was used as substrate for the production of laccase.Proximate analysis was performed of the substrate rice bran to analyse crude protein, fat, ash, moisture and fibre content. The fermentation was performed at room temperature and flasks were placed on orbital shaker at 100 rpm and 30˚C. Enzyme activity was checked using (ABTS) as substrate at 420nm, for every 24 hour to observe the maximum enzyme production. Fermentation parameters like substrate concentration, incubation period, pH, temperature and nitrogen source (corn steep liquor and ammonium sulphate) were optimized. The concentration of substrate optimized for rice bran was 7.5g/100ml and optimum production of 6.11 IU/ml of enzyme was observed. The optimum day for the production of enzyme was day 7 and the amount of enzyme produced was 6.91 IU/ml. The optimum pH and temperature were 4 and 40˚C respectively, and the amounts of enzyme produced were 7.48 IU/ml and 7.96 IU/ml respectively. Two nitrogen sources optimized were maize steep liquor 1ml and ammonium sulphate 0.2 g, and the enzyme produced was recorded 7.67 IU/ml and 9.41 IU/ml respectively. Large scale fermentation batch of one litter was carried out under the optimized conditions and the enzyme produced was 9730 IU/L. Triplicates of each parameter were prepared. The enzyme was purified using the purification techniques like ammonium sulphate precipitation, then by dialysis excess salt was removed, and then gel filtration was performed to collect different fractions on the basis of size of molecules and molecular mass of the laccase was analysed by SDS-PAGE. The size of the protein was found to be 70kDa. Characterization of laccase was performed in terms of optimum pH, temperature and in response to inducers and inhibitors. The optimum pH and temperature of the purified enzyme was 6 and 40˚C respectively. The inducer copper sulphate enhanced the activity of enzyme up to 9.7 U/ml and then inhibitors EDTA and 2-merceptoethanol reduced the activity level up to 4.17IU/ml and 3.98 IU/ml respectively. To study and analyse the effects of optimization parameters Pearson correlation, descriptive statistics and one way Anova were used. The optimum production of laccase was achieved using agro waste rice bran. The enzyme produced was economical and it can provide effective solutions for bioremediation of hazardous compounds and pollutants. Mitochondrial geneCytochrome c oxidase subunit 1 (CO1)of 500bps was used as a marker for identification at specie level.Genomic DNA was extracted by each blood and tissue sample of eleven bird species (33 samples). Amplification of CO1 gene was a done by using a universal set of primers (BIRDF1 and BIRDR1)containing region of almost 750 bps (Hebert et al. 2003).Amplicons were purified and sequenced Sanger sequencing method (Sanger et al. 1977). Forward and reverse sequences were analyzed using softwaresEMBOSS merger,ClustalW, BioEdit and nBLAST. Phylogenetic analysis of selected bird species was done. Each sequence was aligned with its reference sequences of CO1 gene present on NCBI. Every nucleotide position which did not align with the reference sequence was studied to identify SNPs. Fixation index (FST) were used to measure species diversity within a same sub population relative to that found in the entire population. Consensus sequences (500bps) generated was used to construct their phylogenetic tree to see their evolutionary relationship with other bird species. All species showed their closest linkage with their respective species. Pakistani population of peafowl and chicken species showed the close relation with same sequences generated in China. Tranopans showed its closest linkage with T. temminckii. In conclusion, seven species ofPhasianinaesub-family of Pakistani bird species was genetically characterized first time in Pakistan by using CO1 as a barcode. It proves that DNA barcoding is an efficient and accurate molecular tool for species identifica¬tion and phylogenetic implication. This study leads to establish a DNA Data Bank that helped scientists to investigate the biodiversity, taxonomic classification, specie identification, in forensic purposes and to study the genetic and phenotypic evolution of these species. DNA barcoding through CO1 gene works as a functional tool for detectingmeat mislabeling and preventing illegitimate trade. This study has established foundations for molecular biologists to study taxonomic uncertainties at sub species level using SNP based identifying marker. Animals in this group were given 3 shots of Oxytetracycline LA at 20mg/kg BW (IM) repeated every 48 hrs. Streptomycin @ 13mg/kg BW IM was given for six consecutive days. Furthermore, flunixin meglumine @ 2mg/kg BW IM for 3 days in combination with antibiotic. Selevit injection containing selenium and vitamin E was administered (IM) for consecutive five days. At next day all the animals in this group were vaccinated with a combined vaccine of RB51 and FMD (Oil based) at a dose of 3mL/animal through deep IM route. In group B, all the animals were given Saafi (Herbal Product) orally consecutively for six days. Furthermore, flunixin meglumin @ 2 mg/kg BW IM was administered for three days. All these treatments were given simultaneously. Then injection Selevit containing selenium and vitamin E were administered (IM) for consecutive five days. After treatment all animals were vaccinated with RB51 + FMD (oil based) 3mL/ animals deep intramuscularly. In group C, flunixin meglumin @ 2 mg/kg BW IM was administered for three days. After NSAID, Selevit injection containing vit. E and selenium was administered (IM) for five days regularly. After treatment, animals were vaccinated with RB51 + FMD (oil based) 3 mL/ animals (IM). In D group five negative adult animals were included in this group and vaccinated subcutaneously with RB51 and FMD (Oil based) at a dose of 3 mL/animal. Summary 40 Data regarding this study was analyzed with ANOVA and completely randomized design was used to compare the mean between different groups using statistical package for social science (SPSS) version 20. P < 0.05 was considered significant. The results showed OD values after 1st, 3rd and 6th month were significantly different in all groups. The mean values depicted that OD values was significantly higher (2.886±0.099) in vaccinated (positive) animal’s vs the other three groups. There was no significant difference in OD values of herbal treated, antibiotic treated and health control animals. The lowest OD values was found in healthy control animals, though significantly similar with herbal treated, antibiotic treated. The mean OD value at 3rd month after treatment is significantly higher (1.794±0.090) in herbal treated and vaccinated positive groups but the OD value of healthy group was lowest (1.794±0.090) after 3rd month of treatment. After 6th month of treatment the OD value of positive vaccinated group was found significantly highest (1.146±0.194). On the other hand lowest value (0.595±0.079) was found in healthy control group. All the groups have found non-significant difference in all group. Regarding Real time PCR the value range from 0-40 Ct values. In the present study the Ct values of the control positive was found to be 13 which indicate control positive. The antibiotic treated group had a Ct value found in this range 35.61 to 38.13. The herbal treated group were all positive and the values were 31.34 to 37.42 Ct. The vaccinated immune booster group. The Ct values in the group was found to be 30.83 to 34.21. The birth weight and placenta dropping time was normal in antibiotic treated animals. There were no significant results found in all groups. Summary 41 This study concluded that herbal regimen is effective in vitro against brucellosis but in vivo it is yet not to be evaluated. The antibiotics can be effective to treat the Brucellosis with this protocol. This was help to control bovine brucellosis, and extensive economic losses. A number of endoparasites are prevalent in Khyber Pakhtun Khwa province and district Malakand. Anthelmintic resistance(AR) is a very important emerging issue in the control of parasites. The current project was designed to study the point-prevalence of various gastro-intestinal parasites of goats in Kot Manzaray BaBa of Malakand Agency and to determine the presence of AR in the parasites prevalent there. Keeping in view the importance of small ruminants in our country and due to the routine use of dewormer, the present project has been designed to achieve our objective. For this purpose Eight Teddy goat farms were selected from different localities of Kot Manzari Baba District Malakand for studies on AR using faecal egg count reduction test and egg hatch test to further confirm the results of the FECRT. Information regarding flock management and worm control practices was collected through a well-structured questionnaire. The questionnaire was filled out by 20 (each) goat farmers, quacks (traditional healers), qualified veterinarians and veterinary assistants through interviews and/or group discussions. Results revealed that majority of the goat flocks were found to have nematode populations resistant to oxfendazole and levamisole. None of the goats were found to have nematode populations resistant to ivermectin accept the Haemonchus contortus which showed a little bit indication of resistance in the study. Majority of the goat flocks were found to have nematode populations susceptible to ivermectin. The frequency of resistance was comparable among Haemonchus contortus, Trichostrongylus colubriformis and Teladorsagia circumcincta. Veterinary assistants look after for maximum number of flocks for their healthcare. A total of 300 venous blood samples of 5 ml each were collect¬ed from sheep of different areas of two Districts i.e. Abbottabad and Battagram, with 150 samples from each District. Then serums were dispatched to the National Agriculture Research Centre (NARC), for the diagnosis of Brucella melitensis by Rose Bengal Plate Test (RBPT) and Indirect Enzyme-linked immunosorbent assay (I-ELISA). The prevalence of the Brucella in sheep was determined as described by (Thrusfield 2013. The prevalence of brucellosis in Hazara Region by RBPT and I-ELISA was recorded as 4.67 % and 4.33 % respectively. From 150 samples of each district, 14 (9.33 %) and 13 (8.67 %) of samples were tested positive by RBPT and I-ELISA respectively in District Abbottabad whereas, no (0 %) samples were tested positive by RBPT and I-ELISA in District Battagram. Furthermore, on Tehsil wise sero-prevalence, only 2 (2.67 %) samples out of 75 were tested positive for brucellosis in sheep by both RBPT and I-ELISA in Tehsil Havelian, while in Tehsil Abbottabad 12 (16 %) and 11 (14.67 %) samples out of 75 were tested positive for brucellosis in sheep by RBPT and I-ELISA from district Abbottabad whereas, no positive samples were detected in Tehsil Battagram and Tehsil Allai by both RBPT and I-ELISA from district Battagram. Statistically there was a significant (>0.05) difference between the prevalence of brucellosis in sheep and area of Hazara Region. The demographic variables (age, sex and breed) were analyzed using Chi-square test on SPSS version 17.0 and statistically there was no significant (>0.05) difference between the prevalence of brucellosis in sheep and demographic variables i.e. age, sex and breed. The present study has shown that brucellosis is prevalent in sheep in the Hazara region. Female animals are more sensitive than their male counterparts. Sheep of breeding age are more vulnerable than the very young ones and Balkhi breed is more prone to the disease as compared to the Kaghani and Ramghani breeds. The Sensitivity and Specificity of the diagnostic test (RBPT) was determined as described by (Thrusfield 2013). The calculated sensitivity of Rose Bengal Plate Test was 100 % and its specificity was 99.6s5 %. The results of this study will be published to communicate the interesting findings and to increase the knowledge of veterinary and public health management. Therefore, we suggest the implementation of control measures, raising public knowledge on prevention methods, a complete surveillance system for the brucellosis in Hazara Region. HCV is affecting people worldwide; more than 170 million peoples have been affected so far and the number is increasing day by day. Its prevalence in Pakistan is about 3% to 6%. There is lot of variation in its genome and it is classified into 6 major genotypes and these genotypes are further classified into many subtypes. Size of HCV is about 9500 bps which only encodes single polyprotein. This 3000 to 3300 amino acids polyprotein is processed by cellular and viral proteases to generate 10 polypeptides consisting of 4 structural (Core, E1, E2 and P7) and 6 non-structural proteins (NS2, NS3, NS4A, NS4B, NS5A, NS5B). The most immunogenic gene from all the genes is E1. It involves in the interaction with the host cell and easily escapes from the immune system of host due to the presence of hypervariable regions in E1 gene. To isolate the E1 gene, RNA extraction was done using the kit method. RNA was converted to cDNA which was then amplified in two rounds of PCR using nested primers from HCV core region. After confirming the presence of HCV RNA in serum samples, PCR amplification of HCV E1 gene was carried out using gene specific nested primers. Amplified E1 gene products were Sanger sequenced and aligned with standard sequence to find out genetic variations. E1 gene sequences were converted to protein sequences for which secondary protein structures were made and analyzed. No noticeable change was seen in these secondary protein structures. The protein sequences were also analyzed for the presence of B-cell and T-cell epitopes; two T-cell epitopes (QAFTFRPRR, FLVGQAFTF) were found which may inform the development of a proper vaccine against HCV. It is an economically important tick-borne disease of buffalo in tropical and sub-tropical areas of the world. In current study, we developed and optimized PCR first for detecting Anaplasma at genus level in buffaloes. One hundred (100) blood samples were collected from buffaloes around the Lahore region. The stained thin blood films were examined microscopically and 37% blood samples were found positive for intra-erythrocytic bodies which were then selected for DNA extraction. The DNA was extracted using commercially available kit for eventual use in optimization of PCR for diagnosis of bovine Anaplasmosis. The primers were designed targeting 16S rRNA gene of Anaplasma. For the detection, the PCR product was run in 2% agarose gel stained with ethidium bromide and thirty seven samples showed the amplification band at 179bp. The selected samples were sent for ABI sequencing to Singapore for the accurate detection of the Anaplasma species. The sequencing results were blasted with database of Genbank and we observed homology with Anaplasma phagocytophilum. We found 37% prevalence of Anaplasmosis in buffaloes through PCR. However more studies are required to confirm the species of Anaplasma infecting buffaloes (Bobalus bobalis) by designing species specific primers. Furthermore, additional studies are needed to establish the epidemiology of Anaplasmosis by using molecular tools in different geographical areas of the country for their better control. An epidemiological questionnaire focusing on trade animals as well as husbandry and sanitary practices that could be associated with the risk of Brucellosis infection was completed. Serum samples were collected and analyzed using Rose Bengal Plate Test. The serum samples positive for Brucellosis through RBPT further subjected to enzyme linked Immunosorbent assay. The samples of cattle collected from publically owned abattoir are subjected to the RBPT and the result was 18% and privately owned abattoir have an RBPT result of 10% and samples of the buffaloes are subjected to the test of RBPT as an screening test which was respectively 20% and 8%. The samples are further subjected to the iELISA and the results was shown that through ELISA the result was respectively 12% and 4%. While results for the cattle declared the prevelance of brucella infection through indirect ELISA was 8% and 2% at public and private abattoir respectively. The abattoir workers had a high risk to the exposure to the transmission of the disease as it is world widely more zoonotic the workers found positive for the brucellosis at public abattoir through RBPT shown no positive results. While in private abattoir through RBPT there is 6.66% animals were positive. However we concluded the results through indirect ELISA the results found were 66.66% and 15.62% for the butchers and meat sellers respectively at public abattoir.so the butchers are prone to the infection of the brucella antibodies.in comparison with the private abattoir 33.33% was detected in the abattoir cleaner. Exuberant granulation typically is an irregular and unhealthy appearing tissue with many grooves and clefts. Histologically exuberant granulation tissue has remains of fibrin deposits, which have not been cleared by the acute inflammatory response. Horses are predisposed to trauma wounds that can be labour intensive and expensive to manage. Wounds localized at the trunk region are less problematic and heal faster than wounds located at the distal aspect of the limb. Complications such as wound infection, formation of exuberant granulation tissue (EGT) and hypertrophic scarring are frequent. The EGT or “Proud Flesh” is considered similar to the keloid of the human skin and together with wound contamination/infection, is currently the most complicated aspect of wound management in equines. The present project was designed to evaluate comparative efficacy of surgical and chemical debridement of such kind of wounds in horses. The clinical study was conducted on 20 already traumatized horses of either sex. The animals were divided into four groups (I, II, III and IV) comprising five animals each. These animals were kept in S.P.C.A, University of Veterinary and Animal Sciences, Lahore. In a group I, Surgical excision was carried out to remove excessive growth from wounded area. Local anaesthesia with two percent lignocaine was in filtered for removal. Antiseptics were applied and over-granulation was smoothly removed from one margin of skin to other. Group II animals were treated with ten percent copper sulphate ointment, Group III animals were treated with two percent silver nitrate ointment and Group IV animals were treated with white lotion. Each technique had its own advantages and disadvantages. The results proved that surgical removal was a best choice for management of hyper-granulated wounds. It also had good scar formation SUMMARY 72 and faster recovery as compared to other chemical agents. It allows the closure of large wounds and faster epithelization to margins. The results of present study clearly indicated that surgical excision of hyper-granulated wound on early stages yields good recovery and proper union. Although silver nitrate has better penetrating power in wounds but it has no ability to epithelize the wound which terminates in slow recovery. Moreover silver is an expensive and less affordable to a common man. Copper sulphate has good recovery but it causes irritation while application and animal feels pain. ZNOPs improve the anti-oxidant health status of broilers at low concentrations compared with bulk zinc oxide. The study was consist of 100 day-old broiler chicks and randomly divided into 4 groups, each group consists of five replicates (n= 5 in each replicate). Group A was control fed with basal diet (BD) only, Group B was supplemented with 80 mg ZnO/kg BD, Group C and D was supplemented with 40 and 80 mg ZNOPs/kg BD respectively for 35 days. The parameters were delayed type hyper sensitivity, malondialdehyde, glucose, total protein, albumin, globulin, triiodothyronine, thyroxin, creatinine, bilirubin, urea and uric acid. Statistical analysis was conducted with Statistical Packages for Social Sciences (SPSS version 20 USA). The data was analyzed using one way ANOVA. The results of serum glucose concentration was found significantly higher (P < 0.05) in Group B and Group C compared to the Group A but was found similar to Group D Delayed type hyper sensitivity, Serum Anti-oxidant status, Serum Proteins, Serum thyroid hormone level, Kidney Function Tests, Serum Bilirubin Concentration were found non-significant (P < 0.05) in all the groups compared to the control group. ZNOPs do not cause nephrotoxicity hence they are safe for kidney .These particles does not disturb the thyroid hormones. However their antioxidant and protein effects were not observed in our study. 900. Clinico-Epidemiological And Experimental Observations On Feline Lower Urinary Tract Disease Among Domesticated Cats

Nicks of this problem also found in Pakistan, however the veterinarians are usually unable to properly diagnose this disease due to lack of knowledge as well as the ancillary diagnostic equipment availability for this disease. Present study was divided into two phases. Phase – 1 included clinico-epidemiological data. To this end, target of more than 502 domesticated client-owned cats of either sex, age, breed, etc showing signs of feline lower urinary tract disease (FLUTD) as per Buffington (1994) were examined accordingly from 3 different cities (Lahore, Faisalabad, and Islamabad) of Pakistan). All data collected was based on a predesigned proforma by using structured interview of the owners. Diagnosis was solely based on serum-cortisol levels, urinalysis, radiography and ultrasonography. Phase II involved experimental trial. The data obtained from whole of the study was then presented in tabulated form as frequencies and percentages. Treatment and outcome of the disease were also analyzed accordingly. According to the present study conducted it is proved that iFLUTD is present among the cats in Pakistan. Its proper cognizance among the Pakistani veterinarians is still non-existent and is misdiagnosed as colic or constipation issues in cats. The present study was undertaken to bring iFLUTD into the reportive of small animal practitioners working in Pakistan. The present study debunked various previous notions like iFLUTD is associated with commercial diets and canned foods only if we talk about this region majority of cases were noticed that had home-cooked food given by the owner. Moreover, cases in Siamese breed are larger than Persian breed. It has been strongly associated with Indoor housing management. Additional work is still needed to explore untouched areas of epidemiology including factors other than those being studied in the previous literature. Academicians in veterinary pathology and veterinary medicine of Pakistani universities should embrace this malady in the Doctor of Veterinary Medicine (DVM) curricula. According to the present study results it is concluded that two factors like stress and pain accelerate the sympathetic nervous system outflow compared to normal felines leading to the inflammatory response. Thus the stress factor must be reduced in the form of making hiding places for cats at home to reduced down the fear factor along with enhancing the feeling of owes for that particular place. Moreover, some more practices should be performed by the owner to reduce down the stress factor like playing with the pet, giving full attention, placing toys and other attractive things like yarn balls at the feline places (where they live/placed). There was no significant difference found between the groups based on the food with health score along with the therapeutic judgment. Hence, it is recommended that more experiments should be performed on larger scale to assess GAG therapy on increased number of felines and need of hour is to conduct more veterinary studies to get information and authenticity for its use against iFLUTD. From this study conducted, I recommend to the owners that the cats must be provided with the indoor hiding places and play with their pets in order to reduce the stress factor that increases the risk of idiopathic lower urinary tract disease.
